Emotions play a central role in our daily lives, influencing the way we think and act, our health and sense of well-being, and films are by excellence the form of art that exploits our affective, perceptual and intellectual activity, holding the potential for a significant impact. Video is becoming a dominant and pervasive medium, and online video a growing entertainment activity on the web and iTV, mainly due to technological developments and the trends for media convergence. In addition, the improvement of new techniques for gathering emotional information about videos, both through content analysis or user implicit feedback through user physiological signals complemented in manual labeling from users, is revealing new ways for exploring emotional information in videos, films or TV series, and brings out new perspectives to enrich and personalize video access. In this work, we reflect on the power that emotions have in our lives, on the emotional impact of movies, and on how to address this emotional dimension in the way we classify and access movies, by exploring and evaluating the design of iFelt in its different ways to classify, access, browse and visualize movies based on their emotional impact.  相似文献

Computational fluid dynamics (CFD) is increasingly used to analyze wind turbines, and the next logical step is to develop CFD‐based optimization to enable further gains in performance and reduce model uncertainties. We present an aerodynamic shape optimization framework consisting of a Reynolds‐averaged Navier Stokes solver coupled to a numerical optimization algorithm, a geometry modeler, and a mesh perturbation algorithm. To efficiently handle the large number of design variables, we use a gradient‐based optimization technique together with an adjoint method for computing the gradients of the torque coefficient with respect to the design variables. To demonstrate the effectiveness of the proposed approach, we maximize the torque of the NREL VI wind turbine blade with respect to pitch, twist, and airfoil shape design variables while constraining the blade thickness. We present a series of optimization cases with increasing number of variables, both for a single wind speed and for multiple wind speeds. For the optimization at a single wind speed performed with respect to all the design variables (1 pitch, 11 twist, and 240 airfoil shape variables), the torque coefficient increased by 22.4% relative to the NREL VI design. For the multiple‐speed optimization, the torque increased by an average of 22.1%. Depending on the CFD mesh size and number of design variables, the optimization time ranges from 2 to 24h when using 256 cores, which means that wind turbine designers can use this process routinely. BACKGROUND: Mushroom polysaccharides play an important role in functional foods because they exhibit biological modulator properties such as antitumour, antiviral and antibacterial activities. The present study involved the production, purification and characterisation of intracellular and extracellular free and protein‐bound polysaccharides from Pleurotus ostreatus and the investigation of their growth‐inhibitory effect on human carcinoma cell lines. RESULTS: Several fermentation parameters were obtained: batch polysaccharide productivities of 0.013 ± 8.12 × 10?5 and 0.037 ± 0.0005 g L?1 day?1 for intracellular and extracellular polysaccharides respectively, a maximum biomass concentration of 9.35 ± 0.18 g L?1, Pmax = 0.935 ± 0.018 g L?1 day?1, µmax = 0.218 ± 0.02 day?1, YEP/X = 0.040 ± 0.0015 g g?1 and YIP/X = 0.014 ± 0.0003 g g?1. Some polysaccharides exhibited superoxide dismutase (SOD)‐like activity of 50‐200 units. Fourier transform infrared analysis of the polysaccharides revealed absorption bands characteristic of such biological macromolecules. Cytotoxicity assays showed that both intracellular and extracellular polysaccharides exhibited antitumour activity towards several tested human carcinoma cell lines in a dose‐dependent manner. CONCLUSION: The polysaccharides of P. ostreatus exhibited high SOD‐like activity, which strongly supports their biological effect on tumour cell lines. The extracellular polysaccharides presented the highest antitumour activity towards the RL95 carcinoma cell line and should be further investigated as an antitumour agent. Dermorphin analogues, containing a (S)‐ and (R)‐4‐amino‐1,2,4,5‐tetrahydro‐2‐benzazepin‐3‐one scaffold (Aba) and the α‐methylated analogues as conformationally constrained phenylalanines, were prepared. Asymmetric phase‐transfer catalysis was unable to provide the (S)‐α‐Me‐o‐cyanophenylalanine precursor for (S)‐α‐MeAba in acceptable enantiomeric purity. However, by using a Schöllkopf chiral auxiliary, this intermediate was obtained in 88 % ee. [(S)‐Aba 3‐Gly 4]dermorphin retained μ‐opioid affinity but displayed an increased δ‐affinity. The corresponding R epimer was considerably less potent. In contrast, the [(R)‐α‐MeAba 3‐Gly 4]dermorphin isomer was more potent than its S epimer. Tar‐MD simulations of both non‐methylated [Aba 3‐Gly 4]dermorphin analogues showed a degree of folding at the C‐terminal residues toward the N terminus of the peptide, without however, adopting a stabilized β‐turn conformation. The α‐methylated analogues, on the other hand, exhibited a type I/I′ β‐turn conformation over the α‐MeAba 3 and Gly 4 residues, which was stabilized by a hydrogen bond involving Tyr 5‐HN and D ‐Ala 2‐CO.  相似文献

