Inhibition of the biosynthesis of complex N‐glycans in the Golgi apparatus influences progress of tumor growth and metastasis. Golgi α‐mannosidase II (GMII) has become a therapeutic target for drugs with anticancer activities. One critical task for successful application of GMII drugs in medical treatments is to decrease their unwanted co‐inhibition of lysosomal α‐mannosidase (LMan), a weakness of all known potent GMII inhibitors. A series of novel N‐substituted polyhydroxypyrrolidines was synthesized and tested with modeled GH38 α‐mannosidases from Drosophila melanogaster (GMIIb and LManII). The most potent structures inhibited GMIIb (Ki=50–76 μm , as determined by enzyme assays) with a significant selectivity index of IC50(LManII)/IC50(GMIIb) >100. These compounds also showed inhibitory activities in in vitro assays with cancer cell lines (leukemia, IC50=92–200 μm ) and low cytotoxic activities in normal fibroblast cell lines (IC50>200 μm ). In addition, they did not show any significant inhibitory activity toward GH47 Aspergillus saitoiα1,2‐mannosidase. An appropriate stereo configuration of hydroxymethyl and benzyl functional groups on the pyrrolidine ring of the inhibitor may lead to an inhibitor with the required selectivity for the active site of a target α‐mannosidase.  相似文献

Mesenchymal stem cells (MSCs) should be expanded in vitro while maintaining their multilineage potential before differentiation to one mesenchymal lineage for application to regeneration therapy. The effect of fetal calf serum (FCS) on undesirable differentiation during subcultivations for the expansion was investigated. The expression level of the aggrecan gene, which is a marker of chondrogenic differentiation, gradually and markedly increased during the subcultivations of MSCs with the addition of 10% FCS and without additional cytokines. The percentage of cells positive for CD90 and CD166, which are markers of MSCs, decreased, and the percentage of large polygonal cells and the average cell adhesion area increased during the expansion. There was a marked difference in the increase in the aggrecan expression level between the two expansion cultures employing different FCS lots, although their proliferation rates were almost the same. The decrease in FCS concentration resulted in a higher percentage of CD90(+)CD166(+) cells, a lower percentage of large polygonal cells, and a lower level of aggrecan expression. Consequently, FCS components could stimulate MSC differentiation to chondrocytes and a lower concentration could decrease this differentiation.  相似文献

We assessed the effect of electron beam irradiation of packaged fresh blueberries at doses greater than 1.0 kGy on the quality attributes of the fruits. Irradiation experiments were conducted using a 10 MeV (18 kW) linear accelerator with single beam fixture. Fruits were stored at 5 °C and 70.4% RH for 14 days and tested at days 0, 3, 7 and 14 for physico-chemical, textural, microstructural, and sensory characteristics. Control samples consisted of non-irradiated fruits. Irradiation at doses higher than 1.1 kGy did affect (P<0.05) the texture of blueberries as the fruits became considerably softer and less acceptable throughout storage. Only irradiation at 3.2 kGy affected the color of blueberries by the end of storage. Irradiation slightly reduced the respiration rates of the blueberries by the end of storage. In terms of overall quality, texture and aroma, only fruits exposed to 3.2 kGy were found unacceptable by the sensory panelists. Irradiation at the dose levels used in this study did not affect the density, pH, water activity, moisture content, acidity and juiciness of blueberries. Electron beam irradiation of blueberries up to 1.6 kGy is a feasible decontamination treatment that maintains the overall fruit quality attributes.  相似文献

Bowman–Birk inhibitors (BBI) from soybean and related proteins are naturally occurring protease inhibitors with potential health‐promoting properties within the gastrointestinal tract. In this work, we have investigated the effects of soybean BBI proteins on HT29 colon adenocarcinoma cells, compared with non‐malignant colonic fibroblast CCD‐18Co cells. Two major soybean isoinhibitors, IBB1 and IBBD2, showing considerable amino acid sequence divergence within their inhibitory domains, were purified in order to examine their functional properties, including their individual effects on the proliferation of HT29 colon cancer cells. IBB1 inhibited both trypsin and chymotrypsin whereas IBBD2 inhibited trypsin only. Despite showing significant differences in their enzyme inhibitory properties, the median inhibitory concentration values determined for IBB1 and IBBD2 on HT29 cell growth were not significantly different (39.9±2.3 and 48.3±3.5 μM, respectively). The cell cycle distribution pattern of HT29 colon cancer cells was affected by BBI treatment in a dose‐dependent manner, with cells becoming blocked in the G0–G1 phase. Chemically inactive soybean BBI had a weak but non‐significant effect on the proliferation of HT29 cells. The anti‐proliferative properties of BBI isoinhibitors from soybean reveal that both trypsin‐ and chymotrypsin‐like proteases involved in carcinogenesis should be considered as potential targets of BBI‐like proteins.  相似文献

