Pulp-enriched cloudy apple juices were manufactured from two apple varieties by blending of apple puree with natural cloudy apple juice in order to enrich bioactive secondary plant substances. Finishing of the puree with a 0.6-mm mesh screen and final product homogenisation revealed as the optimum processing technology for the novel 100% fruit beverage. The presence of large particles originating from the puree prevented long-term cloud stability, but due to the proportion of cloud-stable juice a complete phase separation did not occur. Optimal drinkability was achieved at a viscosity of 11.5 mPa s. Polyphenol compositions and concentrations were determined by means of RP-HPLC/PDA. The results ranged from 109 to 610 mg/l. All samples showed the typical polyphenol pattern of apples with dominating hydroxycinnamic acids followed by flavanols and flavonols. The puree addition to the cloudy apple juices increased the polyphenol concentrations by average 100%. Relatively, the highest increase could be observed for dimeric procyanidins. Different technological variants did not affect significantly the polyphenol concentrations. The dietary fibre contents of the pulp-enriched cloudy apple juices ranged from 5.8 to 9.4 g/l.  相似文献

Aerodynamic modelling of HAWT rotors by means of “engineering methods” has reached a saddle point, where no further development can be expected without a breakthrough in understanding the physics of unsteady, rotating three‐dimensional flows. However, such a breakthrough becomes ever more necessary, as the size of the wind turbines increases. With the experimental work in that direction being mostly limited to observing the phenomena and interpreting the associated mechanisms, and its increased cost, alternatives are being sought. The use of CFD techniques and state‐of‐the‐art Navier–Stokes solvers is considered a very serious contender, a belief shared by the members of the present consortium, which has worked on the VISCEL JOR3‐CT98‐0208 Joule III project. This project's goal was to determine the aerodynamic characteristics as well as the aeroelastic behaviour of wind turbine blades across their broad range of operational conditions, from attached to highly separated flow regimes. The work programme included specific tasks for the validation and assessment of existing 3D solvers, for the parametric study of 3D flow around realistic blades and for the investigation of aeroelastic stability, at the blade section level. The U.S. Geological Survey evaluated 20 years of total phosphorus (P) and total nitrogen (N) concentration data for 18 Lake Champlain tributaries using a new statistical method based on weighted regressions to estimate daily concentration and flux histories based on discharge, season, and trend as explanatory variables. The use of all the streamflow discharge values for a given date in the record, in a process called “flow-normalization”, removed the year-to-year variation due to streamflow and generated a smooth time series from which trends were calculated. This approach to data analysis can be of great value to evaluations of the success of restoration efforts because it filters out the large random fluctuations in the flux that are due to the temporal variability in streamflow. Results for the full 20 years of record showed a mixture of upward and downward trends for concentrations and yields of P and N. When the record was broken into two 10-year periods, for many tributaries, the more recent period showed a reversal in N from upward to downward trends and a similar reversal or reduction in magnitude of upward trends for P. Some measures of P and N concentrations and yields appear to be related to intensity of agricultural activities, point-source loads of P, or population density. Total flow-normalized P flux aggregated from the monitored tributaries showed a decrease of 30 metric tons per year from 1991 to 2009, which is about 15% of the targeted reduction established by the operational management plan for the Lake Champlain Basin.  相似文献

Polymeric photovoltaic (PV) solar cells have been fabricated using six solvents: chloroform (CHCl3), toluene (T), chlorobenzene (CB), orthodichlorobenzene (ODCB), 1,2,3,4-tetrahydronaphthalene (THN) and 1,2,4-trichlorobenzene (TCB). The active layers were composed of poly(3-hexyl)thiophene (P3HT) and [6,6]-phenyl C61 butyric acid methyl ester (PCBM). Special care has been taken to keep all experimental parameters constant (thickness of the active layers, donor/acceptor weight ratio, area of active surface and electrodes) in order to avoid artefacts and truly study the effect of solvents. Studies using atomic force microscopy (AFM) and optical absorption (UV-vis) showed the relationship between the photovoltaic performance and the evaporation rate of solvents. The use of solvents with high boiling point results in a higher degree of organization in the structure of P3HT. A direct comparison with devices processed with thermal treatment has also been performed. As often reported thermal annealing increases photo-conversion efficiency of devices created from common solvents, due to better separation of phase between the two materials of the blend. In the case of solvents with high boiling point such as THN and TCB, neither phase separation nor modification of P3HT crystallization induced by thermal annealing has been observed. However thermal treatment appears to enhance performance, ensuing the evaporation of remaining solvent in the active layers. An overview of the effect of solvent on the electrical properties of films containing pure P3HT and P3HT:PCBM blend reported in the literature has been completed for the discussion.  相似文献

The diversity of lactic acid bacteria associated with Hussuwa fermentation, a Sudanese fermented sorghum food, was studied using a polyphasic taxonomical approach. Predominant strains could be well characterised based on a combination of phenotypic tests and genotypic methods such as ARDRA, rep-PCR and RAPD-PCR, as well as 16S rRNA gene sequencing of representative strains. Thus, the majority (128 of 220, 58.3%) of strains exhibited phenotypic properties typical of heterofermentative lactobacilli and of these, 100 strains were characterised more closely using the genotyping methods. The majority (97/100) strains could be characterised as Lactobacillus fermentum strains. Seventy-two of 220 strains (32.7%) showed phenotypic properties that are characteristic of pediococci. Of 41 selected strains investigated by genotyping techniques, 38 (92.7%) could be characterised as Pediococcus acidilactici strains, while three (7.3%) could be characterised as Pediococcus pentosaceus strains. The Hussuwa fermentation thus appears to be dominated by L. fermentum strains and P. acidilactici strains. For this reason, we selected representative and predominant strains as potential starter cultures for Hussuwa fermentation. These strains, L. fermentum strains BFE 2442 and BFE 2282 and P. acidilactici strain BFE 2300, were shown on the basis of RAPD-PCR fingerprinting to predominate in a model fermentation when used as starter cultures inoculated at 1 × 106 CFU/g and to lower the pH of the fermentation to below pH 4.0 within 48 h. These cultures should be studied for further development as starter preparations in pilot scale studies in actual field fermentations.  相似文献

