Genomic diversity of Portuguese accessions of Avena species—diploid A. strigosa and hexaploids A. sativa and A. sterilis—was evaluated through molecular and cytological analysis of 45S rDNA, and other repetitive sequences previously studied in cereal species—rye subtelomeric sequence (pSc200) and cereal centromeric sequence (CCS1). Additionally, retrotransposons and microsatellites targeting methodologies—IRAP (inter-retrotransposon amplified polymorphism) and REMAP (retrotransposon-microsatellite amplified polymorphism)—were performed. A very high homology was detected for ribosomal internal transcribed sequences (ITS1 and ITS2) between the species analyzed, although nucleolar organizing regions (NOR) fluorescent in situ hybridization (FISH) analysis revealed distinct number of Nor loci between diploid and hexaploid species. Moreover, morphological diversity, evidenced by FISH signals with different sizes, was observed between distinct accessions within each species. pSc200 sequences were for the first time isolated from Avena species but proven to be highly similar in all genotypes analyzed. The use of primers designed for CCS1 unraveled a sequence homologous to the Ty3/gypsy retrotransposon Cereba, that was mapped to centromeric regions of diploid and hexaploid species, being however restricted to the more related A and D haplomes. Retrotransposon-based methodologies disclosed species- and accessions-specific bands essential for the accurate discrimination of all genotypes studied. Centromeric, IRAP and REMAP profiles therefore allowed accurate assessment of inter and intraspecific variability, demonstrating the potential of these molecular markers on future oat breeding programs. 相似文献

Mass spectrometry-based untargeted metabolomics often results in the observation of hundreds to thousands of features that are differentially regulated between sample classes. A major challenge in interpreting the data is distinguishing metabolites that are causally associated with the phenotype of interest from those that are unrelated but altered in downstream pathways as an effect. To facilitate this distinction, here we describe new software called metaXCMS for performing second-order (\"meta\") analysis of untargeted metabolomics data from multiple sample groups representing different models of the same phenotype. While the original version of XCMS was designed for the direct comparison of two sample groups, metaXCMS enables meta-analysis of an unlimited number of sample classes to facilitate prioritization of the data and increase the probability of identifying metabolites causally related to the phenotype of interest. metaXCMS is used to import XCMS results that are subsequently filtered, realigned, and ultimately compared to identify shared metabolites that are up- or down-regulated across all sample groups. This study is aimed at the thermal analysis of sealant mortar (usually a mixtures of bentonite and cemem with addition of sand) used in geothermal cooling and heating. In particular, thermal conductivity and diffusivity measurements were performed on differem sealant mixtures by using Hot Disk thermal constants analyzer in order to identify the interesting thermal properties of grouting materials. The grouting materials that we considered are of porous nature and, if used in the presence of groundwater, have different levels of imbibitions. It is important to know the thermal behavior of these materials at different water content. A first set of measurements was performed on a not-tinted material at room temperature; then the samples were led to saturation conditions by contact capillary imbibitions with a cotton wool layer moistened in water. The determination of thermal conductivity in these test conditions appears to be critical compared to the measuremems on non-timed sample. The thermal conductivity tests have revealed how the thermal behavior of the samples analyzed is essentially determined by the density and water content of the material: in fact, the thermal conductivity increases of two to three times the value of the not-tinted material. 相似文献

In the present study, we have investigated the occurrence of yeast flora on several agricultural products coming from crop-growing environments in Cameroon, to provide better knowledge of the biodiversity of yeast flora, and to thus define the impact of this biodiversity on food products. The yeast biodiversity was investigated using traditional culture-dependent methods, along with culture-independent methods. The culture-dependent approach was carried out using both direct and enrichment procedures, to detect the broadest possible presence of yeast species. A total of 151 strains belonging to 26 different yeast species were isolated and identified using restriction pattern analysis of the internal transcribed spacer region 5.8S-ITS and sequence analysis of D1/D2 domain of 26S rRNA gene. The enrichment isolation procedures carried out in high-sugar media allowed the recognition of fermentative species such as Saccharomyces cerevisiae and Torulaspora delbrueckii, which have previously not been detected using direct isolation methodology. The results of culture-independent method using DGGE patterns and sequencing of the DNA bands revealed a lower number of yeast species when compared with the culture-dependent methodology even if the identification of several yeast species not detected by traditional microbiological procedures such as Candida tropicalis and Hanseniaspora uvarum is allowed. Thus, these multiphasic approaches to study yeast biodiversity (culture-dependent and -independent methods) have allowed us to get a more complete picture of the microbial diversity in these natural environments. 相似文献

