The cerebral synthesis of cholesterol is mainly handled by astrocytes, which are also responsible for apoproteins’ synthesis and lipoproteins’ assembly required for the cholesterol transport in the brain parenchyma. In Alzheimer disease (AD), these processes are impaired, likely because of the astrogliosis, a process characterized by morphological and functional changes in astrocytes. Several ATP-binding cassette transporters expressed by brain cells are involved in the formation of nascent discoidal lipoproteins, but the effect of beta-amyloid (Aβ) assemblies on this process is not fully understood. In this study, we investigated how of Aβ1-42-induced astrogliosis affects the metabolism of cholesterol in vitro. We detected an impairment in the cholesterol efflux of reactive astrocytes attributable to reduced levels of ABCA1 transporters that could explain the decreased lipoproteins’ levels detected in AD patients. To approach this issue, we designed biomimetic HDLs and evaluated their performance as cholesterol acceptors. The results demonstrated the ability of apoA-I nanodiscs to cross the blood–brain barrier in vitro and to promote the cholesterol efflux from astrocytes, making them suitable as a potential supportive treatment for AD to compensate the depletion of cerebral HDLs.  相似文献

Gangliosides constitute a subgroup of glycosphingolipids characterized by the presence of sialic acid residues in their structure. As constituents of cellular membranes, in particular of raft microdomains, they exert multiple functions, some of them capital in cell homeostasis. Their presence in cells is tightly regulated by a balanced expression and function of the enzymes responsible for their biosynthesis, ganglioside synthases, and their degradation, glycosidases. The dysregulation of their abundance results in rare and common diseases. In this review, we make a point on the relevance of gangliosides and some of their metabolic precursors, such as ceramides, in the function of podocytes, the main cellular component of the glomerular filtration barrier, as well as their implications in podocytopathies. The results presented in this review suggest the pertinence of clinical lipidomic studies targeting these metabolites.  相似文献

We modified synthetic isoprene rubber by means of the in situ generation of silica particles through the sol–gel process starting from tetraethoxysilane (TEOS) as an inorganic oxide precursor. Different reaction conditions were investigated with variations in the initial TEOS content, the reaction time of the sol–gel process, and the presence of a coupling/surfactant agent (octyltriethoxysilane). Organic–inorganic hybrid materials with a silica content up to 70 phr were obtained with the complete conversion of TEOS to silica for a long enough sol–gel reaction time. A very homogeneous dispersion of silica particles was observed in all cases together with a very good adhesion between the filler and matrix. The size of the in situ generated silica was controlled by the appropriate addition of octyltriethoxysilane. Swelling and extraction tests and dynamic mechanical analysis indicated that the vulcanization process of isoprene rubber was perturbed by the sol–gel process; this led to a slight decrease in the crosslinking degree. Metastable tetragonal ZrO2 phase has been observed in ZrO2–SiO2 binary oxides prepared by the sol–gel method. There are many studies concerning the causes of ZrO2 tetragonal stabilization in binary oxides such as Y3O2–ZrO2, MgO–ZrO2, or CaO–ZrO2. In these binary oxides, oxygen vacancies cause changes or defects in the ZrO2 lattice parameters, which are responsible for tetragonal stabilization. Since oxygen vacancies are not expected in ZrO2–SiO2 binary oxides, tetragonal stabilization should just be due to the difficulty of zirconia particles growing in the silica matrix. Furthermore, changes in the tetragonal ZrO2 crystalline lattice parameters of these binary oxides have recently been reported in a previous paper. The changes of the zirconia crystalline lattice parameters must result from the chemical interactions at the silica–zirconia interface (e.g., formation of Si–O–Zr bonds or Si–O groups). In this paper, FT-IR and 29Si NMR spectroscopy have been used to elucidate whether the presence of Si–O–Zr or Si–O is responsible for tetragonal phase stabilization. Moreover, X-ray diffraction, Raman spectroscopy, and transmission electron microscopy have also been used to study the crystalline characteristics of the samples.  相似文献

The vibration and noise radiation from fluid-loaded cylindrical shells are controlled by using multiple stiffeners and passive constrained layer damping treatment. Dynamic and fluid finite elements are developed to study the fundamental phenomena governing the coupling between the stiffened shell, with and without damping, and the fluid domain surrounding it. The models are used to predict the response of the shell and to evaluate the effect of stiffening rings and damping treatment on both the structural vibration and noise radiation in the fluid domain. The geometry of the shell and fluid domain allows the formulation of a harmonic-based model, which uncouples the fluid–structural response of modes corresponding to different numbers of circumferential nodes.In this study, it is shown that stiffening of the shell reduces the amplitude of the vibration and noise radiation, particularly for high-order lobar modes. The attenuation of the shell response and sound radiation can be increased significantly through the application of passive constrained layer damping treatments on the inner surface of the stiffening rings.  相似文献

This paper presents a global approach to the validation of the parameters that enter into the neutronics simulation tools for advanced fast reactors with the objective to reduce the uncertainties associated to crucial design parameters. This global approach makes use of sensitivity/uncertainty methods; statistical data adjustments; integral experiment selection, analysis and “representativity” quantification with respect to a reference system; scientifically based cross-section covariance data and appropriate methods for their use in multigroup calculations. This global approach has been applied to the uncertainty reduction on the criticality of the Advanced Burner Reactor (both metal and oxide core versions) presently investigated in the frame of the GNEP initiative. The results obtained are very encouraging and allow to indicate some possible improvements of the ENDF/B-VII data file.  相似文献

The effect of organic production on broiler carcass and meat quality was assessed. Two hundred and fifty Ross male chickens were assigned to two different systems of production: conventional, housing in an indoor pen (0.12 m(2)/bird); organic, housing in an indoor pen (0.12 m(2)/bird) with access to a grass paddock (4 m(2)/bird). At 56 and 81 days of age, 20 chickens per group were slaughtered to evaluate carcass traits and the characteristics of breast and drumstick muscles (m. pectoralis major and m. peroneus longus). The organic chickens had carcasses with a higher breast and drumstick percentages and lower levels of abdominal fat. The muscles had lower pHu and water holding capacity. Instead cooking loss, lightness values, shear values, Fe, polyunsaturated fatty acids of n-3 series and TBA-RS were higher. The sensory quality of the breast muscle was better. Organic production system seems to be a good alternative method, due to better welfare conditions and good quality of the carcass and meat. A negative aspect was the higher level of TBA-RS in the muscles, probably due to greater physical activity.  相似文献

Pd-loaded Ce0.6Zr0.4O2 solid solutions supported on Al2O3 are investigated as catalysts for the reduction of NO by CO. The attention is focused on the role of the Ce0.6Zr0.4O2 and of the Pd dispersion on the catalytic activity. The system shows a very high activity below 500 K, which is almost independent on the Pd dispersion. The high activity is attributed to a promoting effect of the Ce0.6Zr0.4O2 on the NO conversion. Investigation of the influence of high temperature treatments disclosed a thermal stabilisation of both Ce0.6Zr0.4O2 and Al2O3 in the Ce0.6Zr0.4O2/Al2O3 system.  相似文献
