Inflammatory bowel diseases (IBD) are commonly considered as Crohn’s disease and ulcerative colitis, but the possibility that the alterations in gut microbiota and oxidative stress may affect the course of experimental colitis in obese physically exercising mice treated with the intestinal alkaline phosphatase (IAP) has been little elucidated. Mice fed a high-fat-diet (HFD) or normal diet (ND) for 14 weeks were randomly assigned to exercise on spinning wheels (SW) for 7 weeks and treated with IAP followed by intrarectal administration of TNBS. The disease activity index (DAI), grip muscle strength test, oxidative stress biomarkers (MDA, SOD, GSH), DNA damage (8-OHdG), the plasma levels of cytokines IL-2, IL-6, IL-10, IL-12p70, IL-17a, TNF-α, MCP-1 and leptin were assessed, and the stool composition of the intestinal microbiota was determined by next generation sequencing (NGS). The TNBS-induced colitis was worsened in obese sedentary mice as manifested by severe colonic damage, an increase in DAI, oxidative stress biomarkers, DNA damage and decreased muscle strength. The longer running distance and weight loss was observed in mice given IAP or subjected to IAP + SW compared to sedentary ones. Less heterogeneous microbial composition was noticed in sedentary obese colitis mice and this effect disappeared in IAP + SW mice. Absence of Alistipes, lower proportion of Turicibacter, Proteobacteria and Faecalibacterium, an increase in Firmicutes and Clostridium, a decrease in oxidative stress biomarkers, 8-OHdG content and proinflammatory cytokines were observed in IAP + SW mice. IAP supplementation in combination with moderate physical activity attenuates the severity of murine colitis complicated by obesity through a mechanism involving the downregulation of the intestinal cytokine/chemokine network and oxidative stress, the modulation of the gut microbiota and an improvement of muscle strength. 相似文献

The bias against women in academia is a documented phenomenon that has had detrimental consequences, not only for women, but also for the quality of science. First, gender bias in academia affects female scientists, resulting in their underrepresentation in academic institutions, particularly in higher ranks. The second type of gender bias in science relates to some findings applying only to male participants, which produces biased knowledge. Here, we identify a third potentially powerful source of gender bias in academia: the bias against research on gender bias. In a bibliometric investigation covering a broad range of social sciences, we analyzed published articles on gender bias and race bias and established that articles on gender bias are funded less often and published in journals with a lower Impact Factor than articles on comparable instances of social discrimination. This result suggests the possibility of an underappreciation of the phenomenon of gender bias and related research within the academic community. Addressing this meta-bias is crucial for the further examination of gender inequality, which severely affects many women across the world. 相似文献

The influence of hydrophobicity on antibacterial activity versus the effect on the viability of mammalian cells for peptide/peptoid hybrids was examined for oligomers based on the cationic Lys-like peptoid residue combined with each of 28 hydrophobic amino acids in an alternating sequence. Their relative hydrophobicity was correlated to activity against both Gram-negative and Gram-positive species, human red blood cells, and HepG2 cells. This identified hydrophobic side chains that confer potent antibacterial activity (e. g., MICs of 2–8 μg/mL against E. coli) and low toxicity toward mammalian cells (<10 % hemolysis at 400 μg/mL and IC50>800 μg/mL for HepG2 viability). Most peptidomimetics retained activity against drug-resistant strains. These findings corroborate the hypothesis that for related peptidomimetics two hydrophobicity thresholds may be identified: i) it should exceed a certain level in order to confer antibacterial activity, and ii) there is an upper limit, beyond which cell selectivity is lost. It is envisioned that once identified for a given subclass of peptide-like antibacterials such thresholds can guide further optimisation. 相似文献

ATR, a DNA damage signaling kinase, is required for cell cycle checkpoint regulation and detecting DNA damage caused by genotoxic factors including Al3+ ions. We analyzed the function of the HvATR gene in response to chemical clastogen-maleic acid hydrazide (MH). For this purpose, the Al-tolerant barley TILLING mutant hvatr.g was used. We described the effects of MH on the nuclear genome of hvatr.g mutant and its WT parent cv. “Sebastian”, showing that the genotoxic effect measured by TUNEL test and frequency of cells with micronuclei was much stronger in hvatr.g than in WT. MH caused a significant decrease in the mitotic activity of root cells in both genotypes, however this effect was significantly stronger in “Sebastian”. The impact of MH on the roots cell cycle, analyzed using flow cytometry, showed no differences between the mutant and WT. 相似文献

Due to the growing stock of torrent control structures and decreasing investments, the protection level can be reduced in the long term. Therefore, the focus of the future investments has been shifted on efficiency improvement and maintenance of existing structures. The maintenance of the existing structures is necessary to ensure safety from natural hazards, which are increasing as a consequence of the climate change. Due to the increasing number of structures, there is a strong need to develop a good maintenance management for these infrastructure works. This study is based on data from the condition rating programme of the Austrian Service for Torrent and Avalanche Control. For the structures relevant in this work, 75,343 records were available. The data were examined for different construction type and construction materials. The reliability of the structures was determined by means of the Markov chain. The study describes a homogeneous Markov chain model formed by the data of the condition rating. The result of this study is a proposal for a time to failure (TTF) and a time to repair (TTR) for several construction types with different construction materials as well as the time-dependent use of construction materials in the sector of the Austrian torrent control infrastructure. 相似文献

People are an important source of pollution indoors, through activities such as cleaning, and also from “natural” emissions from breath and skin. This paper investigates natural emissions in high‐occupancy environments. Model simulations are performed for a school classroom during a typical summer in a polluted urban area. The results show that classroom occupants have a significant impact on indoor ozone, which increases from ~9 to ~20 ppb when the pupils leave for lunch and decreases to ~14 ppb when they return. The concentrations of 4‐OPA, formic acid, and acetic acid formed as oxidation products following skin emissions attained maximum concentrations of 0.8, 0.5, and 0.1 ppb, respectively, when pupils were present, increasing from near‐zero concentrations in their absence. For acetone, methanol, and ethanol from breath emissions, maximum concentrations were ~22.3, 6.6, and 21.5 ppb, respectively, compared to 7.4, 2.1, and 16.9 ppb in their absence. A rate of production analysis showed that occupancy reduced oxidant concentrations, while enhancing formation of nitrated organic compounds, owing to the chemistry that follows from increased aldehyde production. Occupancy also changes the peroxy radical composition, with those formed through isoprene oxidation becoming relatively more important, which also has consequences for subsequent oxidant concentrations. 相似文献
