European perch (Perca fluviatilis) harvested from three lakes of Central Italy were studied in different seasonal periods of a year to evaluate their nutritional quality and some safety aspects related to the pollution of the aquatic environment. The lakes considered, located in the Latium region, differed with respect to their volcanic (Bolsena and Bracciano Lakes) or artificial (Salto Lake) origin. Fillets of fish caught in the three lakes were characterised by good protein (17–19%) and mineral contents and low lipid levels (0.6–1.2%) throughout the year. Total lipids were characterised by low cholesterol levels (41.9–74.7 mg/100 g) and high percentages of total n − 3 polyunsaturated fatty acids (27.7–33.8% of total fatty acids), in particular docosahexaenoic acid (14.2–25.3% of total fatty acids). The qualitative analysis of the stomach content of perch confirmed their predatory feeding behaviour. The chemical and nutritional profiles of perch from the three lakes were comparable except for rubidium and cesium levels, which were higher in the muscle tissues of perch from the volcanic lakes. These minerals may represent elements of traceability of the origin of fish. Low levels of organochlorine pesticides and polychlorinated biphenyls, well below the Italian and European action limits, were detected in the muscle tissue of perch from all three lakes.  相似文献

Tyrosine kinase substrate with four SH3 domains (Tks4) scaffold protein plays roles in cell migration and podosome formation and regulates systemic mechanisms such as adult bone homeostasis and adipogenesis. Mutations in the Tks4 gene (SH3PXD2b) cause a rare developmental disorder called Frank-Ter Haar syndrome (FTHS), which leads to heart abnormalities, bone tissue defects, and reduced adiposity. We aimed to produce a human stem cell-based in vitro FTHS model system to study the effects of the loss of the Tks4 protein in different cell lineages and the accompanying effects on the cell signalome. To this end, we used CRISPR/Cas9 (clustered, regularly interspaced, short palindromic repeats (CRISPR)/CRISPR associated (Cas9)) to knock out the SH3PXD2b gene in the HUES9 human embryonic stem cell line (hESC), and we obtained stable homo- and heterozygous knock out clones for use in studying the potential regulatory roles of Tks4 protein in embryonic stem cell biology. Based on pluripotency marker measurements and spontaneous differentiation capacity assays, we concluded that the newly generated Tks4-KO HUES9 cells retained their embryonic stem cell characteristics. We propose that the Tks4-KO HUES9 cells could serve as a tool for further cell differentiation studies to investigate the involvement of Tks4 in the complex disorder FTHS. Moreover, we successfully differentiated all of the clones into mesenchymal stem cells (MSCs). The derived MSC cultures showed mesenchymal morphology and expressed MSC markers, although the expression levels of mesodermal and osteogenic marker genes were reduced, and several EMT (epithelial mesenchymal transition)-related features were altered in the Tks4-KO MSCs. Our results suggest that the loss of Tks4 leads to FTHS by altering cell lineage differentiation and cell maturation processes, rather than by regulating embryonic stem cell potential.  相似文献

Two studies were conducted to examine infants' social responses to attractive and unattractive faces. In Study 1, 60 12-month-olds interacted with a stranger who wore a professionally constructed attractive or unattractive mask. The infants showed more positive affective tone, less withdrawal, and more play involvement with the stranger in the attractive condition. In Study 2, 43 12-month-olds played with an attractive and an unattractive doll. The infants played significantly longer with the attractive doll. These results extend and amplify earlier findings showing that young infants exhibit visual preferences for attractive over unattractive faces. Both visual and behavioral preferences for attractiveness are evidently exhibited much earlier in life than was previously supposed. Tumor microenvironments are often characterized by an increase in oxidative stress levels. We studied the response to oxidative stimulation in human primary (IGR39) or metastatic (IGR37) cell lines obtained from the same patient, performing patch-clamp recordings, intracellular calcium ([Ca2+]i) imaging, and RT-qPCR gene expression analysis. In IGR39 cells, chloramine-T (Chl-T) activated large K+ currents (KROS) that were partially sensitive to tetraethylammonium (TEA). A large fraction of KROS was inhibited by paxilline—a specific inhibitor of large-conductance Ca2+-activated BK channels. The TEA-insensitive component was inhibited by senicapoc—a specific inhibitor of the Ca2+-activated KCa3.1 channel. Both BK and KCa3.1 activation were mediated by an increase in [Ca2+]i induced by Chl-T. Both KROS and [Ca2+]i increase were inhibited by ACA and clotrimazole—two different inhibitors of the calcium-permeable TRPM2 channel. Surprisingly, IGR37 cells did not exhibit current increase upon the application of Chl-T. Expression analysis confirmed that the genes encoding BK, KCa3.1, and TRPM2 are much more expressed in IGR39 than in IGR37. The potassium currents and [Ca2+]i increase observed in response to the oxidizing agent strongly suggest that these three molecular entities play a major role in the progression of melanoma. Pharmacological targeting of either of these ion channels could be a new strategy to reduce the metastatic potential of melanoma cells, and could complement classical radio- or chemotherapeutic treatments.  相似文献

