High prevalence of both criteria and extra-criteria antiphospholipid antibodies (aPL) has been reported in COVID-19 patients. However, the differences in aPL prevalence decreased when an age-matched control group was included. The association of aPL with thrombotic events in COVID-19 is very heterogeneous. This could be influenced by the fact that most of the studies carried out were conducted on small populations enriched with elderly patients in which aPL was measured only at a single point and they were performed with non-standardized assays. The few studies that confirmed aPL in a second measurement showed that aPL levels hardly changed, with the exception of the lupus anticoagulant that commonly reduced. COVID-19 coagulopathy is an aPL-independent phenomenon closely associated with the onset of the disease. Thrombosis occurs later in patients with aPL presence, which is likely an additional prothrombotic factor. B2-glycoprotein deficiency (mainly aPL antigen caused both by low production and consumption) is very common during the SARS-CoV2 infection and has been associated with a greater predisposition to COVID-19 complications. Mass spectrometry (MS)-based techniques can be a powerful tool to identify neuropsychiatric disorder biomarkers, improving prediction and diagnosis ability. Here, we evaluate the efficacy of MS proteomics applied to human peripheral fluids of schizophrenia (SCZ) patients to identify disease biomarkers and relevant networks of biological pathways. Following PRISMA guidelines, a search was performed for studies that used MS proteomics approaches to identify proteomic differences between SCZ patients and healthy control groups (PROSPERO database: CRD42021274183). Nineteen articles fulfilled the inclusion criteria, allowing the identification of 217 differentially expressed proteins. Gene ontology analysis identified lipid metabolism, complement and coagulation cascades, and immune response as the main enriched biological pathways. Meta-analysis results suggest the upregulation of FCN3 and downregulation of APO1, APOA2, APOC1, and APOC3 in SCZ patients. Despite the proven ability of MS proteomics to characterize SCZ, several confounding factors contribute to the heterogeneity of the findings. In the future, we encourage the scientific community to perform studies with more extensive sampling and validation cohorts, integrating omics with bioinformatics tools to provide additional comprehension of differentially expressed proteins. The produced information could harbor potential proteomic biomarkers of SCZ, contributing to individualized prognosis and stratification strategies, besides aiding in the differential diagnosis. 相似文献

The skin is the largest organ in the human body, comprising the main barrier against the environment. When the skin loses its integrity, it is critical to replace it to prevent water loss and the proliferation of opportunistic infections. For more than 40 years, tissue-engineered skin grafts have been based on the in vitro culture of keratinocytes over different scaffolds, requiring between 3 to 4 weeks of tissue culture before being used clinically. In this study, we describe the development of a polymerizable skin hydrogel consisting of keratinocytes and fibroblast entrapped within a fibrin scaffold. We histologically characterized the construct and evaluated its use on an in vivo wound healing model of skin damage. Our results indicate that the proposed methodology can be used to effectively regenerate skin wounds, avoiding the secondary in vitro culture steps and thus, shortening the time needed until transplantation in comparison with other bilayer skin models. This is achievable due to the instant polymerization of the keratinocytes and fibroblast combination that allows a direct application on the wound. We suggest that the polymerizable skin hydrogel is an inexpensive, easy and rapid treatment that could be transferred into clinical practice in order to improve the treatment of skin wounds. 相似文献

To survive in today’s competitive global market, companies must perform strategic changes in order to increase productivity, eliminating wasted materials, time, and effort. This study will examine how to optimize the time and effort required to supply raw material to different production lines in a manufacturing plant in Juarez, Mexico by minimizing the distance an operator must travel to distribute material from a warehouse to a set of different production lines with corresponding demand. The core focus of this study is similar to that of the Vehicle Routing Problem in that it is treated as a combinatorial optimization problem. The artificial bee colony algorithm is applied in order to find the optimal distribution of material with the aim of establishing a standard time for this duty by examining how this is applied in a local manufacturing plant. Managing project portfolios has been a challenge to many IT organizations due to the size and complexity of their initiatives that are often cross-functional, fast changing, and transformational in nature. A governance process on project solicitation, evaluation, and monitoring is thus essential to ensure the resulting portfolio creates tangible values, balances across priorities, and supports business objectives. An optimization model to streamline the decision processes for IT portfolios and programs is proposed. We consider project characteristics such as the extent of strategic alignment, expected benefit, development cost, and cross-project synergy to maximize the portfolio value. We also consider team proficiency and resource availability to determine a project portfolio that could be implemented within the overall development time. The multi-objective model identifies the optimal mix among project types and the solution procedure efficiently produces recommendations that are superior to those found with current empirical techniques. We also describe an evolutionary algorithm to find approximate solutions to the optimization model. Possible extensions on how the optimization procedure can go beyond projects to also streamline decisions such as the renewal or replacement of in-flight applications is discussed. 相似文献
