Protein design to understand peptide ligand recognition by tetratricopeptide repeat proteins

Protein design aims to understand the fundamentals of protein structure by creating novel proteins with pre-specified folds. An equally important goal is to understand protein function by creating novel proteins with pre-specified activities. Here we describe the design and characterization of a tetratricopeptide (TPR) protein, which binds to the C-terminal peptide of the eukaryotic chaperone Hsp90. The design emphasizes the importance of both direct, short-range protein-peptide interactions and of long-range electrostatic optimization. We demonstrate that the designed protein binds specifically to the desired peptide and discriminates between it and the similar C-terminal peptide of Hsp70.     

Hydrogenation of 3,4-epoxy-1-butene over Cu–Pd/SiO2 catalysts prepared by electroless deposition

Electroless deposition has been used to prepare Cu–Pd/SiO₂ bimetallic catalysts wherein initial Cu coverages are limited only to the pre-existing Pd surface. Cu loading on the Pd surface can be systematically varied by modification of deposition kinetic parameters. In this case deposition time was used as the kinetic variable for the preparation of a series of Cu–Pd catalysts. These materials have been characterized using atomic absorption, CO chemisorption, and FT-IR (adsorption of CO), and then evaluated for the hydrogenation of 3,4-epoxy-1-butene, a functionalized olefin having many potential reaction pathways. Catalyst performance and characterization results suggest that Cu is not distributed in a monodisperse manner on the Pd surface, indicating the existence of autocatalytic deposition of Cu on Cu sites. The FT-IR results suggest that although CO adsorption on all sites is suppressed by Cu addition, initial Cu deposition occurs more readily on certain sites. The bimetallic Cu–Pd sites that are formed exhibit unusually high activity for EpB conversion and formation of unsaturated alcohols and aldehydes. This bimetallic effect on catalyst activity and selectivity is best explained, not by the existence of either ligand or ensemble effects, but rather by the bifunctional nature of the Cu–Pd sites present on the surface of these catalysts.     

Mass transfer in aerated fermentation broths in a stirred tank reactor

The volumetric mass transfer coefficients, k_La, measured in a pilot plant (0.1 m³) and an industrial (67.5 m³) fermentor during an actual fermentation process are presented. Problems related to the estimation of the phsyical properties as well as to the correlation of experimental data and to scale up procedures are discussed. Although the scale up factor was rather high, both sets of data could be represented by single correlation. Comparison of the experimental data with several available correlations demonstrated the need for pilot plant experiments and scale up procedures, since it is almost impossible to take into account all relevant system properties.     

The mechanism of CO oxidation over Cu(110): Effect of CO gas energy

The effect of the temperature of gas phase CO upon the kinetics of the oxygen titration reaction: CO_g +O_a CO_2,g, has been studied. It is found that the reaction's rate is independant of CO gas temperature between 300 and 623 K. The activation energy (6.5 kcal/mole), dependence upon CO pressure (first-order), and independence upon oxygen coverage for 0.1 _o 0.4 are all independant of the CO gas phase temperature. This result rules out any Eley-Rideal type mechanism whereby CO reacts directly from the gas phase with an oxygen adatom without first being accommodated to the surface temperature in an absorbed state. Creating Disk Images of Born Digital Content: A Case Study Comparing Success Rates of Institutional Versus Private Collections

Although libraries maintain collections of material on computer disks in climate controlled environments, a significant amount of digital heritage is in private collections. In this study we compared the outcomes of creating disk images of the State Library of South Australia’s born digital collections with those of a private collector to gauge the effect of long term storage conditions. The failure rates of both collections give cause for concern, and the results from the private collection are alarming.

Das Projekt ?E-Mail made in Germany“ wurde als Reaktion auf die Enthüllungen von Edward Snowden initiiert. Es ist das erklärte Ziel, den E-Mail-Nutzern in Deutschland einen hohen Sicherheits- und Datenschutzstandard anzubieten. Das Produkt wurde mit einer breit angelegten Werbekampagne beworben. Im Mittelpunkt der Kampagne stand ein 30sekündiger TV-Spot, der auf die Notwendigkeit von sicherer E-Mail-Kommunikation eingeht. Kritiker bemängeln jedoch, dass dabei ein falsches Sicherheitsverständnis von ?E-Mail made in Germany“ vermittelt wird. Dieser Kritik wurde mit einer Laborstudie nachgegangen.     

Fostering Library as a Place for Distance Students: Best Practices From Two Universities

The concept of library as a place is now in action in many academic libraries: from physical library spaces to Web presences. In the constantly growing market of online and distance learning, however, libraries must foster library as a place for students who may never set foot in the physical building. This article provides a profile of two universities serving two very different bodies of distance students as well as the challenges associated with these populations. These libraries work to provide services to distance students that cultivate the library as a place to go for learning, research, and support by focusing on six categories: access, environment, resources, instruction, availability at the point of need, and “being real.” Tangible examples and best practices are provided throughout. This article is based on a presentation given at the 15th National Conference of the Association of College and Research Libraries in Philadelphia in March 2011.     

Single machine batch scheduling with release times and delivery costs

We study single machine batch scheduling with release times. Our goal is to minimize the sum of weighted flow times (or completion times) and delivery costs. Since the problem is strongly $\mathcal{NP}$ -hard even with no delivery cost and identical weights for all jobs, an approximation algorithm is presented for the problem with identical weights. This uses the polynomial time solution we give for the preemptive version of the problem. We also present an evolutionary metaheuristic algorithm for the general case. Computational results show very small gaps between the results of the metaheuristic and the lower bound.     

Computing all efficient solutions of the biobjective minimum spanning tree problem

A common way of computing all efficient (Pareto optimal) solutions for a biobjective combinatorial optimisation problem is to compute first the extreme efficient solutions and then the remaining, non-extreme solutions. The second phase, the computation of non-extreme solutions, can be based on a “k-best” algorithm for the single-objective version of the problem or on the branch-and-bound method. A k-best algorithm computes the k-best solutions in order of their objective values. We compare the performance of these two approaches applied to the biobjective minimum spanning tree problem. Our extensive computational experiments indicate the overwhelming superiority of the k-best approach. We propose heuristic enhancements to this approach which further improve its performance.
