Infantile hemangioma (IH) is the most common infantile tumor, affecting 5–10% of newborns. Propranolol, a nonselective β-adrenergic receptor (ADRB) antagonist, is currently the first-line treatment for severe IH; however, both its mechanism of action and its main cellular target remain poorly understood. Since betablockers can antagonize the effect of natural ADRB agonists, we postulated that the catecholamine produced in situ in IH may have a role in the propranolol response. By quantifying catecholamines in the IH tissues, we found a higher amount of noradrenaline (NA) in untreated proliferative IHs than in involuted IHs or propranolol-treated IHs. We further found that the first three enzymes of the catecholamine biosynthesis pathway are expressed by IH cells and that their levels are reduced in propranolol-treated tumors. To study the role of NA in the pathophysiology of IH and its response to propranolol, we performed an in vitro angiogenesis assay in which IH-derived endothelial cells, pericytes and/or telocytes were incorporated. The results showed that the total tube formation is sensitive to propranolol only when exogenous NA is added in the three-cell model. We conclude that the IH’s sensitivity to propranolol depends on crosstalk between the endothelial cells, pericytes and telocytes in the context of a high local amount of local NA.  相似文献

BACKGROUND: Tedisamil is a new bradycardic agent proven to exert anti-ischemic and antiarrhythmic effects by blockade of the different cardiac and vascular K+ currents. HYPOTHESIS: It was the aim of the present study to compare the favorable anti-ischemic effects of tedisamil, with two long established representatives in the treatment of coronary artery disease (CAD), namely, the beta1 blocker esmolol and the Ca2 antagonist gallopamil. METHODS: The hemodynamic and neurohumoral effects of the new potassium channel blocker tedisamil, an agent with negative chronotropic and class III antiarrhythmic properties, were compared with the ultra-short-acting beta1-selective adrenoceptor blocker esmolol and the calcium antagonist gallopamil. A total of 22 patients with angiographically proven CAD and reproducible ST-segment depression in the exercise electrocardiogram was included in two studies with an almost identical design and inclusion criteria. The investigation was carried out using right heart catheterization and bicycle ergometry. A subgroup of 8 patients receiving 0.3 mg/kg body weight tedisamil intravenously (i.v.) in an open dose-finding study was compared with a group of 14 patients who had received esmolol (i.v. bolus of 500 micrograms/kg, maintenance dose 200 micrograms/kg/min) and gallopamil (initial dose 0.025 mg/kg, maintenance dose 0.0005 mg/kg/h) in a second intraindividual comparison. RESULTS: Tedisamil and esmolol reduced heart rate at rest by 13% (p < 0.001), and 6% (p < 0.05), and at maximum working levels by 8% (p < 0.01) and 9% (p < 0.05), respectively. Gallopamil increased heart rate at rest by 7% (p < 0.05), with only slight changes occurring during exercise. Corresponding findings for each drug were observed for cardiac output both at rest and during exercise [tedisamil: at rest -10% (NS), max. exercise -8%; esmolol: at rest -14% (NS), max. exercise -18% (NS); gallopamil: no significant changes]. Compared with tedisamil, stroke volume was reduced by esmolol [at rest and max. workload: -9% (NS)] and gallopamil [rest: -6% (NS), max. exercise: -2% (NS)]. Of the indirect parameters of ventricular function, that is, mean capillary wedge pressure (PCWPm) and right ventricular ejection fraction, only PCWPm demonstrated significant differences between tedisamil and gallopamil (+18% and -6% at rest, +17% and -21% during exercise, respectively; p < 0.001). Compared with gallopamil, both tedisamil and esmolol were superior in their effects on rate-pressure product, myocardial oxygen consumption, and ST-segment depression, whereas plasma lactate concentration was more reduced by tedisamil and gallopamil. Tedisamil led to a fall in norepinephrine levels in particular. CONCLUSION: Tedisamil and esmolol showed almost equipotent anti-ischemic effects at the doses administered. Tedisamil acts mainly by reductions in heart rate, and esmolol, though to a lesser degree, also by reductions in systolic blood pressure. The mechanism of gallopamil is to reduce afterload and to improve coronary perfusion. At the doses applied, however, it has lower antianginal potency compared with tedisamil and esmolol.  相似文献

BACKGROUND: The CarboMedics valve is a relatively new, low-profile, bileaflet, mechanical prosthesis. The results of a prospective follow-up study after valve replacement with this prosthesis in a university hospital are presented. METHODS: We implanted 640 CarboMedics prostheses in 583 patients in the aortic (n = 359), mitral (n = 167), or aortic and mitral positions (double valve replacement; n = 57). Patient ages ranged from 11 to 81 years (mean age, 58 +/- 12.3 years). RESULTS: Overall hospital mortality was 9.0%; however, when high-risk urgent cases were removed from the calculation, the operative mortality fell to 4.5%. Follow-up was 98% complete, comprising 2,027 patient-years for a mean follow-up of 44 months (range, 6 to 72 months). Actuarial freedom from complications (linearized rates in parentheses) was as follows: late mortality, 85% +/- 2.0% (2.3%/patient-year); thromboembolism, 92% +/- 1.1% (1.6%/patient-year); anticoagulation-related hemorrhage, 87% +/- 1.2% (2.8%/patient-year); prosthetic valve endocarditis, 98% +/- 0.5% (0.1%/patient-year); and overall valve-related morbidity and mortality, 76% +/- 2.1% (4.3%/ patient-year). CONCLUSIONS: The CarboMedics valve shows a low rate of valve-related complications comparable with other new mechanical heart valve prostheses.  相似文献

