A wide variety of biological activities including the major metabolic actions of insulin is regulated by phosphatidylinositol (PI) 3-kinase. However, the downstream effectors of the various signaling pathways that emanate from PI 3-kinase remain unclear. Akt (protein kinase B), a serine-threonine kinase with a pleckstrin homology domain, is thought to be one such downstream effector. A mutant Akt (Akt-AA) in which the phosphorylation sites (Thr308 and Ser473) targeted by growth factors are replaced by alanine has now been shown to lack protein kinase activity and, when overexpressed in CHO cells or 3T3-L1 adipocytes with the use of an adenovirus vector, to inhibit insulin-induced activation of endogenous Akt. Akt-AA thus acts in a dominant negative manner in intact cells. Insulin-stimulated protein synthesis, which is sensitive to wortmannin, a pharmacological inhibitor of PI 3-kinase, was abolished by overexpression of Akt-AA without an effect on amino acid transport into the cells, suggesting that Akt is required for insulin-stimulated protein synthesis. Insulin activation of p70 S6 kinase was inhibited by approximately 75% in CHO cells and approximately 30% in 3T3-L1 adipocytes, whereas insulin-induced activation of endogenous Akt was inhibited by 80 to 95%, by expression of Akt-AA. Thus, Akt activity appears to be required, at least in part, for insulin stimulation of p70 S6 kinase. However, insulin-stimulated glucose uptake in both CHO cells and 3T3-L1 adipocytes was not affected by overexpression of Akt-AA, suggesting that Akt is not required for this effect of insulin. These data indicate that Akt acts as a downstream effector in some, but not all, of the signaling pathways downstream of PI 3-kinase.  相似文献

Loss of genetic material, including loss of loci on chromosome arms 6q, 9p, and 10q, occurs frequently in cutaneous melanoma but infrequently in benign melanocytic nevi or other melanocytic lesions, suggesting that these genetic alterations are important in the development and progression of melanoma. To examine whether allelic loss is of prognostic importance in melanoma, disease-free survival was related to loss of heterozygosity on 6q, 9p and 10q in 83 individuals with sporadic primary cutaneous melanoma. Loss of chromosome arms 6q and 10q were each significantly associated with a poorer clinical outcome (P=0.013 and P=0.001 respectively). In a subgroup of 41 subjects whose primary tumours were allelotyped, the fractional allelic loss (FAL) at 39 autosomal arms also significantly correlated with disease-free survival (P=0.013), with an increase in FAL associated with a poorer outcome; this association remained significant when controlled for tumour thickness (P=0.035). In addition, a greater proportion of cells were immunopositive for Ki67 antigen, p53 and p21WAF1 protein in the primary melanomas than in the benign melanocytic nevi, however, only p53 over-expression was significantly associated with improved survival (P=0.041).  相似文献

The authors have succeeded in exciting a new type of leaky surface acoustic wave (LSAW) having only a shear horizontal (SH) component that has a large electromechanical coupling factor, a large reflection coefficient, and excellent temperature stability, by combining interdigital transducers (IDTs) and reflectors made of heavy-metal films such as gold (Au), tantalum (Ta), and tungsten (W) on the ST-cut 90/spl deg/ X propagation (direction perpendicular to the X-axis) quartz substrate. This LSAW does not have a propagation decay. The square of the electromechanical coupling factor is 2.1-2.7 times larger than, the reflection coefficient is 30 times larger than, and the temperature characteristic is the same as those of a Rayleigh wave on an ST-cut X propagation quartz substrate. The authors applied this SH LSAW to resonators and resonator filters. As a result, we succeeded in developing the low loss and very small-sized resonators and resonator filters (1/5-1/4 of conventional device sizes) with IDTs with a small number of finger pairs and very small reflectors, for the first time.  相似文献

