Non-enzymatic glycation is an unavoidable reaction that occurs across biological taxa. The final products of this irreversible reaction are called advanced glycation end-products (AGEs). The endogenously formed AGEs are known to be bioactive and detrimental to human health. Additionally, exogenous food-derived AGEs are debated to contribute to the development of aging and various diseases. Receptor for AGEs (RAGE) is widely known to elicit biological reactions. The binding of RAGE to other ligands (e.g., high mobility group box 1, S100 proteins, lipopolysaccharides, and amyloid-β) can result in pathological processes via the activation of intracellular RAGE signaling pathways, including inflammation, diabetes, aging, cancer growth, and metastasis. RAGE is now recognized as a pattern-recognition receptor. All mammals have RAGE homologs; however, other vertebrates, such as birds, amphibians, fish, and reptiles, do not have RAGE at the genomic level. This evidence from an evolutionary perspective allows us to understand why mammals require RAGE. In this review, we provide an overview of the scientific knowledge about the role of RAGE in physiological and pathological processes. In particular, we focus on (1) RAGE biology, (2) the role of RAGE in physiological and pathophysiological processes, (3) RAGE isoforms, including full-length membrane-bound RAGE (mRAGE), and the soluble forms of RAGE (sRAGE), which comprise endogenous secretory RAGE (esRAGE) and an ectodomain-shed form of RAGE, and (4) oxytocin transporters in the brain and intestine, which are important for maternal bonding and social behaviors.  相似文献

Trastuzumab-emtansine (T-DM1) is a therapeutic agent molecularly targeting human epidermal growth factor receptor 2 (HER2)-positive metastatic breast cancer (MBC), and it is especially effective for MBC with resistance to trastuzumab. Although several reports have described T-DM1 resistance, few have examined the mechanism underlying T-DM1 resistance after the development of acquired resistance to trastuzumab. We previously reported that YES1, a member of the Src family, plays an important role in acquired resistance to trastuzumab in HER2-amplified breast cancer cells. We newly established a trastuzumab/T-DM1-dual-resistant cell line and analyzed the resistance mechanisms in this cell line. At first, the T-DM1 effectively inhibited the YES1-amplified trastuzumab-resistant cell line, but resistance to T-DM1 gradually developed. YES1 amplification was further enhanced after acquired resistance to T-DM1 became apparent, and the knockdown of the YES1 or the administration of the Src inhibitor dasatinib restored sensitivity to T-DM1. Our results indicate that YES1 is also strongly associated with T-DM1 resistance after the development of acquired resistance to trastuzumab, and the continuous inhibition of YES1 is important for overcoming resistance to T-DM1.  相似文献

In this paper two watermarking algorithms for image content authentication with localization and recovery capability of the tampered regions are proposed. In both algorithms, a halftone version of the original gray-scale image is used as an approximated version of the host image (image digest) which is then embedded as a watermark sequence into given transform domains of the host image. In the first algorithm, the Integer Wavelet Transform (IWT) is used for watermark embedding which is denominated WIA-IWT (Watermarking-based Image Authentication using IWT), while in the second one, the Discrete Cosine Transform (DCT) domain is used for this purpose, we call this algorithm WIA-DCT (Watermarking-based Image Authentication using DCT). In the authentication stage the tampered regions are detected using the Structural Similarity index (SSIM) criterion, which are then recovered using the extracted halftone image. In the recovery stage, a Multilayer Perceptron (MLP) neural network is used to carry out an inverse halftoning process to improve the recovered image quality. The experimental results demonstrate the robustness of both algorithms against content preserved modifications, such as JPEG compression, as well as an effective authentication and recovery capability. Also the proposed algorithms are compared with some previously proposed content authentication algorithms with recovery capability to show the better performance of the proposed algorithms.  相似文献

