The association of immune markers and clinicopathologic features and patient outcome has not been extensively studied in Merkel cell carcinoma (MCC). We correlated tumoral PD-L1 and IDO1 expression, and intratumoral CD8+ and FoxP3+ lymphocytes count with clinicopathologic variables, Merkel cell polyomavirus (MCPyV) status, and patient outcomes in a series of 132 MCC. By univariate analyses, tumoral PD-L1 expression >1% and combined tumoral PD-L1 >1% and high intratumoral FoxP3+ lymphocyte count correlated with improved overall survival (OS) (p = 0.016, 0.0072), MCC-specific survival (MSS) (p = 0.019, 0.017), and progression-free survival (PFS) (p = 0.043, 0.004, respectively). High intratumoral CD8+ and FoxP3+ lymphocyte count correlated with longer MSS (p = 0.036) and improved PFS (p = 0.047), respectively. Ulceration correlated with worse OS and worse MSS. Age, male gender, and higher stage (3 and 4) significantly correlated with worse survival. MCPyV positivity correlated with immune response. By multivariate analyses, only ulceration and age remained as independent predictors of worse OS; gender and stage remained for shorter PFS. Tumoral PD-L1 expression and increased density of intratumoral CD8+ lymphocytes and FoxP+ lymphocytes may represent favorable prognosticators in a subset of MCCs. Tumoral PD-L1 expression correlated with intratumoral CD8+ and FoxP3+ lymphocytes, which is supportive of an adaptive immune response.  相似文献

Surface offset is one of the most useful operations in Computer Aided Geometric Design (CAGD). However, an implementation of this operator is not trivial primarily because the offset surface, in general, does not have the same representation as the original surface. Hence, it is difficult or impossible to represent an exact offset in a system with limited surface forms. For this reason, some CAGD surface offset operators produce results that are, at times, unsatisfactory. In this article, we discuss surface offset approximation methods in B-spline environment: both the original surfaces and their offsets are B-spline surfaces. We have examined the 16-year time series of particulate organic carbon (POC) concentration in the surface waters of the North Atlantic derived from SeaWiFS and MODIS-Aqua data. The annual mean POC concentrations are the highest in the northern North Atlantic, reaching 120 mg m?3. Moving south, the mean annual POC concentrations decrease to minimum values of about 30 mg m?3 at around 30° N and increase in the equatorial region to about 70 mg m?3. The seasonal amplitude of POC concentration in the northern North Atlantic region is larger when compared to other regions. The annual mean surface POC concentrations in the entire North Atlantic basin show a statistically significant trend with an average decrease of 0.79 mg m?3 year?1. Regionally averaged 16-year mean POC biomass integrated over the optical depth, euphotic depth, and mixed-layer depth is estimated at about 1.27, 4.34, and 4.59 g m?2, respectively. Even larger biomass of 6.26 g m?2 is estimated if one chooses to use in the calculations the greatest from the daily values of the estimates listed above at each pixel of the satellite data. Comparisons of POC biomass with primary productivity allowed us to assess temporal and spatial patterns of POC losses.  相似文献

Neurodegenerative disorders such as Alzheimer’s disease (AD) are distinguished by the irreversible degeneration of central nervous system function and structure. AD is characterized by several different neuropathologies—among others, it interferes with neuropsychiatrical controls and cognitive functions. This disease is the number one neurodegenerative disorder; however, its treatment options are few and, unfortunately, ineffective. In the new strategies devised for AD prevention and treatment, the application of plant-based natural products is especially popular due to lesser side effects associated with their taking. Moreover, their neuroprotective activities target different pathological mechanisms. The current review presents the anti-AD properties of several natural plant substances. The paper throws light on products under in vitro and in vivo trials and compiles information on their mechanism of actions. Knowledge of the properties of such plant compounds and their combinations will surely lead to discovering new potent medicines for the treatment of AD with lesser side effects than the currently available pharmacological proceedings.  相似文献

The author's research on the time-domain methods for electromagnetic simulation is reported with focus on the expanded-node FDTD and TLM approaches. The theorem of formal equivalence of FDTD and TLM together with its fundamental consequences (published recently or yet unpublished) is summarized. As an example of application of the theorem, a new family of nonequidistant TLM algorithms with controlled stability margin is originally derived by transformation from FDTD. Several meshing techniques for the modeling of irregular geometries are compared, and the locally modified grids based on local integral approximations are shown to provide optimum accuracy to computer resources ratio for practical microwave problems. © 1996 John Wiley & Sons, Inc.  相似文献

