Identification of HLA-A24-restricted epitopes with high affinities to Hsp70 using peptide arrays

Heat shock protein 70 (Hsp70) family members are known as facilitators of immune responses by interacting with receptors on antigen-presenting cells leading to Hsp70-peptide uptake and antigen cross priming. Here, identification of human leukocyte antigen (HLA)-A24-restricted epitopes was achieved using peptide arrays for evaluation of their affinities to Hsp70 and HLA-A24 binding prediction tools. Using Hsp70 as the model antigen, the GYPVTNAVI and VFQHGKVEI peptides were identified as antigens. These peptides actually bound to HLA-A24 in the stabilization assay using T2-A*2402 cells, and induced a strong peptide-reactive cytotoxic T lymphocyte response in HLA-A24 transgenic mice after vaccination.     

Efficient Synthesis of α-Monoglycerides via Solventless Condensation of Fatty Acids with Glycerol Carbonate

Highly pure α-monoglycerides (5a–e) were successfully prepared in high yields by the condensation of fatty acids such as lauric, myristic, palmitic, stearic and oleic (2a–e) with glycerol carbonate (4-hydroxymethyl-1,3-dioxolan-2-one) (1) in the presence of triethylamine as catalyst. Reaction conditions and spectroscopic identification of products will be presented in this article.     

Detailed Microphysical Modeling of the Formation of Organic and Sulfuric Acid Coatings on Aircraft Emitted Soot Particles in the Near Field

The development of a detailed microphysical model that describes the complex multicomponent interactions between organic vapors and soot particles emitted from aircraft gas turbine engines is presented. Our model formulation includes both soot surface activation by organic vapors and organic vapor condensation on the activated part of the soot surfaces. To enable this formulation, approaches to estimate chemical and physical properties of aerosols containing complex mixtures of sulfuric acid, water, and organic molecules were developed. Relevant distributions of a list of organic surrogates at the engine exit plane were used to represent complex organic emissions from aircraft engines. A parametric study was performed using this new formulation to understand the effects of ambient conditions, organic emissions levels, and mass accommodation coefficient values on the evolution of near field volatile particulate matter emissions from aircraft engines at ground level.

Synthesis of Bio-Based Polyamide/Acid-Functionalized Multiwalled Carbon Nanotube Nanocomposites Using Vanillin

Synthesis of bio-based polyamide/acid-functionalized multiwalled carbon nanotube nanocomposites (PA/FCNT NCs) is reported in this investigation. New aliphatic–aromatic bio-based polyamide (PA) was synthesized through direct polycondensation reaction between bio-based diacid derived from a renewable resource; vanillin and diamine containing ether linkages. To obtain a homogeneous dispersion of multiwalled carbon nanotubes (MWCNTs) in the PA matrix, acid-functionalized MWCNTs (FCNTs) were used and PA nanocomposites with three different FCNT contents (1, 5 and 7?wt%) were prepared. The resulting NCs were characterized by Fourier transform infrared spectroscopy (FT-IR), X-ray diffraction (XRD), field emission scanning electron microscopy (FE-SEM), and thermogravimetric analysis (TGA).     

Multifaceted Analysis of IL-23A- and/or EBI3-Including Cytokines Produced by Psoriatic Keratinocytes