The effects of heat treatment, involving solutionizing at temperature of 370 °C for a relatively short period of time (3 or 5 h), followed by quenching in water, on tribological behavior of ZA-27 alloys were examined.Dry sliding wear tests were conducted on as-cast and heat-treated ZA-27 samples using block-on-disk machine over a wide range of applied loads. To determine the wear mechanisms, the worn surfaces of the samples were examined by scanning electron microscopy (SEM). The tribological results were related to the microstructure and mechanical properties.The heat treatment resulted in reduction in the hardness and tensile strength but increase in elongation. The heat-treated alloy samples attained improved tribological behavior over the as-cast ones, both from the aspects of friction and wear. The improved tribological behavior of the heat-treated alloys, in spite of reduced hardness, could be the result of breaking the dendrite structure, when the fraction of interdendrite regions was considerably decreased and a very fine α and η mixture was formed at the same time. The wear response of the samples has been corroborated through characteristics of worn surfaces and dominant wear mechanisms.  相似文献

The aim of this work was to develop and validate static headspace gas chromatography with flame ionization detection method for determination of five aldehydes selected as markers for lipid oxidation, present in freshly baked and stored crackers with high fat content. Chromatographic conditions defined in this method allow the separation of propanal, pentanal, hexanal, heptanal and octanal within 15 min. Relative standard deviations (RSD) of the retention times for repeatability and intermediate precision for all compounds were less than 1.75 and 0.22 %, respectively. For the obtained peak areas, RSD for repeatability and intermediate precision were less than 3.86 and 4.32 %, respectively. Recovery was within the range of 60–99 %. Each analyte showed an excellent linear behaviour over the set concentration range, with correlation coefficient (R 2 ) values above 0.99. Limits of quantification for propanal, pentanal, hexanal, heptanal and octanal were 0.10, 0.05, 0.08, 0.02 and 0.02 μg, respectively. The proposed method accomplishes the requirements for the method selectivity, precision, sensitivity, accuracy and linearity needed for determination of aldehydes in bakery products. Quantification of aldehydes was achieved for both freshly baked and stored crackers over a long period of time.  相似文献

The content of tocopherols, B vitamins, as well as free soluble, soluble conjugated and insoluble bound phenolic compounds was determined in untreated, steeped and sprouted wheat grains. Antioxidant capacity of whole wheat grains and their phenolic fractions was also evaluated. Sprouting significantly increased the levels of tocopherols, niacin, riboflavin, as well as free and bound phenolic compounds improving nutritional value and antioxidant capacity of wheat grains/flour. After sprouting for 5 days, the content of total phenolics, flavonoids and ferulic acid calculated as the sum of its fractions was increased by 9.9, 30.7 and 21.6%, respectively. The content of α‐, β+γ‐ and δ‐tocopherols was increased for 3.59‐fold, 2.33‐fold and 2.61‐fold respectively, while the content of niacin, as predominant B vitamin, was increased for 1.19‐fold after sprouting. The total antioxidant capacity of untreated, steeped and sprouted whole wheat grains was 19.44, 20.37 and 22.70 mmol Trolox Eq/kg, respectively. Sprouted wheat, as a rich source of bioavailable phytochemicals, should be used to improve the nutritional quality of food.  相似文献

Natural abundances of stable isotopes in lotic food webs yield valuable information about sources of organic matter for consumers and trophic structure. However, interpretation of isotopic information can be challenging in the face of variability in organisms at the base of food webs. Unionid and dreissenid mussels, commonly used as baseline organisms in lakes, are uncommon in many river settings and can have variable diets, thus making them unsuitable as a universal baseline for many river food web studies and often forcing reliance on more common benthic insects for this purpose. Turnover rates of body carbon and nitrogen in insects are relatively rapid (1 to 50 days half‐life). These rapid turnover rates in primary consumers can result in considerable temporal variability in δ13C that rivals that of algae (>10‰ range within a site). This suggests that using primary consumers as a surrogate baseline for algae may not circumvent the problem of temporal variability and the resultant mismatch of sources with longer‐lived, slow‐growing secondary and tertiary consumers. There are several strategies for reducing the influence of these confounding factors when bivalves with a known diet are not present. These include sampling over large spatial scales and correlating δ13C of consumers with the source of interest (e.g. benthic algae), sampling baseline organisms multiple times in the weeks preceding sampling of larger consumers (particularly in response to large changes in discharge) and using algal‐detrital separation methods and multiple tracers as much as possible. Incorporating some of these recommendations and further exploring variability at the base of the food web will potentially provide greater insights into consumer–resource coupling in running waters and more robust conclusions about food web structure and energy flow in these dynamic systems. Measurements of the liquid–solid mass transfer coefficient kS were performed in a three-phase multiple impeller and stationary catalytic basket reactor, called a Robinson–Mahoney reactor. Local coefficients were determined using naphtol particle dissolution in water and n-heptane with or without gas flow. Experiments have shown that local coefficients kS loc depend on the particle position in the basket, agitation speed, and liquid properties. A correlation linking kS to liquid velocity was established in a reactor simulating hydrodynamics in a piece of catalytic basket.
