Cancer-associated fibroblasts (CAFs) are one of the most abundant and critical components of the tumor stroma. CAFs can impact many important steps of cancerogenesis and may also influence treatment resistance. Some of these effects need the direct contact of CAFs and cancer cells, while some involve paracrine signals. In this study, we investigated the ability of head and neck squamous cell carcinomas (HNSCC) patient-derived CAFs to promote or inhibit the colony-forming ability of HNSCC cells. The effect of cisplatin on this promoting or inhibiting influence was also studied. The subsequent analysis focused on changes in the expression of genes associated with cancer progression. We found that cisplatin response in model HNSCC cancer cells was modified by coculture with CAFs, was CAF-specific, and different patient-derived CAFs had a different “sensitizing ratio”. Increased expression of VEGFA, PGE2S, COX2, EGFR, and NANOG in cancer cells was characteristic for the increase of resistance. On the other hand, CCL2 expression was associated with sensitizing effect. Significantly higher amounts of cisplatin were found in CAFs derived from patients who subsequently experienced a recurrence. In conclusion, our results showed that CAFs could promote and/or inhibit colony-forming capability and cisplatin resistance in HNSCC cells via paracrine effects and subsequent changes in gene expression of cancer-associated genes in cancer cells. 相似文献

Leptin is an adipokine that regulates appetite and body mass and has many other pleiotropic functions, including regulating kidney function. Increased evidence shows that chronic kidney disease (CKD) is associated with hyperleptinemia, but the reasons for this phenomenon are not fully understood. In this review, we focused on potential causes of hyperleptinemia in patients with CKD and the effects of elevated serum leptin levels on patient kidney function and cardiovascular risk. The available data indicate that the increased concentration of leptin in the blood of CKD patients may result from both decreased leptin elimination from the circulation by the kidneys (due to renal dysfunction) and increased leptin production by the adipose tissue. The overproduction of leptin by the adipose tissue could result from: (a) hyperinsulinemia; (b) chronic inflammation; and (c) significant lipid disturbances in CKD patients. Small unmanned aerial vehicles (UAV) are typically driven by Lithium polymer batteries. The batteries have their own dynamics, which changes during discharge. Classical approaches to altitude control assume a time-invariant system and therefore fail. Adaptive controllers require an identified system model which is often unavailable. Battery dynamics can be characterized and used for a battery model-based controller. This controller is useful in situations when no feedback from actuators (such as RPM or thrust) is available. After measuring the battery dynamics for two distinct types of batteries, a controller is designed and experimentally verified, showing a consistent performance during entire discharge test and a consequent flight verification. 相似文献

Summary The dynamic mechanical behaviour of 4% aqueous solutions and networks of poly (N, N-diethylacrylamide) and copolymers of diethylacrylamide

(T > Tc) the mechanical behaviour of solutions has a liquid-like character (the loss modulus G″ is higher than the storage modulus
G′ for a constant frequency ω=1 Hz), in the region of globular conformations (T > Tc) a heterogeneous physical network is built in solutions, and the mechanical behaviour has a solid-like character (G′ > G″).
In networks the collapse is reflected in an increase of storage modulus G′; the magnitude of this increase decreases with
ionization. The dependence of the loss modulus G″ on the temperature of solutions and networks allows us to conclude that

