Once merely thought of as the protein responsible for the overall physical nature of the human immunodeficiency virus type 1 (HIV-1), the Gag polyprotein has since been elucidated to have several roles in viral replication and functionality. Over the years, extensive research into the polyproteins’ structure has revealed that Gag can mediate its own trafficking to the plasma membrane, it can interact with several host factors and can even aid in viral genome packaging. Not surprisingly, Gag has also been associated with HIV-1 drug resistance and even treatment failure. Therefore, this review provides an extensive overview of the structural and functional roles of the HIV-1 Gag domains in virion integrity, functionality and infectivity.  相似文献

Chemoradiation-induced mucositis is a debilitating condition of the gastrointestinal tract eventuating from antineoplastic treatment. It is believed to occur primarily due to oxidative stress mechanisms, which generate Reactive Oxygen Species (ROS). The aim of this scoping review was to assess the role of oxidative stress in the development of Oral Mucositis (OM). Studies from the literature, published in MEDLINE and SCOPUS, that evaluated the oxidative stress pathways or antioxidant interventions for OM, were retrieved to elucidate the current understanding of their relationship. Studies failing inclusion criteria were excluded, and those suitable underwent data extraction, using a predefined data extraction table. Eighty-nine articles fulfilled criteria, and these were sub-stratified into models of study (in vitro, in vivo, or clinical) for evaluation. Thirty-five clinical studies evaluated antioxidant interventions on OM’s severity, duration, and pain, amongst other attributes. A number of clinical studies sought to elucidate the protective or therapeutic effects of compounds that had been pre-determined to have antioxidant properties, without directly assessing oxidative stress parameters (these were deemed “indirect evidence”). Forty-seven in vivo studies assessed the capacity of various compounds to prevent OM. Findings were mostly consistent, reporting reduced OM severity associated with a reduction in ROS, malondialdehyde (MDA), myeloperoxidase (MPO), but higher glutathione (GSH) and superoxide dismutase (SOD) activity or expression. Twenty-one in vitro studies assessed potential OM therapeutic interventions. The majority demonstrated successful a reduction in ROS, and in select studies, secondary molecules were assessed to identify the mechanism. In summary, this review highlighted numerous oxidative stress pathways involved in OM pathogenesis, which may inform the development of novel therapeutic targets.  相似文献

Sydnone Ethylenes. I Stereoselective Synthesis of Sydnone Ethylenes from Sydnone Aldehydes Sydnones are formylated in 4-position by the VILSMEIER -HAAK reaction. N(3)-Methyl- and N(3)-(p-Anisyl)-sydnone-(4)-carboxaldehyde react stereoselectively to sydnone ethylenes with diethyl ethoxycarbonylmethanephosphonate by HORNER reaction and with arylmethylphosphonium salts by WITTIG reaction in the presence of strong bases. In the latter case, especially in the absence of lithium cations, Z-selectivity is observed which is explained by electronic interaction between the sydnone cyclus and the aromatic substituent of the phosphine ylid in the first step of the reaction. The configuration of the obtained Z- and E-sydnone ethylenes is determined by means of 1H-NMR- and IR-spectra.  相似文献

Despite the fact that plants are not traditionally considered as hosts for human enteric pathogens, recent evidence suggests that non-typhoidal Salmonella and enterovirulent Escherichia coli recognize plants and rely on a specific set of genes to multiply in plant-associated environments, eventually causing dramatic outbreaks of illness. The advent of sensitive functional genomics tools, including differential fluorescence induction and in vivo expression technology, set the stage for the characterization of the genes and behaviors used by enterics to colonize, persist and proliferate within plants and the associated microbial communities. Meta-analysis of published data suggests that Salmonella and enterovirulent E. coli persist in plants using strategies that differ from those in phytobacteria. Virulence genes were upregulated in E. coli O157:H7 in the presence of lettuce leaf exudates, however Salmonella SPI-1 genes associated with gastroenteritis were dispensable during plant growth. Microarray and mutant studies of plant responses to human pathogens reveal that plants generally do not recognize Salmonella and enterovirulent E. coli as phytopathogens or beneficial symbionts, although the full spectrum of plant responses to enterics remains to be elucidated. Defining plant responses to human enteric pathogens becomes increasingly important as the feasibility of breeding for resistance to these organisms is being evaluated.  相似文献