BACKGROUND: Despite the long history of sherry wine vinegar, new alternatives of consumption are being developed, with the aim of diversifying its market. Several new acetic‐based fruit juices have been developed by optimising the amount of sherry wine vinegar added to different fruit juices: apple, peach, orange and pineapple. Once the concentrations of wine vinegar were optimised by an expert panel, the aforementioned new acetic fruit juices were tasted by 86 consumers. Three different aspects were taken into account: habits of consumption of vinegar and fruit juices, gender and age. RESULTS: Based on the sensory analysis, 50 g kg?1 of wine vinegar was the optimal and preferred amount of wine vinegar added to the apple, orange and peach juices, whereas 10 g kg?1 was the favourite for the pineapple fruit. Based on the olfactory and gustatory impression, and ‘purchase intent’, the acetic beverages made from peach and pineapple juices were the most appreciated, followed by apple juice, while those obtained from orange juice were the least preferred by consumers. CONCLUSION: New opportunities for diversification of the oenological market could be possible as a result of the development of this type of new product which can be easily developed by any vinegar or fruit juice maker company. © 2012 Society of Chemical Industry  相似文献

Peat deposits can concentrate chalcophilic metals such as Zn and Cd by biogeochemical processes; as a result, there is a possibility that the solubility, mobility, and bioavailability of these metals could increase when such deposits are drained and cropped, initiating oxidation of organic matter and sulfides under aerobic conditions. We use spectroscopic, chemical, and bioassay approaches to characterize high Zn (88-15,800 mg kg(-1)), Cd (0.55-83.0 mg kg(-1)), and S (3.52-9.54 g kg(-1)) peat soils collected from locations in New York State and Ontario that overlie Silurian-age metal-enriched dolomite bedrock. Total and KNO3-extractable trace metals were determined by ICP emission spectrometry, and labile Cd and Zn were measured in the KNO3 extracts by anodic stripping voltammetry. A greenhouse bioassay with maize and canola was conducted to determine the bioavailability and toxicity of the soil Zn and Cd. The electronic oxidation states of sulfur in the peat soils were determined by X-ray absorption near edge spectroscopy (XANES) and Zn and S distribution in soil particles by energy-dispersive X-ray absorption (EDX) spectroscopy. Sulfur-XANES analyses show that a high percentage (35-45%) of the total soil S exists in the most reduced electronic oxidation states (such as sulfides and thiols), while <5% exists in the most oxidized forms (such as sulfate). EDX analyses indicate a microscopic elemental association between Zn and S in these soils. Despite the EDX evidence of close association between Zn and S in soil particles, conventional X-ray diffraction analyses of the bulk soils did not detect a mineral phase of sphalerite (ZnS) in any of the soils. The distribution coefficients (Kd) for Zn and Cd increased with soil pH and indicated stronger Cd retention than Zn in the peats. The results of the bioassaytests showed that most of the high-Zn soils were very phytotoxic, with plant shoot Zn levels exceeding 400 mg kg(-1). Conversely, Cd concentrations in the plant shoots were generally below 2 mg kg(-1), showing a tendency toward low Cd phytoavailability relative to Zn. The information gained from the spectroscopic analyses (S-XANES and EDX) was used to explain the macroscopic observations (Cd and Zn Kd values and phytoavailability data) in these peat soils; we conclude that sulfur biogeochemical cycling may play an important role in Zn and Cd retention in these organic soils.  相似文献

