Lectin A (LecA) from Pseudomonas aeruginosa is an established virulence factor. Glycoclusters that target LecA and are able to compete with human glycoconjugates present on epithelial cells are promising candidates to treat P. aeruginosa infection. A family of 32 glycodendrimers of generation 0 and 1 based on a bifurcated bis‐galactoside motif have been designed to interact with LecA. The influences both of the central multivalent core and of the aglycon of these glycodendrimers on their affinity toward LecA have been evaluated by use of a microarray technique, both qualitatively for rapid screening of the binding properties and also quantitatively (Kd). This has led to high‐affinity LecA ligands with Kd values in the low nanomolar range (Kd=22 nm for the best one).  相似文献

Diapause, which occurs through the production of dormant eggs, is a strategy used by some zooplankton to avoid winter months of persistent low temperatures and low food availability. However, reports of active zooplankton under the ice indicate that other strategies also exist. This study was aimed at evaluating whether the composition of storage lipids in the fall differs between diapausing and active overwintering Daphnia. We assessed the quantity of storage lipids and fatty acid (FA) composition of Daphnia species, along with FA content of seston, in six boreal, alpine and subarctic lakes at the onset of winter, and evaluated the association between storage lipids and Daphnia overwintering strategy. We found that active overwintering Daphnia had >55% body fat and the highest FA concentrations. Polyunsaturated FA, especially stearidonic acid (18:4n-3; SDA) and high ratios of n-3:n-6, were preferentially retained to a greater extent in active overwintering Daphnia than in those that entered diapause. Daphnia FA composition was independent of that of the seston diet, indicating that Daphnia adjusted their storage lipids according to the physiological requirements of a given overwintering strategy. The occurrence of an active overwintering strategy has consequences for zooplankton community structure, and can have important implications for the transfer of high-quality energy at higher trophic levels.  相似文献

Alkali and alkaline earth aluminoborate glasses feature high resistance to cracking under sharp contact loading compared to other oxide glasses. However, due to the high content of hygroscopic B2O3, it is expected that applications of these glasses could be hindered by poor chemical durability in aqueous solutions. Indeed, the compositional and structural dependence of their dissolution kinetics remains unexplored. In this work, we correlate the dissolution rates of aluminoborate glasses in acidic, neutral, and basic solutions with the structural changes induced by varying the aluminum-to-boron ratio. In detail, we investigate a total of seventeen magnesium, lithium, and sodium aluminoborate glasses with fixed modifier content of 25 mol%. We show that the structural changes induced by alumina depend on the network modifier. We also demonstrate a correlation between the chemical durability at various pH values and the structural changes in Mg-, Li- and Na-aluminoborate glasses. The substitution of alumina by boron oxide leads to a general decrease in chemical corrosion in neutral and acidic solutions. The lowest dissolution rate value is observed in Mg-aluminoborate glasses, as a consequence of the intermediate character of magnesium which can increase the network cross-linking. For basic solutions, the chemical durability is almost constant for the different amount of alumina in the three series, likely because B2O3 is susceptible to nucleophilic attack, which is favored in high-OH solutions.  相似文献

The synthesis of alternating copolymers of tetraalkylindenofluorene with bithiophene and terthiophene using Suzuki polycondensation route is reported. We report on the optical and electrochemical properties of these copolymers. AFM analysis of the microscopic morphology of thin deposits showed that the copolymer with terthiophene units produced the more ordered films, with well-defined fibrillar structures, resulting from highly-regular dense packing due to strong π–π interchain interactions, in contrast to the amorphous bithiophene copolymer. Upon testing these materials in FETs the terthienyl copolymers displayed the higher charge mobilities among the studied compounds, with values of over 10?4 cm2 V?1 s?1 being obtained.  相似文献

Thermal barrier coatings (TBC) are commonly used in modern gas turbines for aeronautic and energy production applications. The conventional methods to fabricate such TBCs are EB-PVD or plasma spray deposition. Recently, the spark plasma sintering (SPS) technique was used to prepare new multilayered coatings. In this study, complete thermal barrier systems were fabricated on single crystal Ni-based superalloy (AM1®) substrate in a one-step SPS process. The lifetime of TBC systems is highly dependent on its ability to form during service a dense, continuous, slow-growing alumina layer (TGO) between an underlying bond coating and a ceramic top coat. In the present paper, we show that such kind of layer (called proto-TGO in the following) can be in situ formed during the SPS fabrication of TBC systems. This proto-TGO is continuous, dense and its nature has been determined using TEM-EDS-SAD and Raman spectroscopy. This amorphous oxide layer in the as-fabricated samples transforms to α-Al2O3 during thermal treatment under laboratory air at 1100 °C. Oxidation kinetics during annealing are in good agreement with the formation of a protective α-Al2O3 layer.  相似文献

This paper presents a method for allocating tolerances to dimensions in kinematic couplings, which are exact constraint devices used to locate one object with respect to another. The objective is to reduce the manufacturing cost without exceeding limits on the variation of the coupled position and orientation. The allocation procedure uses parametric models of the contacting surfaces and a solution for the resting position of the coupled body. A multivariate error analysis provides a relation between variation in manufactured dimensions and variation in the position and orientation of the coupled body. Optimal tolerances are then determined using a non-linear constrained optimization algorithm that minimizes the manufacturing cost while satisfying constraints on the variation of the coupled position and orientation. The method provides a useful tool when designing mass-produced kinematic couplings intended for applications where coupled bodies are exchanged.  相似文献
