In the present work, a rapid and reproducible molecular method, based on the combination of conventional and multiplex RAPD-PCR reactions, was developed for typing Lactobacillus sanfranciscensis isolates from traditional sourdoughs. At first, four random primers, two used singly and two combined with the primer RD1, were chosen on the basis of their differentiating capability and reproducibility. The four resulting profiles for each isolate were integrated into a unique profile to be statistically treated by cluster analysis. The method was validated on 58 L. sanfranciscensis isolates coming from three traditional Italian sourdoughs. This new RAPD method was useful for determining the genomic diversity within the L. sanfranciscensis species. In particular, the intraspecific diversity of this species seemed to be related to the sourdough origin. 相似文献

To modify the properties of edible soy protein isolate (SPI) films, 0.5% anthocyanin-rich red raspberry (Rubus strigosus) extract (ARRE) (0.5 g raspberry powder in 95% ethyl alcohol/water/85% lactic acid [80:19:1. v/v/v]) was incorporated into film-forming solutions. ARRE resulted in an SPI film having significantly enhanced tensile strength (P < 0.05) and % elongation at break (P < 0.05), as well as increased water swelling ratio (P < 0.05) and in vitro pepsin digestibility (P < 0.05). The resultant films also showed significantly decreased water solubility and water vapor permeability (P < 0.05). In addition, ARRE increased darkness, redness, and yellowness film appearance as evidenced by a lower L* (P < 0.05), greater positive a* (P < 0.05), and a higher b* (P < 0.05) than the control film. Scanning electron microscopy images revealed that extract-added films had denser and more compact cross-section microstructure. Fourier transform infrared spectra illustrated that ARRE-created hydrogen bonding involved conformational changes of soy protein without destroying its backbone structure. SDS-PAGE electrophoretograms revealed that the extract induced intermolecular interaction of the soy protein monomers. Natural plant extracts would be a promising ingredient to make SPI films with different physicochemical properties and applications. PRACTICAL APPLICATION: This study characterizes the potential physicochemical changes of SPI film with incorporated raspberry extract. Upon the above modification, the resultant film was found to enhance the applications of pure SPI film in food packaging. For example, SPI-ARRE film could prolong the usage life of SPI film due to increased strength, or could be useful as a desiccant (drying agent) such as a water-absorbing sheet for preserving dried foods due to its increased hydrophilic surface and water-swelling ratio. The effects of transverse hydrodynamic dispersion on altering transformation-induced compound-specific isotope analysis (CSIA) signals within groundwater pollution plumes have been assessed with reactive transport modeling accommodating diffusion-induced isotope fractionation (DIF) and implementing different parameterizations of local transverse dispersion. The model reproduced previously published field data showing a negative carbon isotope pattern (-2 ‰) at the fringes of a nondegrading PCE plume. We extended the study to reactive transport scenarios considering vinyl chloride as a model compound and assessing, through a detailed sensitivity analysis, the coupled effects of transverse hydrodynamic dispersion (with and without DIF) and aerobic fringe degradation on the evolution of carbon and chloride isotope ratios. Transformation-induced positive isotope signals were increasingly attenuated with distance from the source and higher degradation rate. The effect of DIF on the overall isotope signal attenuation was greatest near the source and for low values of groundwater flow velocity, transverse dispersion coefficient, molecular weight, rate constant, and isotope fractionation factor, α, of the degradation reaction. Models disregarding DIF underestimate the actual α. The approximately twice larger DIF effect for chlorine than for carbon together with the low α for oxidation resulted in strong chlorine CSIA depletions for VC at the plume fringe. 相似文献

Members of the YERO57c/YJGFc/UK114 protein family have been identified in bacteria and eukaryotes. The budding yeast Saccharomyces cerevisiae contains two different proteins of this family, Hmf1p and Mmf1p. We have previously shown that Mmf1p is a mitochondrial protein functionally related to its human homologue and able to influence the maintenance of mitochondrial DNA. Deletion of Mmf1 results in loss of the mitochondrial genome. Using a multicopy suppression approach, we have identified a protein of the mitochondrial large ribosomal subunit, MRPL40, which stabilizes mtDNA in Deltammf1 cells. Overexpression of MRPL40 did not prevent loss of mtDNA in a mutant strain lacking the mitochondrial protein Abf2p. Thus, MRPL40 does not have a general effect on mtDNA stability, but it may be specific for the mmf1-null strain. We also show that the Deltamrpl40 cells present a similar phenotype to the mmf1-null strain, having reduced mtDNA stability and growth rate. Furthermore, we observed that rho(+)Deltamrpl40 haploid cells can be obtained when tetrads are directly dissected on medium containing a non-fermentable carbon source. Thus, replication and segregation of the mtDNA can occur in the absence of MRPL40. We also show that another mitochondrial ribosomal protein, MRPL38, is able to overcome the Deltammf1-associated defect. Together, our results suggest a link between Mmf1p and the two mitochondrial ribosomal proteins. 相似文献

