Lipedema is an adipose tissue disorder characterized by the disproportionate increase of subcutaneous fat tissue in the lower and/or upper extremities. The underlying pathomechanism remains unclear and no molecular biomarkers to distinguish the disease exist, leading to a large number of undiagnosed and misdiagnosed patients. To unravel the distinct molecular characteristic of lipedema we performed lipidomic analysis of the adipose tissue and serum of lipedema versus anatomically- and body mass index (BMI)-matched control patients. Both tissue groups showed no significant changes regarding lipid composition. As hyperplastic adipose tissue represents low-grade inflammation, the potential systemic effects on circulating cytokines were evaluated in lipedema and control patients using the Multiplex immunoassay system. Interestingly, increased systemic levels of interleukin 11 (p = 0.03), interleukin 28A (p = 0.04) and interleukin 29 (p = 0.04) were observed. As cytokines can influence metabolic activity, the metabolic phenotype of the stromal vascular fraction was examined, revealing significantly increased mitochondrial respiration in lipedema. In conclusion, despite sharing a comparable lipid profile with healthy adipose tissue, lipedema is characterized by a distinct systemic cytokine profile and metabolic activity of the stromal vascular fraction.  相似文献

Chronic inflammation has been associated with several chronic diseases, such as age-related macular degeneration (AMD). The NLRP3 inflammasome is a central proinflammatory signaling complex that triggers caspase-1 activation leading to the maturation of IL-1β. We have previously shown that the inhibition of the chaperone protein, Hsp90, prevents NLRP3 activation in human retinal pigment epithelial (RPE) cells; these are cells which play a central role in the pathogenesis of AMD. In that study, we used a well-known Hsp90 inhibitor geldanamycin, but it cannot be used as a therapy due to its adverse effects, including ocular toxicity. Here, we have tested the effects of a novel Hsp90 inhibitor, TAS-116, on NLRP3 activation using geldanamycin as a reference compound. Using our existing protocol, inflammasome activation was induced in IL-1α-primed ARPE-19 cells with the proteasome and autophagy inhibitors MG-132 and bafilomycin A1, respectively. Intracellular caspase-1 activity was determined using a commercial caspase-1 activity kit and the FLICA assay. The levels of IL-1β were measured from cell culture medium samples by ELISA. Cell viability was monitored by the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) test and lactate dehydrogenase (LDH) measurements. Our findings show that TAS-116 could prevent the activation of caspase-1, subsequently reducing the release of mature IL-1β. TAS-116 has a better in vitro therapeutic index than geldanamycin. In summary, TAS-116 appears to be a well-tolerated Hsp90 inhibitor, with the capability to prevent the activation of the NLRP3 inflammasome in human RPE cells.  相似文献

Exposure to moisture‐damaged indoor environments is associated with adverse respiratory health effects, but responsible factors remain unidentified. In order to explore possible mechanisms behind these effects, the oxidative capacity and hemolytic activity of settled dust samples (n = 25) collected from moisture‐damaged and non‐damaged schools in Spain, the Netherlands, and Finland were evaluated and matched against the microbial content of the sample. Oxidative capacity was determined with plasmid scission assay and hemolytic activity by assessing the damage to isolated human red blood cells. The microbial content of the samples was measured with quantitative PCR assays for selected microbial groups and by analyzing the cell wall markers ergosterol, muramic acid, endotoxins, and glucans. The moisture observations in the schools were associated with some of the microbial components in the dust, and microbial determinants grouped together increased the oxidative capacity. Oxidative capacity was also affected by particle concentration and country of origin. Two out of 14 studied dust samples from moisture‐damaged schools demonstrated some hemolytic activity. The results indicate that the microbial component connected with moisture damage is associated with increased oxidative stress and that hemolysis should be studied further as one possible mechanism contributing to the adverse health effects of moisture‐damaged buildings.  相似文献

Burnishing is a cold working process that can be used to improve surface finish and surface hardness of workpieces. Conventionally, diamond or hard metal burnishing tools are used. In the present work, a novel burnishing tool was created by depositing amorphous diamond coating (AD) on a stainless steel tip. This tool was used to improve the surface finish and surface hardness of Nitronic-50 HS stainless steel workpieces. Nitronic-50 HS is used in a wide range of applications in industry. The burnishing process was carried out at different burnishing parameters (force, revolution speed, feed and number of tool passes). Burnishing parameters had a significant effect on the finishing process and they had to be optimized to achieve the best results. Remarkable improvements in surface finish (70% decrease in roughness) and hardness (25% increase) could be achieved with this tool and process in the surface finishing of Nitronic-50 stainless steel workpieces. From the tribological point of view, the AD-coated tip performed slightly better than a corresponding tip modified from a commercial polycrystalline diamond tip. AD coating seems to be very suitable for use in mechanical surface finishing tools such as a burnishing tip.  相似文献

Thermal fatigue damage of flip chip solder joints is a serious reliability concern, although it usually remains tolerable with the flip chip connections (of smaller chips) to ceramic boards as practiced by IBM for over a quarter century. However, the recent trend in microelectronics packaging towards bonding large chips or ceramic modules to organic boards means a larger differential thermal expansion mismatch between the board and the chip or ceramic module. To reduce the thermal stresses and strains at solder joints, a polymer underfill is customarily added to fill the cavity between the chip or module and the organic board. This procedure has typically at least resulted in an increase of the thermal fatigue life by a factor of 10, as compared to the non-underfilled case. In this contribution, we first discuss the effects of the underfill to reduce solder joint stresses and strains, as well as underfill effects on fatigue crack propagation based on a finite element analysis. Secondly, we probe the question of the importance of the effects of underfill defects, particularly that of its delamination from the chip side, on the effectiveness of the underfill to increase thermal fatigue life. Finally, we review recent experimental evidence from thermal cycling of actual flip chip modules which appears to support the predictions of our model.  相似文献

