This work deals with the deposition of lead(Pb) thin films by the U V pulsed laser ablation technique,for their further use as photocathode devices in superconducting radio frequency guns.Scanning electron microscopy and atomic force microscopy analyses were performed to study the morphological features of Pb thin films deposited on Si(100) and Nb substrates.The films showed a granular structure with a nearly fully covered surface only for that one deposited on Nb substrata X-ray diffraction measurements indicate the growth of polycrystalline Pb thin films with a preferential orientation along(111) planes.Results of the photoemission performance of Pb thin film deposited on Nb substrate showed a very encouraging average value of quantum efficiency of 6×10~(-5) through a single-photon absorption process,promoting further studies in the realisation of Pb photocathodes by this techniqua  相似文献

The water buffalo mozzarella cheese is a typical Italian cheese which has been introduced in the thriving Brazilian market in the last 10 y, with good acceptance by its consumers. Lactic acid bacteria (LAB) play an important role in the technological and sensory quality of mozzarella cheese. In this study, the aim was to evaluate the diversity of the autochthones viable LAB isolated from water buffalo mozzarella cheese under storage. Samples were collected in 3 independent trials in a dairy industry located in the southeast region of Brazil, on the 28th day of storage, at 4 ºC. The LAB were characterized by Gram staining, catalase test, capacity to assimilate citrate, and production of CO2 from glucose. The diversity of LAB was evaluated by RAPD‐PCR (randomly amplified polymorphic DNA‐polymerase chain reaction), 16S rRNA gene sequencing, and by Vitek 2 system. Twenty LAB strains were isolated and clustered into 12 different clusters, and identified as Streptococcus thermophilus, Enterococcus faecium, Enterococcus durans, Leuconostoc mesenteroides subsp. mesenteroides, Lactobacillus fermentum, Lactobacillus casei, Lactobacillus delbrueckii subsp. bulgaricus, and Lactobacillus helveticus. Enterococcus species were dominant and citrate‐positive. Only the strains of L. mesenteroides subsp. mesenteroides and L. fermentum produced CO2 from glucose and were citrate‐positive, while L. casei was only citrate positive. This is the first report which elucidates the LAB diversity involved in Brazilian water buffalo mozzarella cheese. Furthermore, the results show that despite the absence of natural whey cultures as starters in production, the LAB species identified are the ones typically found in mozzarella cheese.  相似文献

The aim of this work was to evaluate the microemulsification as sample preparation procedure for determination of Cu, Fe, Ni and Zn in vegetable oils samples by High-Resolution Continuum Source Flame Atomic Absorption Spectrometry (HR-CS FAAS). Microemulsions were prepared by mixing samples with propan-1-ol and aqueous acid solution, which allowed the use of inorganic aqueous standards for the calibration. To a sample mass of 0.5 g, 100 μL of hydrochloric acid and propan-1-ol were added and the resulting mixture diluted to a final volume of 10 mL. The sample was manually shaken resulting in a visually homogeneous system. The main lines were selected for all studied metals and the detection limits (3σ, n = 10) were 0.12, 0.62, 0.58 and 0.12 mg kg−1 for Cu, Fe, Ni and Zn, respectively. The relative standard deviation (RSD) ranged from 5% to 11 % in samples spiked with 0.25 and 1.5 μg mL−1 of each metal, respectively. Recoveries varied from 89% to 102%. The proposed method was applied to the determination of Cu, Fe, Ni and Zn in soybean, olive and sunflower oils.  相似文献

The temporo-spatial patterning of lectin-binding sites was examined by lectin histochemistry and quantitative methods in the microvasculature of the optic tectum of 9-, 14-, 20-day-old embryos and 30-day-old chickens. Horseradish peroxidase and colloidal-gold-labelled lectins were used for detection of beta-D-galactose (RCA-I, Ricinus communis agglutinin-I) and of N-acetylglucosamine and sialic residues (WGA, Wheat germ agglutinin) at light and electron microscopical levels. At the light microscopical level, RCA-I and WGA binding sites were detectable in the early embryonic capillaries in a diffuse staining pattern; in later embryonic stages and in adult animals, RCA-I labelling became located on the abluminal surface of the vessels, while WGA staining was detected on the luminal surface. Ultrastructurally, gold labelling for RCA-I was seen intracytoplasmically in endothelial cells in 9-day-old embryos. In 14-to 20-day-old embryos and in chickens, binding sites for RCA-I were detected in endothelial tight junctions and basement membranes. In contrast, labelling of the gold-coupled WGA lectin was distributed almost exclusively on the luminal endothelial surface already in early embryos. The results indicate that the endothelial cells of the optic tectum acquire functional polarity early in their development and that glycoconjugates containing beta-D-galactose residues are involved in the biochemical composition of the tight junctions and basement membrane, which are considered to be key structures in blood-brain barrier (BBB) differentiation.  相似文献