Photovoltaic (PV) water pumping systems have been widely tested and assessed in Indonesia because of the deficiency of potable water in remote areas. Several different PV water pumping systems, such as a.c./d.c. with/without battery and various pumps, have been tested in the field. Further study, evaluation and classification of optimized PV water pumping systems is needed. Three of those pumping systems were installed in Sumba Island in 1983, in three adjacent villages with similar climate and environment conditions. Two of the installations had a.c. direct coupled systems and the other had d.c. The a.c. systems used a 3.65 kWp PV generator each and the d.c. system used 5.76 kWp. After comparing the performance data of both systems, it can be concluded that the a.c. system performs better and needs less maintenance on the pump. In May 1987, as a result of this evaluation, the 5.76 kWp d.c. system was replaced by an a.c. one, and its capacity was increased to 9.22 kWp. Meanwhile, the 3.65 kWp a.c. system was increased to 3.88 kWp.  相似文献

A gel filtration technique afforded a good separation between metal complexed with bacterial extracellular polymers and free metal ions. The complexation of polymers extracted from cultures of Klehsiella aerogenes and activated sludge with cadmium, nickel, manganese and cobalt was demonstrated. The extraction of extracellular polymers from cultures of K. aerogenes and activated sludge reduced the capacity of the cells and flocs to adsorb metal. Adsorption and complexation of metals by cells of K. aerogenes and extracellular polymers extracted from activated sludge were fitted to Freundlich equilibrium isotherms. Saturation of activated sludge polymer binding sites occurred at 10 mg 1?1 metal additions for all the metals studied except manganese which was complexed to a very limited extent. Cells of K. aerogenes exhibited no saturation effects in the range of metal concentrations studied.Precipitation of metals below a concentration of 1 mg 1?1 was minimal, with the exception of cadmium precipitation. At a concentration of 10 mg 1?1, precipitation of cadmium, cobalt and manganese may have been the major mechanism of metal removal. The more soluble metals generally displayed the lowest removals. Concentrations of extracellular polymers and soluble chelating agents may be important in controlling removals of metals which are largely soluble in activated sludge.  相似文献

Based on their predominance in Gari fermentations, as well as suitable technological properties, Lactobacillus plantarum, Lactobacillus fermentum, Weissella paramesenteroides and Leuconostoc mesenteroides strains were investigated for their suitability for development as starter strains for this African traditional fermented cassava product. The strains were grown in optimized growth media in 2 L fermenters, harvested and freeze dried, and then tested in lab-scale cassava mash fermentation trials for their ability to ferment the cassava. The strains performed well and rapidly increased the titratable acidity from 1.1 to 1.3% at 24 h to 1.3–1.6% at 48 h. The benefit of including starter cultures was that it lowered the pH of the product much faster and to lower levels than in the uninoculated control fermentation. The results furthermore indicated that especially the L. plantarum-group strains could be produced as starter strains at low cost. Overall, the results of this study showed that starter strains could be easily and economically produced, and thus represent a feasible possibility for further development for application in the field.  相似文献

A liquid chromatography–tandem mass spectrometry (LC–MS/MS) method was developed and validated for the simultaneous determination of six major ergot alkaloids, ergometrine, ergosine, ergotamine, ergocornine, ergokryptine and ergocristine, as well as their corresponding epimers in food and feed samples. The method involves extraction under alkaline conditions and subsequent clean-up by applying a simple and rapid liquid–liquid partitioning procedure prior to LC–MS/MS analysis. Evaluation of the method revealed good linearity, accuracy and precision. The limits of quantification varied from 0.1 to 1 μg/kg depending on the analyte and matrix. The average extraction and clean-up recoveries in different matrices were between 45 (only for ergometrine in biscuit) and 90%. The uncertainty associated with the analytical method was not higher than 51% and 30%, at concentration levels of 2.5 and 150 μg/kg respectively. Analyte epimerization proved to be minimal during the analytical procedure. The method has been successfully applied to the determination of ergot alkaloids in some Belgian food and feed commodities. Ergot alkaloids were found in 104 out of 122 samples investigated. Ergosine was the most frequently occurring alkaloid, while the highest levels were observed for ergotamine, ergocristine or ergosine, depending on the product type. The pregnancy-specific glycoproteins (Psg) are secreted hormones encoded by multiple genes in rodents and primates, and are thought to act as immune modulators. The only Psg receptor identified is CD9, through which Psg17 induces cytokine production from macrophages cultured in vitro. We examined temporal and spatial aspects of Psg and CD9 expression during mouse pregnancy to determine whether their expression patterns support a role in immune modulation. Using in situ hybridisation, immunohistochemistry and RT-PCR we found Psg expression in trophoblast giant cells and in the spongiotrophoblast. Psg22 is the predominant Psg family member expressed in giant cells. Detectable Psg is associated predominantly with endothelial cells lining vascular channels in the decidua, rather than with maternal immune cell markers. CD9 expression exhibited partial overlap with Psg, but without exclusive co-localisation. CD9 was observed in decidual cells surrounding early implantation sites, and in the endometrium. However, embryo transfer of wild-type embryos to CD9-deficient females indicates that maternal CD9 is not essential for successful pregnancy.  相似文献
