BACKGROUND: Telomerase, an enzyme associated with cellular immortality, is expressed by most malignant cells and is inactive in most normal somatic cells, with the excitation of proliferative stem cells, male germ cells, and activated lymphocytes. The measurement of telomerase activity in clinically obtained tissue samples may provide useful information as both a diagnostic and prognostic marker. In this study, we sought to determine whether telomerase activity might prove helpful in the assessment of benign and malignant thyroid tumors. METHODS: A modified, semiquantitative polymerase chain reaction-based telomeric repeat amplification protocol assay was used for detection of telomerase activity in 59 samples obtained at thyroidectomy, including 15 thyroid cancers, 22 benign thyroid diseases, and 22 adjacent normal thyroid tissues. RESULTS: Four of 13 differentiated thyroid carcinomas (30%) and 2 of 2 medullary carcinomas (100%) expressed telomerase activity. Unexpectedly, we also detected activity in 3 of 22 (14%) adjacent normal thyroid tissues and 6 of 22 (28%) benign thyroid diseases. Pathologic review of the telomerase-positive benign specimens revealed that many contained extensive lymphoid infiltrates with germinal centers (six of nine, 67%), as did two of four telomerase-positive papillary carcinomas. CONCLUSIONS: In contradistinction to other epithelial carcinomas, telomerase does not appear to be frequently reactivated in differentiated thyroid carcinomas.  相似文献

The aim of this study was to evaluate the cumulative net free-living nitrogen (N) fixation and its response to elevated atmospheric carbon dioxide (CO2) in the submerged surface of paddy soil. The study site was an actual paddy area in central Japan that was equipped with a free-air CO2 enrichment (FACE) facility and was composed of four bays, where each bay had an elevated CO2 plot (FACE plot) and an ambient plot. Field incubation was conducted at subplots without N fertilizer application from May 28 to August 18, 2012 (82 days). The CO2 enrichment was an average of +528 ppm during the study period at the subplots. The changes in total N (TN) content in the surface soil (0–1 cm) were determined in comparison to the initial TN content. The cumulative net N fixations during the 82-day study period were 47.1 ± 3.7 and 43.3 ± 5.8 (standard error) kg N ha?1 in the FACE and ambient plots, respectively. The difference between the FACE and ambient plots was not significant (p = 0.05). However, these values were partly affected by the charophyte blooms, which are not involved in N fixation. The results are not conclusive, and the one bay without charophyte blooming displayed a significant increase in the cumulative net N fixation (approximately 10 kg N ha?1) with CO2 enrichment (p < 0.001).  相似文献

A complete series of well-crystallized solid-solution Sr1– X -CaXMoO4 films has been prepared on molybdenum substrates in an electrolytic solution containing Sr2+ and Ca2+ ions by an electrochemical method at room temperature (25°C). The peak positions of excitation and emission of the Sr1–XCaXMoO4 solid-solution films were independent of the Ca content: 285 ± 2 nm for excitation and 536 ± 2 nm for emission at liquid-nitrogen temperature (−196°C).  相似文献

Optical code-division multiple access (OCDMA) for high-speed multimedia transmission is proposed and its performance is investigated. The proposed system introduces time hopping to vary transmission bit rate and power control to control transmission signal performance, which allows various signals with different desired rate and performance. An expression for the bit error rate (BER) of the proposed system is derived and numerical results are shown. An optical power selector (OPS) coupled with a hard limiter are also proposed to improve system performance. The OPS is a device to transmit the instantaneous maximum optical power among input signals. The OPS combined with the optical hard limiter reduces the interference from simultaneous users which results in an improved BER. BER is theoretically derived and numerical results for some parameters are shown. We show that the proposed system significantly improves BER and is an attractive scheme for future multimedia networks.  相似文献

Rare-earth (Nd, Sm, Er, Yb, Sc)-doped zirconia was melted using an arc-imaging furnace, and rapidly quenched with a hammer and anvil apparatus (cooling rate >105 K sec–1). These ZrO2-RO1.5 samples were investigated by X-ray diffraction and transmission electron microscopy. The existing region of metastable tetragonal zirconia is from 2 to 14 mol% of RO1.5 regardless of the species of dopants, RO1.5. In the lattice parameters of the tetragonal phases, the unit cell volume corresponds to the ionic radii of dopants, whereas the tetragonality (c/a) is independent of the species of dopants, but dependent on the content of dopants.  相似文献