We report on the simple, in situ generation of CdS nanocrystals inside electrospun polymer fibres by thermal decomposition of a cadmium thiolate precursor, leading to nanocomposite light-emitting fibres. The modifications induced in the precursor by the thermal decomposition are investigated by a morphological, structural and spectroscopic analysis of the resulting nanocomposite fibres. This approach allows us to overcome nanofabrication difficulties related to disfavoured micro- or nanofluidic molecular flow as given by the direct incorporation of particles in the electrospinning solution. This method therefore enables the synthesis of luminescent, CdS-based composite fibres with emission peaked in the visible range, suitable as building blocks for nanophotonic devices based on light-emitting nanomaterials.  相似文献

The development of new antiviral drugs against SARS-CoV-2 is a valuable long-term strategy to protect the global population from the COVID-19 pandemic complementary to the vaccination. Considering this, the viral main protease (Mpro) is among the most promising molecular targets in light of its importance during the viral replication cycle. The natural flavonoid quercetin 1 has been recently reported to be a potent Mpro inhibitor in vitro, and we explored the effect produced by the introduction of organoselenium functionalities in this scaffold. In particular, we report here a new synthetic method to prepare previously inaccessible C-8 seleno-quercetin derivatives. By screening a small library of flavonols and flavone derivatives, we observed that some compounds inhibit the protease activity in vitro. For the first time, we demonstrate that quercetin (1) and 8-(p-tolylselenyl)quercetin (2d) block SARS-CoV-2 replication in infected cells at non-toxic concentrations, with an IC50 of 192 μM and 8 μM, respectively. Based on docking experiments driven by experimental evidence, we propose a non-covalent mechanism for Mpro inhibition in which a hydrogen bond between the selenium atom and Gln189 residue in the catalytic pocket could explain the higher Mpro activity of 2d and, as a result, its better antiviral profile.  相似文献

The ordered mesoporous silica material MCM-41 was used to prepare a novel drug delivery system (DDS) for oral administration of the diuretic furosemide (FURO), labeled in class IV of the Biopharmaceutics Classification System (BCS). This drug is characterized by both low solubility and permeability and its absorption window is the stomach and the proximal small intestine. Thus, the aim of this work was the development of an immediate formulation that could improve the drug release in its preferential absorptive regions.Three inclusion products MCM-41-FURO-E/D, MCM-41-FURO-S and MCM-41-FURO-E have been prepared in three different ways in order to find the best preparation conditions. They were characterized by XRPD, DSC, FT-IR, TGA and B.E.T. and then the inclusion products displaying the best characteristics were submitted to in vitro studies. They evidenced that the examined inclusion products displayed FURO dissolution enhancement, followed by a complete release within 90 min. Moreover, physical stability studies revealed that MCM-41 is able to stabilize the drug preventing it from re-crystallization. At last, as FURO is light sensitive, the effect of MCM-41 on drug photochemical stability was also investigated and the results indicated the protective action exerted by the matrix.  相似文献

The antioxidant and antifungal activities of the phenolic compounds in different alcoholic extracts of aged cachaça were evaluated. The physico‐chemical analyses were performed in the Laboratório de Qualidade de Aguardente of the Universidade Federal de Lavras according to the methods of the Ministério Agricultura Pecuária e abastecimento. Total phenol content was determined by the Folin–Ciocalteu method, quantification of these compounds was performed by HPLC and antioxidant activities were determined by methods involving inhibition of the DPPH (1,1‐diphenyl‐2 picrylhydrazyl) radical, the β‐carotene/linoleic acid system, ABTS (2,2 azinobis‐[3‐ethyl‐6‐benzothiazolinesulfonic acid]) radical, reducing power and thiobarbituric acid. Determination of antifungal activity was accomplished through the technique of dissemination in discs using Aspergillus carbonarius, Aspergillus niger, Aspergillus flavus, Penicillium commune and Penicillium cladosporoides fungi at the Laboratório de Micologia de Alimentos. The values obtained for the phenolic compounds ranged from 0.41 to 9.69 mg L?1; syringaldehyde, vanillic acid and gallic acid were predominant. A satisfactory antioxidant activity was observed in all of the tests with the alcoholic extracts. A moderate activity against P. commune and P. cladosporoides, but no inhibition of the growth of A. carbonarius, A. niger or A. flavus was observed.
