Centromere protein B (CENP-B) is a centromeric DNA-binding protein that binds to alpha-satellite DNA at the 17 bp CENP-B box sequence. The binding of CENP-B, along with other proteins, to alpha-satellite DNA sequences at the centromere, is thought to package the DNA into heterochromatin subjacent to the kinetochore of mitotic chromosomes. To determine the importance of CENP-B to kinetochore assembly and function, we generated a mouse null for the cenpB gene. The deletion removed part of the promoter and the entire coding sequence except for the carboxyl-terminal 35 amino acids of the CENP-B polypeptide. Mice heterozygous or homozygous for the cenpB null mutation are viable and healthy, with no apparent defect in growth and morphology. We have established mouse embryo fibroblasts from heterozygous and homozygous cenpB null littermates. Microscopic analysis, using immunofluorescence and electron microscopy of the cultured cells, indicated that the centromere-kinetochore complex was intact and identical to control cells. Mitosis was identical in fibroblasts derived from cenpB wild-type, heterozygous and null animals. Our studies demonstrate that CENP-B is not required for the assembly of heterochromatin or the kinetochore, or for completion of mitosis.  相似文献

Two studies were conducted to evaluate the efficacy of attributional retraining as a career counseling technique for college students. Participants who received the attributional retraining treatment viewed an 8-min videotape designed to foster internal, controllable, and unstable attributions for career decision making. Participants in the control groups viewed a similar videotape that lacked any reference to career-related attributions. Results revealed that participants who received attributional retraining exhibited significant changes in career beliefs and attributional style and engaged in significantly more career exploration behavior than the participants in the control groups. An evaluation of attributional retraining as a career-counseling technique for college students is provided, and ideas for further research are suggested. The adaptation of a cylindrical geometry fracture toughness test specimen to testing of polymeric dental biomaterials is demonstrated. The specimen configuration facilitates the fabrication of small specimens and simplifies the experimental study of environmental effects on properties of dental biomaterials. The test method is used to measure critical stress intensity factor, KIC for poly (methyl methacrylate) (PMMA), particulate composite restorative materials, and dental polymers. Values obtained are in agreement with data reported for other specimen configurations.  相似文献

Mastering supramolecular self-assembly to a similar degree as nature has achieved on a subcellular scale is critical for the efficient fabrication of complex nanoscopic and mesoscopic structures. We demonstrate that active, molecular-scale transport powered by biomolecular motors can be utilized to drive the self-assembly of mesoscopic structures that would not form in the absence of active transport. In the presented example, functionalized microtubules transported by surface-immobilized kinesin motors cross-link via biotin/streptavidin bonds and form extended linear and circular mesoscopic structures, which move in the presence of ATP. The self-assembled structures are oriented, exhibit large internal strains, and are metastable while the biomolecular motors are active. The integration of molecular motors into the self-assembly process overcomes the trade-off between stability and complexity in thermally activated molecular self-assembly.  相似文献

To examine the intra-individual heterogeneity of the local inflammatory infiltrate in metastases of untreated melanoma patients we analysed 42 skin and subcutaneous metastases from 11 patients. Metastases were excised on the same date (10 patients) or consecutively (six patients), and processed in a two-step immunoperoxidase technique using monoclonal antibodies directed against T-cells (CD3, CD25) and two macrophage subpopulations (RM 3/1, 27E10). The number of positively stained T-cells and macrophages were counted for each lesion in representative high power fields. The mean values were calculated. We observed limited heterogeneity among T-cells (CD3+) and activated T-cells (CD25+) in simultaneously excised metastases. Contrary to this, markedly heterogeneous CD3+ infiltrates were found in metastases taken on different dates, even if there was a time difference of only 1 day between excisions, suggesting spontaneous variations. However, activated T-cells in follow-up biopsies showed only limited heterogeneity. In spite of inter-individual variations, metastases taken simultaneously, as well as consecutively, showed little intra-individual heterogeneity with respect to macrophages. Our findings should be taken into account for comparative studies of the infiltrate in metastatic melanoma during immunotherapy.  相似文献