The composition and structure of triacylglycerols (TAGs) and partial glycerides of crude palm oil obtained from interspecific hybrid Elaeis oleifera × Elaeis guineensis, grown in Colombia, were fully characterised and compared to data obtained by analysing crude African palm oil. Hybridisation appears to substantially modify the biosynthesis of fatty acids (FAs) rather than their assembly in TAGs. In fact, total FAs analysis showed significant differences between these two types of oil, with hybrid palm oil having a higher percentage of oleic acid (54.6 ± 1.0 vs 41.4 ± 0.3), together with a lower saturated fatty acid content (33.5 ± 0.5 vs 47.3 ± 0.1), while the percentage of essential fatty acid, linoleic acid, does not undergo significant changes. Furthermore, 34 TAG types were identified, with no qualitative differences between African and E. guineensis × E. oleifera hybrid palm oil samples. Short and medium chain FAs (8:0, 10:0, 12:0, 14:0) were utilised, together, to build a restricted number of TAG molecular species. Oil samples from the E. guineensis × E. oleifera hybrid showed higher contents of monosaturated TAGs (47.5–51.0% vs 36.7–37.1%) and triunsaturated TAGs (15.5–15.6% vs 5.2–5.4%). The sn-2 position of TAGs in hybrid palm oil was shown to be predominantly esterified with oleic acid (64.7–66.0 mol% vs 55.1–58.2 mol% in African palm oil) with only 10–15% of total palmitic acid and 6–20% of stearic acid acylated in the secondary position. The total amount of diacylglycerols (DAGs) was in agreement with the values of free acidity; DAG types found were in agreement with the representativeness of different TAG species. 相似文献

Food safety and quality are nowadays a major concern. Any case of food alteration, especially when reported by the media, has a great impact on public opinion. There is an increasing demand for the improvement of quality controls, hence addressing scientific research towards the development of reliable molecular tools for food analysis. DNA barcoding is a widely used molecular-based system, which can identify biological specimens, and is used for the identification of both raw materials and processed food. In this review the results of several researches are critically analyzed, in order to exploit the effectiveness of DNA barcoding in food traceability, and to delineate some best practices in the application of DNA barcoding throughout the industrial pipeline. Predicting the load-carrying capacity of timber beams with holes cannot be performed by usual analysis methods if the failure of the beam is governed by the crack initiation and propagation around the hole at low load levels. Predicting the cracking load is an important design issue because it corresponds to the load-carrying capacity of the timber beam before the crack propagation. One of the models that can be used for the fracture formulation is that of a beam on elastic foundation. In this model a part of the beam is assumed to follow the conditions of beam with elastic foundation which has spring stiffness equal to the fracture properties of the material in the crack surface. Based on beam on elastic foundation model, the cracking load prediction is the target of the paper. Some closed form solutions for the beam with hole are presented. The formulation has been derived for two cases of pure shear and pure bending moment. Finally a semi-empirical formulation for combination of shear and bending moment in the section is presented. The model predictions are compared with the results of an experimental program showing good correlation. The analytical model can therefore be proposed for future revisions of codes of practice such as the Eurocode 5. 相似文献

The aim of this study was to test the ability of visible-near infrared (Vis-NIR) reflectance spectroscopy to predict beef quality traits in the slaughterhouse by directly applying a fiber-optic probe on the carcass surface. Carcasses from 230 young bulls and heifers slaughtered in two commercial abattoirs were included in the experiment. Vis-NIR spectra were recorded on an exposed surface of M. gracilis in the abattoirs 4 to 6 and 14 to 16 h post mortem. Traits evaluated were pH, color indexes (L*, a*, b*, H, and SI), cooking loss, and Warner–Bratzler shear force. Prediction models were satisfactory for pH and color indexes, and promising for cooking loss but not for Warner–Bratzler shear force. Results of this work show that Vis-NIR spectroscopy may be a useful tool for on-line prediction of some physical beef quality traits when applied directly on the carcass surface. 相似文献

We present and evaluate a classification method to estimate tourist presence in an area from cellular network data. Our approach is based on setting up a classifier to label users according to five main classes: residents, commuters, people in-transit, tourists and excursionists. We experiment the approach in some important tourist cities in Italy: Venice, Florence, Turin and Lecce. In the lack of sound groundtruth data, we analysed the composition of different classes obtaining results in line with domain knowledge. Moreover, these results are also supported by an analysis of the locations frequented by the tourists that well conforms with expectations. Finally, the number of users classified as tourists is strongly correlated with official statistics on tourist presence in the area. 相似文献
