To better detect and identify beeswax in ancient organic residues from archaeological remains, we developed a new analytical methodology consisting of the analysis of (i) the trimethylsilylated organic extract by GC/MS and (ii) the crude extract by ESI-MS. Selective scanning modes, such as SIM or MRM, permit separate quantification of each chemical family (fatty acids, monoesters, monohydroxyesters, and diesters) and allow an improvement in sensitivity and selectivity, allowing the crude extract to be treated without further purification. GC/MS (SIM) was revealed to be a powerful method for the detection of components, with a detection limit down to a total lipid extract in the range of approximately 50 ng in a complex matix, such as archaeological degraded material, whereas ESI-MS/MS is instead used for the detection of nonvolatile biomarkers. Identification by GC/MS (SIM) and ESI-MS/ MS (MRM) of more than 50 biomarkers of beeswax in an Etruscan cup at the parts-per-million level provides the first evidence for the use of this material by the Etruscans as fuel or as a waterproof coating for ceramics.  相似文献

For development of the French Laser Integration Line and the Laser Megajoule, we describe the design and the control of a first 383 mm x 398 mm continuous phase-plate prototype. Extensively used in laser fusion facilities for beam smoothing, this optical component was manufactured by deep etching onto a fused-silica substrate, which led to a phase plate engraved directly onto fused silica, with which good optical performance could be achieved. We demonstrate good agreement between the desired simulated component and the manufactured component in terms of focal spot shape. This demonstration was performed by both interferometric and photometric measurements.  相似文献

Accumulating evidence suggests that anxiety sensitivity (fear of arousal-related sensations) plays an important role in many clinical conditions, particularly anxiety disorders. Research has increasingly focused on how the basic dimensions of anxiety sensitivity are related to various forms of psychopathology. Such work has been hampered because the original measure--the Anxiety Sensitivity Index (ASI)--was not designed to be multidimensional. Subsequently developed multidimensional measures have unstable factor structures or measure only a subset of the most widely replicated factors. Therefore, the authors developed, via factor analysis of responses from U.S. and Canadian nonclinical participants (n = 2,361), an 18-item measure, the ASI-3, which assesses the 3 factors best replicated in previous research: Physical, Cognitive, and Social Concerns. Factorial validity of the ASI-3 was supported by confirmatory factor analyses of 6 replication samples, including nonclinical samples from the United States and Canada, France, Mexico, the Netherlands, and Spain (n = 4,494) and a clinical sample from the United States and Canada (n = 390). The ASI-3 displayed generally good performance on other indices of reliability and validity, along with evidence of improved psychometric properties over the original ASI. The illumination of overhead lines by a nuclear electromagnetic pulse (NEMP) in presence of a perfectly conducting ground gives rise to a scattering problem leading to phenomenons both studied here in time and frequency domains. This problem is resolved with the help of an integral equation established in space-time domain and making possible (without approximation) the determination of the time dependance of the current induced on each point of the wire structure. Moreover, the passage in frequency domain allows to express a transfer function defined in one point of the excited line, pointing out its spectral characteristics. Results clearly show the insufficiency of the classic transmission line theory concealing some phenomenons which are described here and for which an interpretation is proposed.  相似文献

Among oil compounds, fatty acids, tocopherols and xanthophylls (lutein and zeaxanthin) are of special interest due to their nutritional properties. The identification and quantification of these compounds in pecan nuts (Carya illinoinensis) could therefore be very useful to produce functional foods rich in compounds of this type. This paper reports studies on their accumulation and the effect of ripening on the content of these high value-added compounds. The total lipid content increased during the ripening. Saturated and polyunsaturated fatty acids decreased significantly, whereas, monounsaturated fatty acids increased during the ripening of pecan nut fruit. Maximum levels of total tocopherol (279.53 mg/kg oil) and xanthophyll (6.18 mg/kg oil) were detected at 20th weeks after the flowering date. These amounts decreased gradually as ripening advances. The early stages of pecan ripening seem to have nutritional and pharmaceutical interests. These results may be useful for evaluating the pecan nut quality and determining the optimal period when the pecans accumulated the maximum of these nutritional and healthy compounds.  相似文献

Alkaline phosphatase (ALP) is widely used as an indicator of proper pasteurization in bovine milk. Due to interest in the use of equine ALP as a time/temperature integrator (TTI) for evaluation of the efficacy of thermal processing of equine milk, its inactivation kinetics were evaluated in whole and skimmed equine milk. Experimentally determined decimal reduction times showed that equine ALP is more readily inactivated in equine milk than its bovine counterpart. Thus, considering the required 6 D reduction of pathogens and the rather low enzyme level present in equine milk, equine ALP will not be suitable as indicator for correct pasteurization of equine milk under the conditions currently used in the reference method for the determination of ALP in milk-based products.  相似文献