This pilot study compared placental growth factor (PIGF) levels in populations with high versus low risk for cardiovascular disease. Previous experiments from our laboratory (Sundaresan et al. 2005, 2009) revealed that the angiogenic factor PIGF was up regulated in modeled microgravity conditions in human lymphocytes leading to possible atherogenesis and pathogenesis in microgravity. Since the findings came from microgravity analog experiments, there is a strong link to its usefulness in the microgravity field as a biomarker. It is important to understand, that these findings came from both studies on expression levels of this cardiovascular marker in human lymphocytes in microgravity (in vitro microgravity analog), and a follow up gene expression study in hind limb suspended mice (in vivo microgravity analog). The relevance is enhanced because in life on earth, PIGF is an inflammatory biomarker for cardiovascular disease. Studies on the levels of PIGF would help to reduce the risk and prevention of heart failures in astronauts. If we can use this marker to predict and reduce the risk of cardiac events in astronauts and pilots, it would significantly help aerospace medicine operations. The investigations here confirmed that in a cardiovascular stressed population such as coronary artery disease (CAD) and acute coronary syndrome (ACS) patients, PIGF could be overexpressed. We desired to re-evaluate this marker in patients with cardiovascular disease in our own study. PIGF is a marker of inflammation and a predictor of short-term and long-term adverse outcome in ACS. In addition, elevated PIGF levels may be associated with increased risk for CAD.PIGF levels were determined in thirty-one patients undergoing cardiovascular catheterization for reasons other than ACS and in thirty-three low-risk asymptomatic subjects. Additional data on traditional cardiovascular risk factors for both populations were also compiled and compared. We found that PIGF levels were significantly higher in the high-risk population as compared to low-risk population. Also we were able to ascertain that PIGF levels were inversely correlated with HDL-cholesterol but directly correlated with the triglyceride levels. With further validation, PIGF may prove a useful addition to the armamentarium of noninvasive biomarkers for cardiovascular disease including a new area of stressful physiological conditions such as microgravity.  相似文献

Saccadic eye movements remain spatially accurate even when the target becomes invisible and the initial eye position is perturbed. The brain accomplishes this in part by remapping the remembered target location in retinal coordinates. The computation that underlies this visual remapping is approximated by vector subtraction: the original saccade vector is updated by subtracting the vector corresponding to the intervening eye movement. The neural mechanism by which vector subtraction is implemented is not fully understood. Here, we investigate vector subtraction within a framework in which eye position and retinal target position signals interact multiplicatively (gain field). When the eyes move, they induce a spatial modulation of the firing rates across a retinotopic map of neurons. The updated saccade metric can be read from the shift of the peak of the population activity across the map. This model uses a quasi-linear (half-rectified) dependence on the eye position and requires the slope of the eye position input to be negatively proportional to the preferred retinal position of each neuron. We derive analytically this constraint and study its range of validity. We discuss how this mechanism relates to experimental results reported in the frontal eye fields of macaque monkeys.  相似文献

We present here a hybrid approach to modeling helium-ion lithography that combines the power and ease-of-use of the Stopping and Range of Ions in Matter (SRIM) software with the results of recent work simulating secondary electron (SE) yield in helium-ion microscopy. This approach traces along SRIM-produced helium-ion trajectories, generating and simulating trajectories for SEs using a Monte Carlo method. We found, both through simulation and experiment, that the spatial distribution of energy deposition in a resist as a function of radial distance from beam incidence, i.e. the point spread function, is not simply a sum of Gauss functions.  相似文献

The possibility to exploit commercially the peculiar characteristics of refractory metallic and ceramic materials and in particular of Zirconium diboride ceramics—a class of promising materials for high temperature applications—often depends to a great extent on the ability to join different ceramics one to the other or to special metallic alloys. As the behaviour of a metal-ceramic joint is ruled by the chemical and the physical properties of the interface, the knowledge of wettability, interfacial tensions and interfacial reactions is mandatory to understand what happens at the liquid metal-ceramic interface during joining processes. In the framework of an extensive study aimed at evaluating the wettability and the interfacial characteristics of different metal-ceramic systems, the behaviour of ZrB2 in contact with liquid Ag and its alloys (Cu, Ti, Zr, Hf) has been studied. ZrB2 pure, with different sintering aids or “alloyed” with other ceramic materials (SiC, Si3N4), have been used. The wetting and spreading experiments have been performed by the sessile drop technique under controlled atmospheres. The wetting and spreading characteristics and the interfacial reactions are discussed as a function of time, compositions of the ceramic and of the alloy involved. The interfacial morphologies, analysed by SEM and EDS, show the presence of regular interfaces and adsorption layers and of different bulk phases which are interpreted in terms of the relevant phase-diagrams.  相似文献