The free amino acid and nucleotide contents of dry-cured ham, shoulder and loin from two genetic lines selected from pigs according to the paternal allele (homozygous AA and heterozygous AG) of the insulin-like growth factor-II gene were studied by HPLC. Their influence on the flavor and taste characteristics was also studied. The increase of lean content caused by the IGF-II mutation could affect proteolysis during the ripening process and therefore the sensory characteristics. The lower intramuscular fat content in the AA ham batch had a positive effect on the free amino acid content. However, similar flavor traits between ham batches were found, but the AG loin batch showed greater value. The enhancing effect of the IMP on the overall flavor intensity was limited by the amino acid and the IMF contents in dry-cured ham and loin, while in dry-cured shoulder, the IMP could be the reason for the significant differences in after taste and cured flavor scores.  相似文献

Increasing world population worsens the serious problem of food security in developing countries. On the other hand in industrialized countries, where the problem of food security is of minor concern, health problems related to food refer to 2 main factors: food safety and environmental sustainability of food production. For these reasons, new ways must be found to increase yields while preserving food quality, natural habitats, and biodiversity. Insects could be of great interest as a possible solution due to their capability to satisfy 2 different requirements: (i) they are an important source of protein and other nutrients; (ii) their use as food has ecological advantages over conventional meat and, in the long run, economic benefits. However, little is known on the food safety side and this can be of critical importance to meet society's approval, especially if people are not accustomed to eating insects. This paper aims to collect information in order to evaluate how insects could be safely used as food and to discuss nutritional data to justify why insect food sources can no longer be neglected. Samples of the intestinal content and carcasses of wood pigeons (Columba palumbus) were evaluated for enterococci with antimicrobial activity. Enterococcus faecium comprised the largest enterococcal species with antagonistic activity, followed by Enterococcusfaecalis and Enterococcus columbae. PCR amplification of genes coding bacteriocins and determination of their nucleotide sequence, and the use of specific antipeptide bacteriocin antibodies and a noncompetitive indirect enzyme-linked immunosorbent assay, permitted characterization of enterococci coding that described bacteriocins and their expression. The efaAfm determinant was the only virulence gene detected in E. faecium, whereas E. faecalis showed a larger number of virulence determinants, and E. columbae did not carry any of the virulence genes examined. Although all E. faecalis isolates manifested a potent direct antimicrobial activity, no activity was detected in supernatants of producer cells. Purification of the antagonistic activity of E. columbae PLCH2 showed multiple chromatographic fragments after matrix-assisted laser desorption-ionization time-of-flight mass spectrometry analysis, suggesting the active peptide(s) had not yet purified to homogeneity. Bacteriocinogenic E. faecium and E. columbae isolates may be considered hygienic for production of enterocins and potentially safe due to their low incidence of potential virulence genes and susceptibility of most relevant clinical antibiotics. However, the presence among the enterococci of E. faecalis strains with a potent antagonistic activity and multiple virulence factors is an issue that must be considered further.  相似文献

Three organic cations with a natural origin (L-carnitine, L-cystine dimethyl ester, and thiamine) were introduced at different loadings in the interlayer of a low-charge montmorillonite, and the performance of the modified clays as adsorbents of the herbicide simazine was investigated using batch adsorption-desorption experiments. The organic cations were selected on the basis of their natural origin and the presence of diverse functional groups in their structures, which was expected to influence simazine adsorption. Elemental analysis and spectroscopy results demonstrated the presence of the organic cations in the modified montmorillonites and their entrance in the clay mineral interlayers. Batch adsorption results showed that modification with thiamine (K(f) = 96-138), cystine dimethyl ester (K(f) = 400-753), and especially carnitine (K(f) > 10 000) enhanced the adsorption of simazine by montmorillonite (K(f) = 28-47). It appeared that the specific interlayer microenvironment provided by the functional groups of each organic cation was an important factor controlling the adsorption efficiency of the modified clays. For carnitine and cystine dimethyl ester, the increase in simazine adsorption was considerably greater than that observed after montmorillonite modification with "classical" alkylammonium cations, such as phenyltrimethylammonium or hexadecyltrimethylammonium. This illustrated how modification of smectitic clay minerals with natural organic cations containing appropriate functional groups can be a useful strategy to improve the performance of organoclays for the removal of specific organic pollutants from the environment.  相似文献
