Health effects of tritium, a β-emitter and a by-product of the nuclear industry, is a subject of significant controversy. This mouse in vivo study was undertaken to monitor biological effects of low level tritium exposure. Mice were exposed to tritiated drinking water (HTO) at 10 KBq/L, 1 MBq/L and 20 MBq/L concentrations for one month. The treatment did not result in a significant increase of apoptosis in splenocytes. To examine if this low level tritium exposure alters radiosensitivity, the extracted splenocytes were challenged in vitro with 2 Gy γ-radiation, and apoptotic responses at 1 and 24 h were measured. No alterations in the radiosensitivity were detected in cells from mice exposed to tritium compared to sham-treated mice. In contrast, low dose γ-irradiation at 20 or 100 mGy, resulted in a significant increase in resistance to apoptotic cell death after 2 Gy irradiation; an indication of the radioadaptive response. Overall, our data suggest that low concentrations of tritium given to mice as HTO in drinking water do not exert cytotoxic effect in splenocytes, nor do they change cellular sensitivity to additional high dose γ-radiation. The latter may be considered as the lack of a radioadaptive response, typically observed after low dose γ-irradiation. 相似文献

Water-based coating formulations contain many components, such as latex binder, pigment, dispersant, thickener, and surfactant. Complex interactions between these components affect properties of the coating in both the wet and dried states. For example, rheology of the paint is dependent on the interactions of components in the formulation. Pigment interaction can affect the degree of dispersion and, therefore, have ramifications on end use properties such as hiding and tint strength of the paint film. Isothermal titration calorimetry has been used to determine the enthalpy of interaction of the coating components. Representative examples will be given to demonstrate structure–property relationships that correlate the interactions of latex, pigment, surfactant, dispersant, and thickener with paint viscosity as well as end use performance parameters such as adhesion and tint strength. 相似文献

The effects of the polyunsaturated omega-3 (n-3) and omega-6 (n-6) fatty acids (FA) on hematopoiesis are complex in that both
FA forms are processed into leukotrienes, eicosanoids, and prostaglandins, which can have independent effects. These FA have

Structural preorganization to fix bioactive conformations at protein binding sites is a popular strategy to enhance binding affinity during late‐stage optimization. The rationale for this enhancement relates to entropic advantages assigned to rigidified versus flexible ligands. We analyzed a narrow series of peptidomimetics binding to thrombin. The individual ligands exhibit at P2 a conformationally flexible glycine, more restricted alanine, N‐methylglycine, N‐methylhomoalanine, and largely rigidified proline moiety. Overall, affinity was found to increase by a factor of 1000, explained partly by an entropic advantage. All ligands adopt the same binding mode with small deviations. The residual mobility of the bound ligands is decreased across the series, and a protein side chain differs in its order/disorder behavior along with changes in the surface‐water network pattern established across the newly generated protein–ligand surfaces. The enthalpy/entropy inventory displays a rather complex picture and emphasizes that thermodynamics can only be compared in terms of relative differences within a structurally similar ligand series. 相似文献

NovaSil (NS) clay, a common anti-caking agent in animal feeds, has been shown to sorb aflatoxins in the GI tract and diminish their bioavailability and adverse effects in short-term animal studies. Based on this evidence, it is hypothesized that clay-based enterosorption of aflatoxins may be a useful strategy for the prevention of aflatoxicosis in human populations. However, the potential toxicity of long-term dietary exposure to NS has not been determined. In this research, 5-6-week-old male and female Sprague-Dawley rats were fed rations containing 0, 0.25, 0.5, 1.0, or 2.0% (w/w) levels of NS for 28 weeks. Analysis of the NS showed negligible levels of dioxin and furan contaminants. Total feed consumption, cumulative feed consumption, body weight, total body weight gain, feed conversion efficiency, cumulative feed conversion efficiency, and relative organ weights were unaffected in either sex at the doses tested. No NS-dependent differences in relative organ weights or gross or histopathological changes were observed. Analysis of hematological parameters, clinical chemistry, and selected vitamin and mineral levels revealed isolated significant differences between some treatments and control groups (mean corpuscular hemoglobin, serum Ca, serum vitamin A, and serum Fe). However, the differences observed in each case were not dose-dependent. These results suggest that dietary inclusion of NS at levels as high as 2.0% (w/w) does not result in overt toxicity. These findings (as well as others) support the use of NS clay for dietary intervention studies in human populations at high risk for aflatoxicosis. 相似文献

There is a lack of information on the effect of swine caecal microbiota on fumonisin metabolism. In this in vitro study, the biotransformation of fumonisin B1 (FB1) by the gut microbiota of adult, healthy pigs was examined. Suspensions of caecal contents and McDougall buffer solution were incubated anaerobically with pure FB1 for 0, 12, 24, 48 and 72 h. After 48 h, the conversion of FB1 to partially hydrolysed FB1 (46%) was nearly equal to the percentage ratio of FB1, while by 72 h it was 49%. In vitro, the conversion of fumonisin B1 to aminopentol was less than 1%. The results show that the caecal microbiota are capable of transforming fumonisin B1 to the above metabolites. Further studies on FB1 metabolism in the small intestine are clearly justified. 相似文献

This article examines the negative suggestion effect, or the impact of exposure to incorrect alternatives on memory for correct information. All experiments used the following design: (a) cued-recall test of general facts (e.g., "second smallest planet") with immediate correct feedback, (b) interpolated exposure to incorrect information related to Test 1 items, and (c) a second test over the same items as Test 1. Test 2 was either multiple choice or cued recall and was given either immediately or 1 week after interpolation. Three experiments confirmed the existence of negative suggestion: Exposure to misinformation hindered subsequent performance on those items, relative to noninterpolated control items. The magnitude of this decrement is unrelated to retention interval, type of second test, number of incorrect alternatives exposed, and number of repetitions of incorrect alternatives. A pilot-scale test rig was designed and constructed to simulate intrusion behavior associated with hydraulic transients initiated by sudden events such as rapid valve closure or uncontrolled change in on/off pump status (“pump trip”) in a water distribution system. After establishing steady state flow conditions, intrusion volumes were determined for 3.2 (1/8-in.) and 6.4 mm (1/4-in.) diam orifices overlaid with a column of water that provided 91 mm (3 ft) of external head. Average intrusion volumes associated with hydraulic transient events were determined by mass balance calculations using cesium as the tracer chemical. Average intrusion volumes were 11.4 and 71.2 mL for the two diameters, respectively. Given similar conditions in a water distribution system (i.e., an available pathway and favorable pressures), pathogens in the soil and water surrounding a water main potentially can intrude into the pipe during short-term pressure transient events. 相似文献
