Recent outbreaks of foodborne illness have been linked to the consumption of contaminated sprouts. The spent irrigation water used to irrigate sprouts can carry many microorganisms, including pathogenic strains of Escherichia coli and Salmonella enterica. These pathogens are believed to originate from the seeds. The U.S. Food and Drug Administration recommends that sprout producers conduct microbiological testing of spent irrigation water from each production lot at least 48 h after seeds have germinated. Microbial analysis for the detection of Salmonella is labor-intensive and takes days to complete. A rapid and automated fiber-optic biosensor assay for the detection of Salmonella in sprout rinse water was developed in this study. Alfalfa seeds contaminated with various concentrations of Salmonella Typhimurium were sprouted. The spent irrigation water was assayed 67 h after alfalfa seed germination with the RAPTOR (Research International, Monroe, Wash.), an automated fiber optic-based detector. Salmonella Typhimurium could be positively identified in spent irrigation water when seeds were contaminated with 50 CFU/g. Viable Salmonella Typhimurium cells were also recovered from the waveguides after the assay. This biosensor assay system has the potential to be directly connected to water lines within the sprout-processing facility and to operate automatically, requiring manual labor only for preventative maintenance. Therefore, the presence of Salmonella Typhimurium in spent irrigation water could be continuously and rapidly detected 3 to 5 days before the completion of the sprouting process.  相似文献

Bologna type sausages were prepared from oxidatively stressed pork (UV-irradiation, 48 h, 5 °C) using a traditional recipe (control) or the same recipe but added green tea extract (500 ppm total phenolic compounds) or rosemary extract (400 ppm total phenolic compounds). Green tea and rosemary extracts protected against formation of TBARS and protein carbonyls. On the contrary, increased thiol loss and a distinct loss of myosin heavy chain and actin due to polymerization by reducible bonds as determined by SDS-page were found by addition of green tea extract. The enhanced protein polymerization was ascribed to the reaction between quinone compounds from the plant extracts and protein thiol groups to yield phenol-mediated protein polymerization. Analysis by ESR spectroscopy revealed increased radical intensities in sausages added plant extracts, which was ascribed to originate from protein-bound phenoxyl radicals, which may protect against other oxidatively induced protein modifications.  相似文献

This study used a cross-national framework to examine country, region, and gender differences in emotional availability (EA), a prominent index of mutual socioemotional adaptation in the parent-child dyad. Altogether 220 Argentine, Italian, and U.S. mothers and their daughters and sons from both rural and metropolitan areas took part in home observations when the children were 20 months old. In terms of country, Italian mothers were more sensitive and optimally structuring, and Italian children were more responsive and involving, than Argentine and U.S. dyads. In terms of region, rural mothers were more intrusive than metropolitan mothers, and boys from metropolitan areas were more responsive than boys from rural areas. In terms of gender, mothers of girls were more sensitive and optimally structuring than mothers of boys, and daughters were more responsive and involving than sons. Understanding how country, region, and gender influence EA exposes forces that shape child development, parent-infant interaction, and family systems. Peroxy-organic acids are formed by the chemical reaction between organic acids and hydrogen peroxide. The peroxy-acid process was applied to two Superfund site soils provided by the U.S. Environmental Protection Agency (EPA). Initial small-scale experiments applied ratios of 3:5:7 (v/v/v) or 3:3:9 (v/v/v) hydrogen peroxide:acetic acid:deionized (DI) water solution to 5g of Superfund site soil. The experiment using 3:5:7 (v/v/v) ratio resulted in an almost complete degradation of the 14 EPA regulated polycyclic aromatic hydrocarbons (PAHs) in Bedford LT soil during a 24-h reaction period, while the 3:3:9 (v/v/v) ratio resulted in no applicable degradation in Bedford LT lot 10 soil over the same reaction period. Specific Superfund site soil characteristics (e.g., pH, total organic carbon content and particle size distribution) were found to play an important role in the availability of the PAHs and the efficiency of the transformation during the peroxy-acid process. A scaled-up experiment followed treating 150g of Bedford LT lot 10 soil with and without mixing. The scaled-up processes applied a 3:3:9 (v/v/v) solution resulting in significant decrease in PAH contamination. These findings demonstrate the peroxy-acid process as a viable option for the treatment of PAH contaminated soils. Further work is necessary in order to elucidate the mechanisms of this process.  相似文献