High-grade serous ovarian carcinoma (HGSOC) is the most frequent and malignant form of ovarian cancer. A local renin–angiotensin system (RAS) has been found in the ovary, and changes in selected components of this system were observed in pathological states and also in ovarian cancer. In the present study, we examined the effect of three peptides, Ang-(1-7), Ang-(1-9) and Ang-(3-7), on proliferation and motility of the OVPA8 cell line, a new well-defined and preclinical model of HGSOC. We confirmed the presence of mRNA for all angiotensin receptors in the tested cells. Furthermore, our findings indicate that all tested angiotensin peptides increased the metabolic serum in the medium by activation of cell defense mechanisms such as nuclear factor kappaB signaling pathway andapoptosis. Moreover, tested angiotensin peptides intensified serum starvation-induced cell cycle arrest at the G0/G1 phase. In the case of Ang-(3-7), a significant decrease in the number of Ki67 positive cells (Ki67+) and reduced percentage of activated ERK1/2 levels in ovarian cancer cells were additionally reported. The angiotensin-induced effect of the accumulation of cells in the G0/G1 phase was not observed in OVPA8 cells growing on the medium with 10% FBS. Moreover, in the case of Ang-(3-7), the tendency was quite the opposite. Ang-(1-7) but not Ang-(1-9) or Ang-(3-7) increased the mobility of reluctant-to-migrate OVAP8 cells cultured in the serum-free medium. In any cases, the changes in the expression of VIM and HIF1A gene, associated with epithelial–mesenchymal transition (EMT), were not observed. In conclusion, we speculate that the adaptation to starvation in nutrient-deprived tumors can be modulated by peptides from the renin–angiotensin system. The influence of angiotensin peptides on cancer cells is highly dependent on the availability of growth factors and nutrients.  相似文献

The CMG complex (Cdc45, Mcm2–7, GINS (Psf1, 2, 3, and Sld5)) is crucial for both DNA replication initiation and fork progression. The CMG helicase interaction with the leading strand DNA polymerase epsilon (Pol ε) is essential for the preferential loading of Pol ε onto the leading strand, the stimulation of the polymerase, and the modulation of helicase activity. Here, we analyze the consequences of impaired interaction between Pol ε and GINS in Saccharomyces cerevisiae cells with the psf1-100 mutation. This significantly affects DNA replication activity measured in vitro, while in vivo, the psf1-100 mutation reduces replication fidelity by increasing slippage of Pol ε, which manifests as an elevated number of frameshifts. It also increases the occurrence of single-stranded DNA (ssDNA) gaps and the demand for homologous recombination. The psf1-100 mutant shows elevated recombination rates and synthetic lethality with rad52Δ. Additionally, we observe increased participation of DNA polymerase zeta (Pol ζ) in DNA synthesis. We conclude that the impaired interaction between GINS and Pol ε requires enhanced involvement of error-prone Pol ζ, and increased participation of recombination as a rescue mechanism for recovery of impaired replication forks.  相似文献

A series of TiO2 photocatalysts were obtained using several calcination temperatures ranging from 350 to 750 °C. The photocatalysts’ characteristics by X-ray diffraction, UV–vis and FTIR diffuse reflectance spectroscopies, X-ray photoelectron spectroscopy, BET and BJH methods showed that sample active in vis region had anatase structure, about 200 m2/g specific surface area, absorbed light for λ > 400 nm and contained 10.1 at.% of C–C species. The photocatalytic activity of the catalysts was estimated by measuring the decomposition rate of phenol in 0.21 mM aqueous solution in visible and ultraviolet light. The experimental data clearly indicate correlation between the absorption intensity of irradiation by obtained powders and their photocatalytic performance in phenol degradation. An increase in absorbance over the entire vis region and the highest photocatalytic activity for phenol degradation in visible light (λ > 400 nm) occurred for photocatalyst calcinated at 350 °C. Photocatalyst processed at 450 °C had the best activity in UV light (250 < λ < 400 nm).  相似文献

Surfaces of bulk platinum and unsupported (Vulcan-free) Pt nanoparticles, that are modified and stabilized with such Keggin type heteropolyacids of molybdenum and tungsten as H3PMo12O40 and H3PW12O40, have been characterized using cyclic voltammetry, FTIR (by reflectance), as well as transmission and scanning electron microscopies. The presence of the polyoxometallate monolayer on platinum results in the partial suppression of the interfacial formation of PtOH/PtO oxides. Both molybdates and tungstates seem to interact with Pt surface via their corner oxygen atoms. The existence of spacious, largely hydrated, polyoxometallate monolayers on platinum does not block access of reactant (oxygen) to the catalytic Pt sites. The electrocatalytic properties of H3PMo12O40 and H3PW12O40 modified Pt nanoparticles towards reduction of oxygen in acid medium have been examined and compared using rotating ring-disk voltammetry. Reactivity of Pt-free H3PMo12O40 and H3PW12O40 with respect to reduction of hydrogen peroxide has also been considered. Our results clearly show that modification of Pt nanoparticles with PW12 (but not with PMo12) results in the enhancement of the electrocatalytic reduction of oxygen.  相似文献