A VLSI-oriented variable-length pipeline structure for data-driven processors is presented. Ordinary inline pipelines have the problem of minimizing the average total processing time through the pipeline, since subdivision of a function along the pipeline is usually optimized for the most complex operations in spite of the fact that simpler operations need fewer stages. As a solution to this problem, a variable-length pipeline scheme in which data go through only the necessary stages according to information contained within is proposed. The scheme has been implemented on a test chip to verify performance. The chip demonstrated a minimum fall-through time (data transmission time from input to output) of 14.4 ns and a data transmission rate in the pipeline of 59 megaword/s (that is, 1/16.9 ns) as a first-in first-out (FIFO) store. By modifying the data transfer control and allocating the processing functions corresponding to the data interval of 16.9 ns, this scheme is applicable as a high-performance processing unit for data-driven processors  相似文献

The high compatibility of fluorene‐based polyester (FBP‐HX) as a polymer matrix for multiwalled carbon nanotubes (MWCNTs) is discussed. A low surface resistivity due to the fine dispersion of MWCNTs in FBP‐HX and polycarbonate (PC) is reported. With a solution‐casting method, a percolation threshold with the addition of between 0.5 and 1.0 wt % MWCNTs was observed in the MWCNT/PC and MWCNT/FBP‐HX composites. Because of the coverage of FBP‐HX on the MWCNTs, a higher surface resistivity and a higher percolation ratio of the MWCNT/FBP‐HX composites were achieved compared with the values for the MWCNT/PC composites. In the MWCNT/FBP‐HX composites, MWCNTs covered with FBP‐HX were observed by scanning electronic microscopy. Because of the coverage of FBP‐HX on the MWCNTs, FBP‐HX interfered with the electrical pathway between the MWCNTs. The MWCNTs in FBP‐HX were covered with a 5‐nm layer of FBP‐HX, but the MWCNTs in the MWCNT/PC composites were in their naked state. MWCNT/PC sheets demonstrated the specific Raman absorption of the MWCNTs only with the addition of MWCNTs of 1 wt % or above because of the coverage of the surface of the composite sheet by naked MWCNTs. In contrast, MWCNT/FBP‐HX retained the behavior of the matrix resin until a 3 wt % addition of MWCNTs was reached because of the coverage of MWCNTs by the FBP‐HX resin, induced by its high wettability. © 2010 Wiley Periodicals, Inc. It has been reported that radon inhalation activates antioxidative functions in liver and has an antioxidative effect against hepatopathy similar to that of the antioxidative effects of ascorbic acid (VC) or α-tocopherol (VE). In this study, we examined the combined effects of radon inhalation and antioxidant vitamin administration on acute alcohol-induced hepatopathy in mice. ICR mice were subjected to intraperitoneal (i.p.) administration of alcohol after pretreating with air only (sham) or radon at a concentration of approximately 2000 Bq/m3 for 24 hours and i.p. administration of VC (300 mg/kg body weight) or VE (300 mg/kg body weight). In mice injected with alcohol, the combined radon and antioxidant vitamins treatment significantly decreased the activities of glutamic oxaloacetic transaminase in serum compared to not only the alcohol-administered group (sham group), but also the radon inhalation with alcohol administration group or the vitamin and alcohol administration group. In addition, radon inhalation significantly increased the antioxidant level, in such as the catalase activity and the total glutathione content in liver compared to the sham group. These results suggested that the combined radon and antioxidant vitamin treatment could effectively inhibit alcohol-induced hepatopathy in mice without any antagonizing action.  相似文献

This paper describes an ultrahigh‐speed permanent‐magnet synchronous motor drive, which is embedded in a turbocharger of an internal‐combustion engine. The electrical drive makes it possible to enhance output power of the turbocharger in a motoring mode and to retrieve combustion energy from exhaust gas in a regenerating mode. Computer simulations and experimental tests are conducted to examine various operation characteristics of a prototype. The experimental data demonstrate 220,000‐r/min operation at 2.2‐kW inverter output power, in good agreement with the simulation results and proving the feasibility of the proposed system. © 2007 Wiley Periodicals, Inc.
