Miltefosine (hexadecylphosphocholine, HePC) is the first effective oral agent for the treatment of visceral leishmaniasis.
The characteristics of HePC incorporation into the human intestinal epithelial cell line Caco-2 were investigated in order
to understand its oral absorption mechanism. The results provide evidence for the involvement of a carrier-mediated mechanism,
since the association of HePC at the apical pole of Caco-2 cells was (1) saturable as a function of time with a rapid initial
incorporation over 5 min followed by a more gradual increase; (2) saturable as a function of concentration over the range
studied (2–200 μM) with a saturable component which followed Michaelis–Menten kinetics (apparent K
m 15.7 μmol/L, V
max 39.2 nmol/mg protein/h) and a nonspecific diffusion component; (3) partially inhibited by low temperature and ATP depletion,
indicating the temperature and energy-dependence of the uptake process. Moreover, we demonstrated, by an albumin back-extraction
method, that HePC is internalized via translocation from the outer to the inner leaflet of the plasma membrane and that HePC
may preferentially diffuse through intact raft microdomains. In conclusion, our results suggest that incorporation of HePC
at the apical membrane of Caco-2 cells may occur through a passive diffusion followed by a translocation in the inner membrane

The acoustic emission technique was applied to standard tests devoted to evaluate sulfide stress cracking susceptibility of steels for oil and gas industry. The mapping of the density of AE signals vs. their location on the specimen gauge length as a function of time allowed early detection of cracking, and gave meaningful information on incubation times and propagation rates. Sulfide stress cracking initiation was correlated with the presence of critical surface defects. A mechanism involving plastic strain and/or metal dissolution was proposed to account for crack propagation. 相似文献

A WebLab is an experiment operated remotely via Internet. Besides the strictly technical aspects of such an experiment, which may contribute to the learning of Chemical Engineering fundamentals, there is also important feedback when teams of students of two different countries are working together: the WebLab becomes an intercultural experience, enhancing the communication skills of the students. A WebLab between Universidade Federal de São Carlos (DEQ/UFSCar) and the Ecole Nationale Supérieure d’Ingénieurs en Arts Chimiques et Technologiques (ENSIACET) is presented in this work. A mass transfer experiment in a bench scale reactor (stirred and aerated) had to be studied by mixed teams, thus emulating challenges that will be common in future working environments. In order to perform the experiment, students in Brazil and in France were put into groups. The students had to make decisions about the procedure for executing the experiments. All the students were able to control the equipment, no matter where they were physically. Students communicated using video conference software. The students’ and teachers’ opinions of this experience were very positive. This methodology is an important contribution to the education of engineers in a world integrated by modern communication technologies. 相似文献

Recent advances in the development of high gauge factor thin films for strain gauges prompt the research on advanced substrate materials. A glass ceramic composite has been developed in consideration of a high coefficient of thermal expansion (9.4 ppm/K) and a low modulus of elasticity (82 GPa) for the application as support material for thin‐film sensors. In the first part, constantan foil strain gauges were fabricated from this material by tape casting, pressure‐assisted sintering, and subsequent lamination of the metal foil on the planar ceramic substrates. The accuracy of the assembled load cells corresponds to accuracy class C6. That qualifies the load cells for the use in automatic packaging units and confirms the applicability of the low‐temperature co‐fired ceramic (LTCC) substrates for fabrication of accurate strain gauges. In the second part, to facilitate the deposition of thin‐film sensor structures to the LTCC substrates, pressure‐assisted sintering step is modified using smooth setters instead of release tapes, which resulted in fabrication of substrates with low average surface roughness of 50 nm. Titanium thin films deposited on these substrates as test coatings exhibited low surface resistances of 850 Ω comparable to thin films on commercial alumina thin‐film substrates with 920 Ω. The presented material design and advances in manufacturing technology are important to promote the development of high‐performance thin‐film strain gauges. 相似文献

Three sympatric Reticulitermes species have been identified in Georgia, USA, based on morphological characters from alates and soldiers: R. flavipes, R. virginicus, and R. hageni, but species identification at individual collection sites is often difficult because alate production is seasonal and soldiers comprise 1–3% of the colony. We therefore set up an experiment to determine if chemical phenotypes and mtDNA haplotypes can be used together to separate species of subterranean termites. Subterranean termites of the order Reticulitermes (Isoptera: Rhinotermitidae) were collected from 20 inspection ports across four soil provinces in Georgia. Each collection was identified to species using dichotomous keys. Two collections, HH11 and BH25, however, could not be unequivocally keyed to species and were classified as unknown. The mitochondrial cytochrome oxidase II (COII) gene was sequenced from individual members of each collection and the variation in cuticular hydrocarbon phenotypes from these same collections was characterized. The cuticular hydrocarbon and mtDNA phylogenetic analyses show agreement with both unknown collections falling out in a separate clade. Specimens from HH11 nad BH25 are different morphologically, chemically, and genetically from the three known sympatric species in Georgia. Our results suggest that these two collections may represent at least one new taxon in Reticulitermes. Furthermore, the association of cuticular hydrocarbon phenotypes and mtDNA haplotypes demonstrates that, when combined with morphological characters, they are useful in separating known species, determining new species, and understanding termite evolution. 相似文献

This paper describes the role of Sb and Nb, components of Sn/V/Nb/Sb mixed oxides catalysts for the gas-phase ammoxidation of propane to acrylonitrile. In samples without Nb and with atomic ratios Sn/V/Sb 1/0.2/x (x = 0 to 3), Sb in the form of amorphous oxide is necessary in order to obtain an active and selective catalyst. However, during reaction the dispersed Sb oxide segregates to α-Sb2O4, and the yield to acrylonitrile decreases considerably. The addition of Nb gives rise to the formation of Nb-containing SbOx and non-stoichiometric rutile-type V/Nb/Sb mixed oxides. The presence of these compounds enhances the catalytic activity and the selectivity to acrylonitrile. Moreover, the catalyst shows a stable catalytic performance, with no segregation of α-Sb2O4. 相似文献