The results from four compensatory fertilization experiments located at different distances (0.5, 2, 4 and 8 km) along a heavy metal deposition gradient extending from the Harjavalta Cu-Ni smelter in SW Finland are presented. The experiments were established in middle-age Scots pine stands growing on dryish sites of sorted glaciofluvial sediments. The soil type in all the experiments is ferric podsol. The treatments in the experiments consisted of liming, a powdered slow-release mineral mixture and stand-specific fertilization which comprised at least methylene urea and ammonium nitrate.Monitoring of deposition and soil solution and studies on soil chemical and microbiological properties, on the nutrient status of trees and needle litterfall, on fine root dynamics and on the growth of the tree stands were carried out during a 5-year period.There was a severe shortage of exchangeable Ca and Mg in the organic layer of the most polluted stands. Although the uppermost mineral soil layer had relatively high exchangeable Ca and Mg concentrations, the trees were not able to utilize these nutrient reserves presumably due to the toxic effects of Cu and Ni on the plant roots and mycorrhizas.The treatments that included limestone markedly decreased the Cu and Ni concentrations in the soil solution and soil organic layer, presumably due to immobilisation through precipitation or absorption. The Ca and Mg concentrations correspondingly increased, which certainly contributed to the partial recovery of fine root and stand growth. The powdered mineral mixture and the combination of methylene urea and ammonium nitrate had no short-term effect on the microbial biomass and activity. All the fertilizer treatments increased volume growth in the most polluted stand. The stand-specific fertilization increased needle mass in heavily polluted stands, but the response of the needle mass to fertilizer treatments was low in the less polluted stands. No clear evidence was found to support the role of nutrient status in tree resistance.  相似文献

Although fatty acid composition is the most important attribute used to control oxidation stability, all edible oils are affected by lipid oxidation irrespective of whether they are highly unsaturated or not. The aim of the study was to compare the oxidation of rapeseed oil (RO) and butter oil (BO) triacylglycerols (TAGs) and their mixtures containing 10% or 20% of the other. Oxidation of the TAGs at 40°C was followed by formation of primary and secondary products. Statistical methods were used to interpret the data. The RO and BO TAGs and their mixtures began to oxidise without any induction periods. In the RO TAGs more hydroperoxides and p‐anisidine reactive compounds were formed than in the BO TAGs. The BO TAGs oxidised more than would be expected by their fatty acid composition. High susceptibility of BO TAGs to oxidation was caused by the easy breakdown of their hydroperoxides. Heptadienal and heptenal were specific products of oxidised RO TAGs and heptanal and nonenal of oxidised BO TAGs. Mixtures of RO and BO TAGs behaved according to which was dominant in the mixture. However, as little as 10% of RO or BO TAG introduced its specific oxidation products to the mixture. © 1999 Society of Chemical Industry  相似文献

A range of probiotic and other intestinal bacteria were examined for their ability to ferment the dietary fibre carbohydrates β‐glucan, xylan, xylo‐oligosaccharides (XOS) and arabinoxylan. β‐Glucan was fermented by Bacteroides spp and Clostridium beijerinckii but was not fermented by lactobacilli, bifidobacteria, enterococci or Escherichia coli. Unsubstituted xylan was not fermented by any of the probiotic bacteria examined. However, many Bifidobacterium species and Lactobacillus brevis were able to grow to high yields using XOS. XOS were also efficiently fermented by some Bacteroides isolates but not by E coli, enterococci, Clostridium difficile, Clostridium perfringens or by the majority of intestinal Lactobacillus species examined. Bifidobacterium longum strains were able to grow well using arabinoxylan as the sole carbon source. These organisms hydrolysed and fermented the arabinosyl residues from arabinoxylan but did not substantially utilise the xylan backbone of the polysaccharide. Arabinoxylan was not fermented by lactobacilli, enterococci, E coli, C perfringens or C difficile and has potential to be an applicable carbohydrate to complement probiotic Bif longum strains in synbiotic combinations. © 2002 Society of Chemical Industry  相似文献

The acute toxicity and the genotoxicity of four colored smokes were studied by an in vitro method based on the exposure of human bronchial epithelial cell cultures to the smokes. All smoke formulations consisted of the oxidizer fuel mixture (potassium chlorate/lactose), talcum and the following dyes: 1,4‐dihydroxy anthraquinone (orange), 1‐(p‐tolylamino)‐4‐hydroxy anthraquinone (violet), 1‐methylamino anthraquinone (red) and 4,4′‐methylidyne‐bis‐3‐methyl‐1‐phenyl‐2‐pyrazolin‐5‐one (yellow). The experiments were carried out in a laboratory scale chamber and in a large container. The toxicity was compared to that of hexachloroethane (HC)‐based reference smoke with known toxicity. All the colored smokes displayed acute toxicity. The order of toxicity in the laboratory scale tests was orange>violet≈red>HC>yellow and in the container tests orange>violet≈yellow>red. The orange smoke appeared genotoxic in all the tests. With the yellow and the violet smokes, the genotoxicity could not be totally excluded. The red smoke showed evidence of weak genotoxicity only in one test series at the highest concentration level.  相似文献
