In this study, the flow of lubricating greases in a labyrinth seal geometry is studied using microparticle image velocimetry (µPIV). The aim is to evaluate the grease velocity distribution inside the gap of a labyrinth seal and to find a relationship between the grease consistency and the transferred speed from the rotating ring in order to choose the correct grease as a sealing medium. In addition, the grease flow characteristics are important for the understanding of fracture due to grease layer displacement. For these purposes, four greases with different rheological properties were used in µPIV experiments. It was found that the grease consistency plays a crucial role in speed development as well as the grease composition and presence of a slip effect at the grease–rotating wall interface.  相似文献

Two out of three diseases of the prostate gland affect aging men worldwide. Benign prostatic hyperplasia (BPH) is a noncancerous enlargement affecting millions of men. Prostate cancer (PCa) in turn is the second leading cause of cancer death. The factors influencing the occurrence of BPH and PCa are different; however, in the course of these two diseases, the overexpression of heat shock proteins is observed. Heat shock proteins (HSPs), chaperone proteins, are known to be one of the main proteins playing a role in maintaining cell homeostasis. HSPs take part in the process of the proper folding of newly formed proteins, and participate in the renaturation of damaged proteins. In addition, they are involved in the transport of specific proteins to the appropriate cell organelles and directing damaged proteins to proteasomes or lysosomes. Their function is to protect the proteins against degradation factors that are produced during cellular stress. HSPs are also involved in modulating the immune response and the process of apoptosis. One well-known factor affecting HSPs is the androgen receptor (AR)—a main player involved in the development of BPH and the progression of prostate cancer. HSPs play a cytoprotective role and determine the survival of cancer cells. These chaperones are often upregulated in malignancies and play an indispensable role in tumor progression. Therefore, HSPs are considered as one of the therapeutic targets in anti-cancer therapies. In this review article, we discuss the role of different HSPs in prostate diseases, and their potential as therapeutic targets.  相似文献

The relationship between the structure and the antiradical and antioxidant activities of three anthocyanidins, namely peonidin, petunidin, and delphinidin, and their glucosides was investigated in this study. The ability of anthocyanins to scavenge free radicals was determined using DPPH assay, whereas the inhibition of peroxidation in liposomes in relation to a model membrane that imitated the composition of a lipid membrane in tumor cells was specified using the fluorimetric method. To explore this issue at the atomistic level, density functional theory studies were applied. It was shown that glycosides performed better than anthocyanidins in protecting membranes against oxidation. The highest redox potential was demonstrated by anthocyanidins with the highest number of hydroxyl groups in the B ring in the order as follows: (Dp > Pt > Pn), and the same relationship was proven for their glucosides. The majority of the compounds studied here proved to be better antioxidants than ascorbic acid. They showed consistent electrodonating properties and though the f-HAT mechanism became more feasible with each consecutive deprotonation. Glycosylation did not have a direct impact on reactivity, apart from peonidin and petunidin in the study of which it was found that this process was responsible for lifting off steric hindrance between B and C rings and rendering certain pathways more feasible. Kinetic and molecular dynamics are essential to properly describe the membrane’s lipid oxidation.  相似文献

The term Western diet (WD) describes the consumption of large amounts of highly processed foods, rich in simple sugars and saturated fats. Long-term WD feeding leads to insulin resistance, postulated as a risk factor for Alzheimer’s disease (AD). AD is the main cause of progressive dementia characterized by the deposition of amyloid-β (Aβ) plaques and neurofibrillary tangles consisting of the hyperphosphorylated tau (p-Tau) protein in the brain, starting from the entorhinal cortex and the hippocampus. In this study, we report that WD-derived impairment in insulin signaling induces tau and Aβ brain pathology in wild-type C57BL/6 mice, and that the entorhinal cortex is more sensitive than the hippocampus to the impairment of brain insulin signaling. In the brain areas developing WD-induced insulin resistance, we observed changes in p-Tau(Thr231) localization in neuronal subcellular compartments, indicating progressive tauopathy, and a decrease in amyloid precursor protein levels correlating with the appearance of Aβ peptides. These results suggest that WD promotes the development of AD and may be considered not only a risk factor, but also a modifiable trigger of AD.  相似文献