The oxidation stability of castor oil fatty methyl ester (FAME), doped with four different phenolic antioxidants, was evaluated using a rapid method of thermal and air-contact degradation. The methodology is based on the induction times observed when the samples are contacted with pure oxygen at elevated pressures and temperatures. The results indicate different performances of the antioxidants as well as synergisms between antioxidants and biodiesel. In general, the addition of antioxidants increased from 6-15 times the stability of castor oil FAME., with BHA (butylated hydroxyanisol) showing the best results for improving antioxidation in castor oil biodiesel.  相似文献

The aim of the present research was to study changes in milk composition and fatty acid profile, specifically conjugated linoleic acid (CLA) and its isomers, in goat milk as affected by dietary supplementation of sun-flower oil and whole linseed (0.81 and 1.84% of dry matter on basal diet, respectively) and to assess the persistency of the response. To achieve this objective, bulk milk from a herd and from 6 individual dairy goats fed a diet supplemented with sunflower oil and whole linseed was monitored for a period of 3 mo. Gas chromatography and silver ion HPLC were used to analyze total CLA content and the isomeric profile of these fatty acids, respectively. The contents of α-linolenic acid increased from 0.35% with the reference diet to 0.62% with the supplemented diet. Similarly, CLA milk content increased from 0.46 to 1.18%. The same pattern was also observed for trans-11 C18:1 (1.38 to 4.05%, respectively) in goat milk after 3 mo of lipid supplementation. In contrast, changes in other trans C18:1 isomers were less remarkable. There was a strong linear correlation between cis-9, trans-11 C18:2, the main CLA isomer, and trans-11 C18:1 under the conditions assayed and their concentration remained stable throughout the duration of the study. Levels of the minor CLA isomers were also enhanced as a consequence of lipid supplementation. The most remarkable increases were observed for 11-13 (trans-trans and trans-cis geometric isomers), whereas trans-7, cis-9 (the second most important CLA isomer from a quantitative point of view) and trans-10, cis-12 increased only slightly with lipid supplementation.  相似文献

In this study, PLGA particle systems were studied for the controlled release of florfenicol, a broad spectrum antibiotic used in veterinary treatments. The emulsion-solvent evaporation technique was used for particle preparation. To evaluate the particle size, entrapment efficiency, and drug release behavior, factors such as solvent type, emulsification time and methods, and drug to polymer ratio were investigated. The results showed that the use of ethyl acetate and 2.5 min of ultrasonication or 30 min of homogenization can lead to sub-micron and micron-sized particles, respectively. Sizes between 200–300 nm and 2–3 μm were obtained for ultrasonication and homogenization procedures, respectively. Entrapment efficiencies were around 20% for all systems and release profiles were size dependent. In addition, a mathematical model was implemented to simulate the florfenicol transport. The model predicts the florfenicol release and takes into account the particle size, polymer molecular weight, and autocatalytic polymer degradation. This work assessed the risk of protozoa in 10 school restaurants in Armenia (Quindío, Colombia) by analyzing the presence of Cryptosporidium spp, Giardia duodenalis, Blastocystis, and Cyclospora cayetanensis DNA in the food, water, and living and inert surfaces of school restaurants and in stools of children who ate at these restaurants. Of the 213 food, water, and surface samples, 6.6% were positive using PCR to test DNA for Blastocystis; 3.8% for Cryptosporidium spp; 0.9% for G. duodenalis; and 0% for C. cayetanensis. In 187 stool samples analyzed via microscopy from children who attended the restaurants, 40 (21.4%) were positive for Blastocystis and 21 (11.2%) were positive for Giardia spp. Via PCR, 20 (10.7%) were positive for Cryptosporidium and 0 (0%) for C. cayetanensis. A higher positivity in children's stools for Blastocystis spp was correlated with lower compliance in property conditions and for higher positivity of Giardia spp in children's stool was related to lower knowledge by food manipulators. Inspection scores can identify restaurants with higher risk for protozoa infection.  相似文献