Poly(9-vinyladenine) was synthesized and utilized as an affinity macroligand entrapped within the gel matrix. Base-specific separation of oligodeoxynucleotides was achieved with high resolution and high speed by electrophoresis, using capillaries filled with conjugated polyacrylamide-poly(9-vinyladenine) gel. Oligothymidylic acids were selectively separated from the mixture of oligothymidylic and oligodeoxyadenylic acids by utilizing a specific hydrogen bonding between poly(9-vinyladenine) and oligothymidylic acids. Migration time and resolution of oligodeoxynucleotides were influenced by several parameters, such as the size of poly(9-vinyladenine), capillary temperature, and concentrations of poly(9-vinyladenine) and urea. Some guidelines are presented, based on the theoretical formulation of the effect of these parameters, in order to find optimum electrophoretic conditions. Analytical capillary affinity gel electrophoresis was developed for the selective and sensitive base recognition of oligodeoxynucleotides with efficiencies as high as several 10(6) plates/m by using a urea-gel capillary with poly(9-vinyladenine) and temperature-programming.  相似文献

The conversion coefficients, H'(d,α)/Φ, for monoenergetic positrons and positron-emitting radionuclides were calculated by using the user code UCICRPM of the Monte Carlo code EGS5 to estimate the radiation dose for medical staff involved in positron emission tomography examinations. From these coefficients, the dose equivalent rates per unit activity at 0.07 and 10 mm depths in a soft tissue for a straight-line source of 2-deoxy-2-[(18)F]fluoro-d-glucose ((18)F-FDG) were calculated by using the developed user code UCF18DOSE. The dose equivalent rates per unit activity at 0.07 and 10 mm depths were measured by using a personal dosemeter (DOSE(3)) under the same conditions as those considered in the calculation. The calculated dose equivalent rates per unit activity at 0.07 and 10 mm depths were 0.116 and 0.0352 pSv min(-1) Bq(-1), respectively, at 20 cm from the (18)F-FDG injection tube.  相似文献

SF6 is used as the main insulation gas for gas‐insulated switchgear (GIS), but it has recently become a gas that must be restricted because of its greenhouse effects. To date, we have studied the insulation characteristics of compressed N2 and CO2 as possible alternatives for SF6. We have reported that N2 or CO2 must be pressurized to 2.0 MPa when it is used as a substitute for SF6 at 0.5 MPa. Therefore, we have proposed a hybrid installation composition that uses gas and solid insulators. Because the central conductor of GIS is covered by a solid insulator in this composition, a high‐pressure gas at 2.0 MPa is not needed. However, the joint of the solid insulator becomes a weak point for discharge development. In this paper, we describe an effective configuration for improvement of the withstand voltage based on experiments. The most effective connector was made of resin without an implant electrode and the most effective configuration was one without a solid–solid interface between the solid insulator of the central conductor and the resin connector. In this experiment, the improvement of breakdown electric field of the hybrid composition was 44% or more compared with the case of only gas insulation (conventional method). In addition, further improvement can be expected by optimizing the insulation creepage distance and configuration. © 2011 Wiley Periodicals, Inc. Lipid packing has a strong influence on the formation and structural dynamics of cell membranes. Techniques to modulate lipid packing may thus enable modification of cellular functions and events. An 18-residue amphiphilic helical peptide derived from the N-terminal segment of epsin-1 (EpN18) is reported to induce positive membrane curvature and to loosen lipid packing in the cell membrane. In this study, it is shown that EpN18, crosslinked to a leucine-zipper peptide K4, is recruited to the cell surface by interacting with a cell-surface-expressed E3 leucine-zipper segment. Cell-surface tethering markedly enhanced loosening of lipid packing, which led to the promotion of membrane translocation of octaarginine. The loosening of lipid packing by EpN18 was also confirmed by analyzing the generalized polarization value with a membrane-environment-sensitive dye, 2-hydroxy-3-{2-[(2-hydroxyethyl)dimethylamino]ethyl}-4-{2-[6-(dibutylamino)-2-naphthyl]ethenyl}pyridiniumdibromide (di-4-ANEPPDHQ). This approach thus shows promise for the control of lipid packing and related cellular events.  相似文献