ZnO thin films were formed in a solution with Zn(NO3)2 and Dimethylaminoborane at 70 °C. The effects of the aluminum ion concentration in the aqueous solution on the surface morphology, crystallographic structure and growth rate were investigated. By adding 10? 4 mol/l of Al, the growth rate was enhanced from 0.13 to 0.35 μm/h. In addition, the surface morphology became flat when the appropriate amount of Al(NO3)3, i.e., 10? 4 mol/l, was added. The Ra and Rz decreased from 20–32 nm and 115–145 nm to 1–11 nm and 7–60 nm, respectively.  相似文献

Despite the importance of multicultural competence in clinical practice and training, there is a surprising dearth of innovative training models from fieldwork sites that would be replicable in other practicum settings. The authors illustrate a multicultural competence training model from a community mental health center that highlights the Guidelines on Multicultural Education, Training, Research, Practice, and Organization Change for Psychologists (American Psychological Association, 2003). The model focuses on providing multicultural training through two separate but linked training teams for the dual purposes of internal reflection and reflective practice. The authors conclude with a discussion of the implications of this training model for practice, research, and organizational change. Inactivation rates of the biofilms of P. fluorescence and P. aeruginosa established on a small slide glass in ozone water (0.9–3.2 mg/L, 1–20 min) were determined in a batch or flow-through system. The effects of ozone water on the biofilm matrices were defined clearly in situ by confocal laser scanning microscopy. These results indicate that ozone is an effective biocide against biofilms and it can remove exopolysaccharides in the biofilm matrices. However, the effective concentration of ozone for disinfection of biofilms varied with the biofilms formed, mainly due to reactions of ozone with constituents of the biofilms.  相似文献

This note considers position and attitude control of large space structures composed of a number of subsystems (substructures) interconnected by flexible links. A decentralized control law of dynamic displacement feedback compatible with subsystems is applied under the assumption that sensors and actuators are collocated. It has been known that the overall closed-loop system is robustly stable against uncertainties in mass, damping, and stiffness, if rigid modes of each subsystem are controllable and observable. The objective of this note is to derive conditions for the overall system to be stable even when some local controllers fail. Since the successful completion of the Human Genome Project, increasing concern is being directed toward the polymorphic aspect of the genome and its clinical relevance. A form of single-strand DNA-conformation polymorphism analysis (SSCP) employing nondenaturing slab-gel electrophoresis (SGE) is applicable to the genetic diagnosis of bladder cancer from urine samples. To bring this technique into routine clinical practice, the use of capillary electrophoresis (CE) is naturally favorable in terms of speed and automation. However, the resolving power of SSCP, a prerequisite basis for reliability required in diagnostics, remains as a challenge for CE systems. We thus focused on this topic and conducted studies on CE instruments equipped with a single capillary or an array of multiple capillaries, using the resolution (Rs) as a quantitative scale for the resolving power. Polymer concentration and buffer are shown to be the decisive parameters. High Rs values of >2.5 are achieved for representative SNPs markers under the optimized conditions, without sacrificing such intrinsic advantages of CE over SGE as the 10-fold quicker migration time and operation that is reproducible, continuous, and automatic. The strategies presented broaden the limits of CE in both the current and related applications.  相似文献

The DNA chip that immobilizes DNA oligonucleotides on a solid plate surface is used for many diagnostic applications. For maximizing the detection sensitivity and accuracy, it is important to control the DNA density on a chip surface and establish a convenient method for optimizing the density. Here, the binding of DNA mismatch-binding protein MutS to the DNA substrate on the chip was investigated, which can be applied for high-throughput single-nucleotide polymorphism analysis in a genome. We prepared the DNA chips where the DNA substrate density was changed simply by using a mixed DNA solution. The binding of MutS was significantly influenced by the amount of DNA substrate on the chip as a consequence of steric crowding, and the moderate density that gave the distance between the DNA substrates greater than the size of the protein was appropriate to obtain accurate kinetic parameters. The substrate density-controlled DNA chip prepared using the mixed DNA solution has distinctive advantages for maximizing the detection capability and kinetic analysis of the binding of MutS and probably also other large proteins.  相似文献