Nanohydroxyapatite (nHAp) powders were produced via aqueous precipitation by adopting four different experimental conditions, assisted or non-assisted by ultrasound irradiation (UI). The nHAp powders were characterized by X-ray diffraction, energy-dispersive X-ray fluorescence, Raman and attenuated total reflection Fourier transform infrared spectroscopies, which showed typical surface chemical compositions of nHAp. Analysis found strong connections between UI and the crystallization process, crystal growth properties, as well as correlations between calcination and substitution reactions.  相似文献

The problem of extracting anatomical structures from medical images is both very important and difficult. In this paper we are motivated by a new paradigm in medical image segmentation, termed Citizen Science, which involves a volunteer effort from multiple, possibly non-expert, human participants. These contributors observe 2D images and generate their estimates of anatomical boundaries in the form of planar closed curves. The challenge, of course, is to combine these different estimates in a coherent fashion and to develop an overall estimate of the underlying structure. Treating these curves as random samples, we use statistical shape theory to generate joint inferences and analyze this data generated by the citizen scientists. The specific goals in this analysis are: (1) to find a robust estimate of the representative curve that provides an overall segmentation, (2) to quantify the level of agreement between segmentations, both globally (full contours) and locally (parts of contours), and (3) to automatically detect outliers and help reduce their influence in the estimation. We demonstrate these ideas using a number of artificial examples and real applications in medical imaging, and summarize their potential use in future scenarios.  相似文献

Gas sensor devices have traditionally comprised thin films of metal oxides, with tin oxide, zinc oxide and indium oxide being some of the most common materials employed. With the recent discovery of novel metal oxide nanostructures, sensors comprising nano-arrays or single nanostructures have shown improved performance over the thin films. The improved response of the nanostructures to different gases has been primarily attributed to the highly single crystalline surfaces as well as large surface area of the nanostructures. In this paper the properties of clean and defected quasi one-dimensional ZnO nanostructures, including hexagonal and triangular nanowires, nanotubes and facetted nanotubes are reviewed. The adsorption of atoms and molecules on the ZnO nanostructures are also reviewed and the findings are compared to studies examining similar reactions on nanostructured metal oxide surfaces for sensing purposes. While both experimental and theoretical approaches have been employed to examine gas sensor reactions, this review focuses on studies that employ electronic structure calculations, which primarily concentrate on using density functional theory. Computational studies have been useful in elucidating the reaction mechanism, binding strength, charge transfer as well as other electronic and structural properties of the nanomaterials and the gas-sensor interaction. Despite these studies there are still significant areas of research that need to be pursued that will assist in the link between theoretical and experimental findings, as well as advancing the current chemical and physical understanding of these novel materials. A summary and outlook for future directions of this exciting area of research is also provided.  相似文献

Abstract: Scanning laser vibrometry is a widely used tool to observe Lamb wave fields for structural health monitoring (SHM) purposes. Lamb waves propagate over long distances in thin‐walled structures and interact with structural inhomogeneities, for example, damages, in spite of wavelengths several times of the damage size. In SHM of sheets and glass‐ or carbon‐fibre‐reinforced plastic plates, this effect is used for determining the position as well as the size of structural faults. With the often employed one‐dimensional vibrometry, a geometrically induced, systematic error occurs when measuring oblique‐angled motion. This error can be, in the specific case of Lamb waves, of a non‐negligible quantity. The nature of this geometrical measurement error in general and concerning Lamb waves in special is discussed analytically for both amplitude and phase data. It is shown that this matter should be taken into account in some applications.  相似文献

The purposes of this study were to (1) explore significant differences in perception of match for a variety of food and wine styles; (2) evaluate the impacts of wine sweetness level, wine acidity level, and wine tannin level on perception of match; and (3) investigate the impact of food and wine expertise on perceived level of match. The highest perceived wine matches for each food item were: Sauvignon Blanc and chêvre, Chardonnay and brie, Cabernet Sauvignon and spicy Italian salami, and Port and milk chocolate. Wine sweetness, acidity, and tannin levels all significantly impacted the level of match with certain food items. Food and wine expertise also significantly impacted the level of match, indicating differences between the more expert and novice participants in the role wine sweetness, acidity, and tannin had on level of match.  相似文献