In this work, sulfonated polyetheretherketone (S-PEEK)-based coatings, nanocrystalline ZnS and hydroxyapatite (n-HA) particles were developed on Zr-2.5Nb zirconium alloy substrates by electrophoretic deposition (EPD) combined with subsequent heat treatment. The properties of suspensions and deposition kinetics were studied. Cationic chitosan polyelectrolyte ensured the stabilization of the suspension and allowed for the co-deposition of all coating components on the cathode. The heating of the coated samples at a temperature of 450 °C and slow cooling resulted in sulfonation of the PEEK and the formation of dense coatings. The coatings were characterized by high roughness, hardness, modulus of elasticity and adhesion strength. The coatings revealed mild hydrophilicity, improved the electrochemical corrosion resistance of the alloy and induced the formation of hydroxyapatite with a cauliflower-like morphology on its surface during the Kokubo test. This work explored the great development potential of advanced sulfonated PEEK-based coatings, incorporating antibacterial and bioactive components by EPD combined with heat treatment to stimulate the surface properties of zirconium alloy for prospective dental and orthopedic applications. The antibacterial and osteoconductive properties of the obtained coatings should be further investigated.  相似文献

African swine fever (ASF) is one of the most dangerous hemorrhagic infectious diseases that affect domestic and wild pigs. Currently, neither a vaccine nor effective treatments are available for this disease. As regards the degree of virulence, ASFV strains can be divided into high, moderate, or low virulence. The main detection methods are based on the use of the polymerase chain reaction (PCR). In order to prevent an uncontrolled spread of ASF, new on-site techniques that can enable the identification of an early-stage disease are needed. We have developed a specific immunological SPR-based assay for ASFV antigen detection directly in liquid samples. The developed assay allows us to detect the presence of ASFV at the dose of 103 HAD50/mL.  相似文献

The development of flat, compact beam‐steering devices with no bulky moving parts is opening up a new route to a variety of exciting applications, such as LIDAR scanning systems for autonomous vehicles, robotics and sensing, free‐space, and even surface wave optical signal coupling. In this paper, the design, fabrication and characterization of innovative, nonvolatile, and reconfigurable beam‐steering metadevices enabled by a combination of optical metasurfaces and chalcogenide phase‐change materials is reported. The metadevices reflect an incident optical beam in a mirror‐like fashion when the phase‐change layer is in the crystalline state, but reflect anomalously at predesigned angles when the phase‐change layer is switched into its amorphous state. Experimental angle‐resolved spectrometry measurements verify that fabricated devices perform as designed, with high efficiencies, up to 40%, when operating at 1550 nm. Laser‐induced crystallization and reamorphization experiments confirm reversible switching of the device. It is believed that reconfigurable phase‐change‐based beam‐steering and beam‐shaping metadevices, such as those reported here, can offer real applications advantages, such as high efficiency, compactness, fast switching times and, due to the nonvolatile nature of chalcogenide phase‐change materials, low power consumption.  相似文献

Al2O3 films were deposited using atomic layer deposition (ALD) and ultrasonic spray pyrolysis (USP) methods on p- and n-type Si substrates, n-type 4H–SiC substrates and 4H–SiC diodes for passivation studies. UV exposure in N2 atmosphere and 5% HF treatment were used as two separate surface preparation procedures prior to Al2O3 deposition. The films deposited with USP technique contain a large amount of fixed negative charge and are vulnerable to water incorporation into the material. The Al2O3 film prepared by ALD method shows much better uniformity and less negative charge. Decrease of the leakage current in the 4H–SiC diodes is observed after Al2O3 passivation using both methods.  相似文献