Liquid crystalline (LC) polyurethanes were made from two diisocyanates (flexible HMDI and stiff TDI) (DIs), mesogenic diol (D) and a polybutadiene-diol (B) with stoichiometric ratios of reactive hydroxy (OH) and isocyanate (NCO) groups ((NCO)DI/((OH)D+(OH)B)=1/1). Two- (B/DIs, D/DIs) and three-component ((D+B)/DIs, D/B=1/1 by weight) polymers were prepared and their dielectric, dynamic mechanical and DSC behavior was investigated. For neat B, the glass transition temperature TgB (∼−46 °C) was detected. Two-component B/HMDI and B/TDI polymers have exhibited a homogeneous structure with the glass transition temperatures TgU∼−9 and 2 °C. On the other hand, for D/DI polymers on cooling from the melt and subsequent heating the glass transitions at TgU∼41 °C (D/HMDI) and 58 °C (D/TDI) together with nematic and smectic mesophases were found. In three-component systems, additional glass transitions at TgB∼−41 °C (B/D/HMDI) and −31 °C (B/D/TDI) were observed. This means that the polymers exhibit a distinct two-phase structure with soft polybutadiene (B) and hard polyurethane (D/DI) phases. In hard polyurethane phase, the glass transitions at TgU and LC mesophases similar to those found in two-component D/DI polyurethanes were detected. This article describes the synthesis and in vitro analysis of poly(ester anhydride) antimicrobial protection coatings. Poly(ester anhydride)s composed of ricinoleic acid, sebacic acid, terephthalic acid, and isophthalic acid were used in this study. The polymers were compatible with various fillers commonly used in paint preparation. The composition of theAutographa gamma sex pheromone was reexamined and only (Z)-7-dodecenyl acetate and (Z)-7-dodecenol were identified by capillary GC, GC-MS, and dimethyl disulfide derivatization and subsequent GC-MS analysis. The fatty acid content of the pheromone glands was also studied, and a series of saturated and unsaturated acids was identified. However, most of the related pheromonal compounds were not detected. The male response to the pheromone components was studied in a flight tunnel and compared with the response to calling females. The best synthetic baits evoked a response similar to that observed to the virgin females, but males spent significantly more time at calling females than at the synthetic baits. The preferred synthetic baits consisted of (Z)-7-dodecenyl acetate alone or of a blend with 5% (Z)-7-dodecenol. Increasing the relative amount of the alcohol caused a gradual reduction in male response, particularly in the last steps of the courtship sequence. The addition of the minor sex pheromone components of the sympatric Plusiinae species,Trichoplusia ni andChrysodeixis chalcites, to theA. gamma pheromone was also investigated in the flight tunnel. Some of these components exhibited a significantly antagonistic effect on theA. gamma male courtship behavior. The most potent antagonists were (Z)-5-dodecenyl acetate and (Z)-9-tetradecenyl acetate. The response ofA. gamma andT. ni males to conspecific and heterospecific females was also compared in the flight tunnel. Unidirectionally reinforced graphite/copper composites have been fabricated using a pressure infiltration casting procedure.
T300 and T650 graphite fibers have been used to reinforce copper and copperchromium alloys. The effects of the chromium level
in the copper matrix on the tensile strength, stiffness, and thermal expansion behavior of the composites have been evaluated
through tensile and three-point bend testing, and thermal cycling. At the 0.5 wt% alloying level, chromium increases the stiffness
and optimizes the thermal expansion behavior of graphite/copper composites. The longitudinal tensile strengths of these composites
are above 1606 MPa, whereas the transverse tensile strengths are lower than 40 MPa due to incomplete infiltration during processing.
Scanning electron microscopy analyses reveal that the unalloyed copper matrix composites experienced extensive fiber/matrix
debonding under tensile loading. The addition of chromium to the copper increases the level of matrix bonding to the graphite
fibers, as evidenced by observations of fractured tensile specimens. Auger electron spectroscopy analyses indicate that a
chromium carbide phase present at the interface is responsible for the improved bonding. 相似文献

Numerical weather prediction (NWP) is in a period of transition. As resolutions increase, global models are moving towards fully nonhydrostatic dynamical cores, with the local and global models using the same governing equations; therefore we have reached a point where it will be necessary to use a single model for both applications. The new dynamical cores at the heart of these unified models are designed to scale efficiently on clusters with hundreds of thousands or even millions of CPU cores and GPUs. Operational and research NWP codes currently use a wide range of numerical methods: finite differences, spectral transform, finite volumes and, increasingly, finite/spectral elements and discontinuous Galerkin, which constitute element-based Galerkin (EBG) methods. Due to their important role in this transition, will EBGs be the dominant power behind NWP in the next 10 years, or will they just be one of many methods to choose from? One decade after the review of numerical methods for atmospheric modeling by Steppeler et al. (Meteorol Atmos Phys 82:287–301, 2003), this review discusses EBG methods as a viable numerical approach for the next-generation NWP models. One well-known weakness of EBG methods is the generation of unphysical oscillations in advection-dominated flows; special attention is hence devoted to dissipation-based stabilization methods. Since EBGs are geometrically flexible and allow both conforming and non-conforming meshes, as well as grid adaptivity, this review is concluded with a short overview of how mesh generation and dynamic mesh refinement are becoming as important for atmospheric modeling as they have been for engineering applications for many years. 相似文献