Interleukin (IL) 23 (p19/p40) plays a critical role in the pathogenesis of psoriasis and is upregulated in psoriasis skin lesions. In clinical practice, anti-IL-23Ap19 antibodies are highly effective against psoriasis. IL-39 (p19/ Epstein-Barr virus-induced (EBI) 3), a newly discovered cytokine in 2015, shares the p19 subunit with IL-23. Anti-IL-23Ap19 antibodies may bind to IL-39; also, the cytokine may contribute to the pathogenesis of psoriasis. To investigate IL23Ap19- and/or EBI3-including cytokines in psoriatic keratinocytes, we analyzed IL-23Ap19 and EBI3 expressions in psoriasis skin lesions, using immunohistochemistry and normal human epidermal keratinocytes (NHEKs) stimulated with inflammatory cytokines, using quantitative real-time polymerase chain reaction (RT-PCR), enzyme-linked immunosorbent assay (ELISA), and liquid chromatography-electrospray tandem mass spectrometry (LC-Ms/Ms). Immunohistochemical analysis showed that IL-23Ap19 and EBI3 expressions were upregulated in the psoriasis skin lesions. In vitro, these expressions were synergistically induced by the triple combination of tumor necrosis factor (TNF)-α, IL-17A, and interferon (IFN)-γ, and suppressed by dexamethasone, vitamin D3, and acitretin. In ELISA and LC-Ms/Ms analyses, keratinocyte-derived IL-23Ap19 and EBI3, but not heterodimeric forms, were detected with humanized anti-IL-23Ap19 monoclonal antibodies, tildrakizumab, and anti-EBI3 antibodies, respectively. Psoriatic keratinocytes may express IL-23Ap19 and EBI3 proteins in a monomer or homopolymer, such as homodimer or homotrimer.     

Admittance Loci Based Design of Plasmonic Sensor Working in Wavelength Interrogation Regime

A prism coupler based plasmonic sensor consisting of a prism, gold (Au) metal film and dielectric sample has been investigated with the use of admittance loci method in wavelength interrogation mode. Prism materials namely fused silica, chalcogenide (2S2G) and silicon have been used to study their effect on surface plasmon sensing in wavelength interrogation mode by admittance loci plots and by corresponding surface plasmon sensing curves. The performance of the plasmonic sensor under wavelength interrogation mode based on the choice of the prism material has been discussed and validated by the dynamic range and sensitivity plots.     

油基钻井液用纳米材料CQ-NZC的封堵性能评价

封堵性是油基钻井液的一个重要评价指标,提高油基钻井液的封堵性有助于井壁稳定,促进快速安全钻井.本文通过使用砂芯漏斗来模拟页岩的微裂缝,建立了一种新的评价油基钻井液封堵性能的方法,利用该方法对自制的纳米材料CQ-NZC在钻井液封堵性及其与氧化沥青封堵剂的复合作用开展了实验研究.结果表明,CQ-NZC在与沥青类封堵剂复配使用时,能够更好地降低滤失量、提高封堵性能.通过微观形态分析发现,CQ-NZC与沥青类封堵剂可以在一定程度上聚结,形成堵孔粒子,提升了钻井液体系的封堵性能.     

Maternal High‐Fat‐Diet Programs Rat Offspring Liver Fatty Acid Metabolism

In offspring exposed in utero to a maternal diet high in fat (HF), we have previously demonstrated that despite similar birth weights, HF adult offspring at 6 months of age had significantly higher body weights, greater adiposity, and increased triacylglycerol (TAG) levels as compared to controls. We hypothesized that a maternal HF diet predisposes to offspring adiposity via a programmed increase in the synthesis of monounsaturated fatty acids in the liver and hence increased substrate availability for liver TAG synthesis. We further hypothesized that programmed changes in offspring liver fatty acid metabolism are associated with increased liver expression of the lipogenic enzyme stearoyl‐CoA desaturase‐1 (SCD‐1). Female rats were maintained on a HF diet rich in monounsaturated fatty acids (MUFA) prior to and throughout pregnancy and lactation. After birth, newborns were nursed by the same dam, and all offspring were weaned to control diet. Plasma and liver fatty acid compositions were determined using gas chromatography/mass spectrometry. Fatty acid C16 desaturation indices of palmitoleic/palmitic and (vaccenic + palmitoleic)/palmitic and the C18 desaturation index of oleic/stearic were calculated. Liver protein abundance of SCD‐1 was analyzed in newborns and adult offspring. Plasma and liver C16 desaturation indices were decreased in HF newborns, but increased in the adult offspring. Liver SCD‐1 expression was increased in the HF adult offspring. These data show that the maternal HF diet during pregnancy and lactation increases offspring liver SCD‐1 protein abundance and alters the liver C16 desaturase pathway.