Supramolecular peptide hydrogels are gaining increased attention, owing to their potential in a variety of biomedical applications. Their physical properties are similar to those of the extracellular matrix (ECM), which is key to their applications in the cell culture of specialized cells, tissue engineering, skin regeneration, and wound healing. The structure of these hydrogels usually consists of a di- or tripeptide capped on the N-terminus with a hydrophobic aromatic group, such as Fmoc or naphthalene. Although these peptide conjugates can offer advantages over other types of gelators such as cross-linked polymers, they usually possess the limitation of being particularly sensitive to proteolysis by endogenous proteases. One of the strategies reported that can overcome this barrier is to use a peptidomimetic strategy, in which natural amino acids are switched for non-proteinogenic analogues, such as D-amino acids, β-amino acids, or dehydroamino acids. Such peptides usually possess much greater resistance to enzymatic hydrolysis. Peptides containing dehydroamino acids, i.e., dehydropeptides, are particularly interesting, as the presence of the double bond also introduces a conformational restraint to the peptide backbone, resulting in (often predictable) changes to the secondary structure of the peptide. This review focuses on peptide hydrogels and related nanostructures, where α,β-didehydro-α-amino acids have been successfully incorporated into the structure of peptide hydrogelators, and the resulting properties are discussed in terms of their potential biomedical applications. Where appropriate, their properties are compared with those of the corresponding peptide hydrogelator composed of canonical amino acids. In a wider context, we consider the presence of dehydroamino acids in natural compounds and medicinally important compounds as well as their limitations, and we consider some of the synthetic strategies for obtaining dehydropeptides. Finally, we consider the future direction for this research area.  相似文献

This study aims to estimate changes in the prevalence of ecstasy use over time, analyze the overlap of ecstasy use and other drug use, and compare other drug use in ecstasy versus marijuana users. The authors hypothesized that ecstasy users early in the "epidemic" would be polydrug users and that associations between ecstasy and other drug use would diminish as the prevalence of ecstasy use increased. Data were drawn from public use data files from the 1995, 1997, 1999, and 2001 National Household Survey on Drug Abuse. Ecstasy use increased in the U.S. population and the prevalence was greater in younger age groups. Ecstasy users were likely to use a variety of other drugs; however, association of ecstasy use with other drug use was strongest early in the "epidemic," diminishing as the number of new users increased. Later, more drug-naive adolescents and young adults began experimenting with ecstasy. These results can orient prevention strategies that target ecstasy users. In the framework of the TBM Program, three ITER vacuum vessel equatorial ports (#16, #18 and #02) have been allocated for the testing of up to six mock-ups of six different DEMO tritium breeding blankets. Each one is called a Test Blanket System (TBS). A TBS consists mainly of the Test Blanket Module (TBM), the in-vessel component facing the plasma, and several ancillary systems, in particular the cooling system and the tritium extraction system. Each port accommodates two TBMs and therefore the two TBSs have to share the corresponding port cell. This paper deals with the design integration aspects of the two TBSs in each port cell performed at ITER Organization (IO) with the corresponding definition of interfaces with other ITER systems. The performed activities have raised several issues that are discussed in the paper and for which design solutions are proposed.  相似文献