A water tunnel in Candida antarctica lipase B that provides the active site with substrate water is hypothesized. A small, focused library created in order to prevent water from entering the active site through the tunnel was screened for increased transacylation over hydrolysis activity. A single mutant, S47L, in which the inner part of the tunnel was blocked, catalysed the transacylation of vinyl butyrate to 20 mM butanol 14 times faster than hydrolysis. The single mutant Q46A, which has a more open outer end of the tunnel, showed an increased hydrolysis rate and a decreased hydrolysis to transacylation ratio compared to the wild‐type lipase. Mutants with a blocked tunnel could be very useful in applications in which hydrolysis is unwanted, such as the acylation of highly hydrophilic compounds in the presence of water.  相似文献

This article focuses on the manufacture and usage of selected sheet metal objects from helmets, a vessel, and two shields from the European Bronze Age and Early Iron Age, which date from the thirteenth to seventh centuries BC. Manufacturing traces on the surface, as well as metallographic investigations and the analyses of the alloy composition with scanning electron microscope–energy-dispersive x-ray spectroscopy, provided an insight into the manufacturing techniques and the production of valuable, high-status objects, and highlighted the potential changes in manufacturing techniques and alloys used during different time periods.  相似文献

Bimodal molecular weight distributions of alternating copolymers of the type (Me2Si?R2Si?SiMe2) n , where R=hexyl and butyl, were obtained by polymerization of BrSiMe2?R2Si?Me2SiBr, using the Wurtz coupling method. Analysis of the samples by pyrolysis GC MS and solution29Si NMR indicated that some randomization occurred, due to cleavage of the original Si?Si bonds in the monomer. The extent of randomization was significantly greater in the high molecular weight fractions. Based on the nature of the rings from the py MGC/MS traces and the number on nonads found in the29Si spectra, two types of randomization processes have been proposed, involving backbiting followed by (1) ring expulsion or (2) redistribution.  相似文献

Laboratory experiments have been conducted to elucidate the degradation of alpha-methylnaphthalene in lake sediments using an advanced oxidation process (AOP) employing peroxy-acids as oxidizing agents. Abiotic degradation of alpha-methylnaphthalene was observed in sediment samples exposed to a 1:1:1 v/v/v mixture of water/organic acid/hydrogen peroxide solution. Sediment samples were collected from Lake Macatawa (Holland, MI) and ranged in total organic carbon content from 2.1% (sandy sediment sample III) to 12.8% (silty-clay sediment sample I) and surface area, which ranged from 3.2m(2)/g (sandy sediment sample III) to 22.0m(2)/g (silty-clay sediment sample I). Analytical measurements performed by gas chromatography revealed varied rates of degradation, depending on the type of acid and the characteristics of the sediment sample. Within 24h, alpha-methylnaphthalene was reduced to 70% and 100% of its original concentration when propionic acid and acetic acid were used as the organic acids, respectively. The formation of products was explored using a gas chromatograph equipped with a mass spectrometer and indicated mainly the formation of lower molecular weight compounds, such as alkyl chains. This AOP method of PAH degradation in sediment/liquid slurry was fast acting and products were most likely biodegradable.  相似文献

A modified version of the fractionator was used to estimate the total number of polystyrene microspheres retained in the airways of hamster lungs at two different time points after inhalation. A systematic three-stage subsampling procedure with known sampling fractions was adopted. First, each lung was cut into slices, from which primary disectors were sampled systematically with a known sampling fraction. From each primary disector, smaller sub-disectors were subsampled, and the corresponding sampling fraction was estimated by point counting. Finally, a few particles were counted at the microscopic level in the sub-disectors, and the final estimate of total particle number (which is unbiased irrespective of any tissue deformations) was easily computed as a product of the counted number times the reciprocal of the successive sampling fractions. The error variance of each estimate was assessed from the data using a new estimator. An average of 6% of the deposited particles were retained on the epithelial surface of the intrapulmonary conducting airways shortly after the inhalation, from which at least one-third was already phagocytosed by macrophages. After 24 h, an average of 87% of the particles retained shortly after the inhalation had been cleared. The proportion of particles ingested by macrophages had increased to at least 87%, in three out of four animals studied.  相似文献

Three approaches that allow the tailoring of long period gratings based refractometric sensors for concentration measurement in fuel blends are employed to assess the fuel quality in biodiesel and biodiesel-petrodiesel blend. To allow the analysis of fuel samples with refractive index higher than fiber cladding one, the samples refractive indices were changed by thermo-optic effect and by dilution in a standard substance with low refractive index. The obtained results show the sensor can detect oil concentration in biodiesel samples with resolution as better as 0.07% and biodiesel concentration in biodiesel-petrodiesel samples with average resolution of 0.09%.  相似文献
