There is critical clinical demand for tissue‐engineered (TE), 3D constructs for tissue repair and organ replacements. Current efforts toward this goal are prone to necrosis at the core of larger constructs because of limited oxygen and nutrient diffusion. Therefore, critically sized 3D TE constructs demand an immediate vascular system for sustained tissue function upon implantation. To address this challenge the goal of this project was to develop a strategy to incorporate microchannels into a porous silk TE scaffold that could be fabricated reproducibly using microfabrication and soft lithography. Silk is a suitable biopolymer material for this application because it is mechanically robust, biocompatible, slowly degrades in vivo, and has been used in a variety of TE constructs. Here, the fabrication of a silk‐based TE scaffold that contains an embedded network of porous microchannels is reported. Enclosed porous microchannels support endothelial lumen formation, a critical step toward development of the vascular niche, while the porous scaffold surrounding the microchannels supports tissue formation, demonstrated using human mesenchymal stem cells. This approach for fabricating vascularized TE constructs is advantageous compared to previous systems, which lack porosity and biodegradability or degrade too rapidly to sustain tissue structure and function. The broader impact of this research will enable the systemic study and development of complex, critically‐sized engineered tissues, from regenerative medicine to in vitro tissue models of disease states.  相似文献

The scaling behavior of Co, Co–Ni and Ni silicides to sub-40 nm gate length CMOS technologies with sub-100 nm junction depths was evaluated. Limitations were found for Co and Co–Ni alloy silicides, which exhibited an increase in sheet resistance at gate lengths below 40 nm and required high processing temperatures to achieve low junction leakage. Ni silicide was shown, in contrast, to have good scaling behavior, with a decrease in sheet resistance for decreasing gate lengths down to 30 nm, lower diode leakage (at similar sheet resistance) and lower silicide to p+ Si contact resistance than Co silicide. Key material issues impacting the applicability of NiSi to CMOS technologies were investigated. Studies of the kinetics of Ni2Si growth were used to design a process that avoids excessive silicidation of small features. The thermal degradation mechanisms of NiSi films were also studied. Thin films degraded morphologically with activation energies of 2.4 eV. Thick films degraded morphologically at low temperatures and by transformation to NiSi2 at high temperatures, suggesting a higher activation energy for the latter mechanism. Pt alloying was shown to help stabilize NiSi films against morphological degradation.  相似文献

The venous blood pH, pCO2, lactate, pyruvate and haematocrit responses to an intravenous (i/v) infusion of halothane-lipid emulsion were assessed in Pietrain, Pietrain X Norwegian Landrace and Gloucester pigs. Halothane-induced lactacidosis was dose related in the Pietrain (stress sensitive) pigs between 0 and 82 μl halothane kg?-1, whereas the crossbred and Gloucester pigs did not show any lactacidosis. It is concluded that the i/v halothane test can be used to quantify a stress sensitive pig's reaction to the drug, and that the crossbred pig does not express halothane sensitivity even at a mild level. Carazolol pretreatment in Pietrain pigs increased the halothane-induced lactacidosis, whereas in a separate study carazolol was found to decrease their rate of post-mortem muscle acidification. This indicates that halothane activates a different mechanism from stress when inducing lactacidosis in stress sensitive pigs.  相似文献

The formalism of the one-dimensional discrete wavelet transform (DWT) based on Daubechies wavelet filters is outlined in terms of finite vectors and matrices. Both the scale-dependent wavelet variance and wavelet covariance are considered and confidence intervals for each are determined. The variance estimates are more accurately determined with a maximal-overlap version of the wavelet transform. The properties of several Daubechies wavelet filters and the associated basis vectors are discussed. Both the Mallat orthogonal-pyramid algorithm for determining the DWT and a pyramid algorithm for determining the maximal-overlap version of the transform are presented in terms of finite vectors. As an example, the authors investigate the scales of variability of the surface temperature and albedo of spring pack ice in the Beaufort Sea. The data analyzed are from individual lines of a Landsat TM image (25-m sample interval) and include both reflective (channel 3, 30-m resolution) and thermal (channel 6, 120-m resolution) data. The wavelet variance and covariance estimates are presented and more than half of the variance is accounted for by scales of less than 800 m. A wavelet-based technique for enhancing the lower-resolution thermal data using the reflected data is introduced. The simulated effects of poor instrument resolution on the estimated lead number density and the mean lead width are investigated using a wavelet-based smooth of the observations  相似文献

Modern video-based endoscopes offer physicians a wide-angle field of view (FOV) for minimally invasive procedures. Unfortunately, inherent barrel distortion prevents accurate perception of range. This makes measurement and distance judgment difficult and causes difficulties in emerging applications, such as virtual guidance of endoscopic procedures. Such distortion also arises in other wide FOV camera circumstances. This paper presents a distortion-correction technique that can automatically calculate correction parameters, without precise knowledge of horizontal and vertical orientation. The method is applicable to any camera-distortion correction situation. Based on a least-squares estimation, our proposed algorithm considers line fits in both FOV directions and gives a globally consistent set of expansion coefficients and an optimal image center. The method is insensitive to the initial orientation of the endoscope and provides more exhaustive FOV correction than previously proposed algorithms. The distortion-correction procedure is demonstrated for endoscopic video images of a calibration test pattern, a rubber bronchial training device, and real human circumstances. The distortion correction is also shown as a necessary component of an image-guided virtual-endoscopy system that matches endoscope images to corresponding rendered three-dimensional computed tomography views.  相似文献