Three series of bithiophene azo dyes functionalized with thiazol-2-yl or benzothiazol-2-yl-diazene acceptor moieties were synthesized through azo coupling reaction using 2,2′-bithiophene, 5-alkoxy-2,2′-bithiophenes, 5-N,N-dialkylamino-2,2′-bithiophenes and thiazolyl- and benzothiazolyl diazonium salts as coupling components. The 5-alkoxy-2,2′-bithiophene precursors yielded the 5-thiazolylazo-5′-alkoxy-2,2′-bithiophenes, while the azo coupling reaction of 5-N,N-dialkylamino-2,2′-bithiophenes with the thiazolyl diazonium salt gave 4-thiazolyl-azo-5-N,N-dialkylamino-2,2′-bithiophenes. A different reactivity behavior was observed for 2,2-bithiophene with thiazolyl diazonium salts which gave rise to the expected azo dyes together with several arylation products. The redox behavior, thermal stability, and the first hyperpolarizability of the novel chromophores were evaluated through cyclic voltammetry, thermogravimetric analysis (TGA) and hyper-Rayleigh scattering (HRS) respectively. By varying the position of the thiazolyldiazene acceptor group on the bithiophene system, the electrochemical behavior as well as the optical (linear and nonlinear) properties of the donor-acceptor π-conjugated systems can readily be tuned. Push-pull 5-thiazolylazo-5′-alkoxy-2,2′-bithiophenes exhibit the most promising redox and the solvatochromic properties and second-order nonlinear optical response. The redox and the optical properties also show notable variations for the different heterocyclic spacers and were also sensitive to the electronic acceptor strength of the (benzo)thiazolyldiazene moieties. 相似文献

Anodic oxidation of molybdenum in weakly acidic, nearly neutral and weakly alkaline electrolytes was studied by voltammetric and electrochemical impedance spectroscopic measurements in a wide potential and pH range. Current vs. potential curves were found to exhibit two pseudo-Tafel regions suggesting two parallel pathways of the dissolution process. Electrochemical impedance spectra indicated the presence of at least two reaction intermediates. X-ray photoelectron spectroscopic (XPS) results pointed to the formation of an oxide containing Mo(IV), Mo(V) and Mo(VI), the exact ratio between different valence states depending on potential and pH of the solution. A physico-chemical model of the processes is proposed and a set of kinetic equations for the steady-state current vs. potential curve and the impedance response are derived. The model is found to reproduce quantitatively the current vs. potential curves and impedance spectra at a range of potentials and pH and to agree qualitatively with the XPS results. Subject to further improvement, the model could serve as a starting point for the optimization of the electrochemical fabrication of functional molybdenum oxide coatings. 相似文献

A fast and easy one-step linker-free approach for the synthesis of palladium nanoparticle/multiwall carbon nanotube (Pd-NP/MWCNT) hybrid materials is described using microwave irradiation for the effective decomposition of Pd2dba3 complex in the presence of MWCNTs. High loadings of Pd nanoparticles (up to 40 wt.%) having sizes between 3 and 5 nm are deposited on the surface of MWCNTs within a time of only 2 min. The Pd-NP/MWCNT materials serve as efficient catalysts in C–C coupling as well as in hydrogenation reactions, all characterized by high conversion rates using a small amount of catalysts, high turnover frequency values and good recyclability. 相似文献