A model of clogging of a pleated filter in gas filtration is presented. The model is obtained by combining a semi-analytical model of the flow in a pleated filter and an empirical model of clogging of the planar filter medium applied locally along the pleated filter channels. The model takes into account the formation of a filtration cake of variable thickness at the porous wall of the pleated filter entrance channels and the resulting evolution of the entrance channels aperture distribution during the filtration/clogging process. Based on the numerical predictions, two main clogging scenarios are identified and analyzed. The optimum pleat density, defined as the pleat density maximizing the filter capacity, is determined and shown to be greater than the pleat density minimizing the pressure drop for a clean filter. Predictions of the evolution of overall pressure drop across the pleated filter due to clogging are compared with experimental data.  相似文献

A mutant of D ‐fructose‐6‐phosphate aldolase (FSA) of Escherichia coli, FSA A129S, with improved catalytic efficiency towards dihydroxyacetone (DHA), the donor substrate in aldol addition reactions, was explored for synthetic applications. The kcat/KM value for DHA was 17‐fold higher with FSA A129S than that with FSA wild type (FSA wt). On the other hand, for hydroxyacetone as donor substrate FSA A129S was found to be 3.5‐fold less efficient than FSA wt. Furthermore, FSA A129S also accepted glycolaldehyde (GA) as donor substrate with 3.3‐fold lower affinity than FSA wt. This differential selectivity of both FSA wt and FSA A129S for GA makes them complementary biocatalysts allowing a control over donor and acceptor roles, which is particularly useful in carboligation multi‐step cascade synthesis of polyhydroxylated complex compounds. Production of the mutant protein was also improved for its convenient use in synthesis. Several carbohydrates and nitrocyclitols were efficiently prepared, demonstrating the versatile potential of FSA A129S as biocatalyst in organic synthesis.  相似文献

A biofilm model for flowing systems in the food industry   总被引:1,自引:0,他引:1  
When bacteria attach to the walls of pipelines, they can form biofilms, which can cause the recontamination of food products. In order to quantify such recontamination, a one-dimensional biofilm model was developed taking into account adsorption, desorption, and the growth of cells. The model consisted of two mass balances describing increases in biofilm formation at the wall and the accumulation of cells in the liquid phase. The necessary parameters for the model were obtained in laboratory biofilm experiments. These experiments involved a flowing system and the use of Staphylococcus aureus as a model pathogen and silicon tubing as a testing material. S. aureus was inoculated into the system for 2 h, and then the system was changed to a sterile medium. Both biofilm formation and the release of cells into the flowing liquid were measured until steady-state conditions were reached (for up to 9 days). The experiments were performed in duplicate for different flow conditions (i.e., for Reynolds numbers of 3.2, 32, and 170). It was shown that at higher Reynolds numbers, the biofilm developed faster, probably owing to an increase in the transfer of nutrients to the surface. The proposed biofilm model was capable of describing the data obtained for the three different flow conditions with the use of the specific growth rate in the biofilm and the desorption coefficient as fit parameters. The specific growth rates were 0.16, 0.27, and 0.49 h(-1) for Reynolds numbers of 3.2, 32, and 170, respectively, and the desorption coefficients were about 1% of these values.  相似文献

Fermentation of grain legumes is an efficient method to reduce the concentration of α‐galactosidic compounds that are known to be flatulence producers. Soluble dietary fibre has also been implicated in flatulence production; however, little information exists about the effectiveness of fermentation in diminishing the effects of these compounds. The objective of this work was to study the effect of natural fermentation (NF) and controlled fermentation (CF) on the content of α‐galactosides and dietary fibre in dry beans (Phaseolus vulgaris) for 24, 48, 72 and 96 h. After 48 h, the pH during NF dropped from 6.15 to 4.00 and the nominal acidity increased six times; for CF, however, although the decrease in pH was similar to that for NF, the nominal acidity increased only three times after 48 h. Insoluble fibre content did not change the pH significantly after 96 h for NF and CF. Soluble fibre suffered an apparent removal after 48 h of NF and underwent a sharp reduction of 66% after 96 h of CF. The concentration of stachyose (the main α‐galactoside in raw beans) diminished notably after 48 h and 96 h NF (72% and 95% respectively), whereas with CF only 11% was removed after 96 h.
