Beta-N-acetylglucosaminidase [EC 3.2.1.30] from Vibrio alginolyticus H-8 was purified by column chromatography on DEAE-Sepharose FF, phenyl-Sepharose HP, Superdex 200HR, and Mono Q. The molecular weight of the enzyme was estimated by SDS-gel electrophoresis (SDS-PAGE) to be 75 kDa. The pI was 4.6. The activity was inhibited by Ag+, Hg2+, iodoacetate, p-chloromercuribenzoate, and N-ethylmaleimide.  相似文献

This paper proposes a new force sensorless robust tracking servo system, which detects and suppresses both the periodic disturbance and the sudden disturbance without force sensor. The proposed system estimates both the periodic disturbance and the sudden disturbance and the sudden disturbance by using a new sudden disturbance observer and the memory of tracking error. In the proposed system, the influence of sudden disturbance is suppressed by the proposed sudden disturbance observer. On the other hand, the influence of periodic disturbance is suppressed by means of ZPET feed‐forward servo system. The experimental results point out that the proposed tracking servo system has a precise tracking response against both the periodic disturbance and the sudden disturbance. © 2008 Wiley Periodicals, Inc. We describe a tellurite-based Er3+-doped fiber amplifier (EDFA) with a flat amplification bandwidth of 76 nm and a noise figure of less than 7 dB. We have demonstrated operation of a transmission system at the 3 ?m band. The system consists of a fluoride optical fibre, a superconducting BaPb1?xBixO3 (BPB) detector and a semiconductor Pb-salt diode laser. The low-level optical signal transmitted through the fibre is detected by the BPB detector up to 600 MHz. The observed results suggest that the BPB detector is suitable for use in infra-red optical transmission.  相似文献

The polycrystalline samples of Ca3Co4-x Ga x O9+δ (0 ≤ x ≤ 0.15) were prepared by a simple thermal hydro-decomposition method. The high density ceramics were fabricated using a spark plasma sintering technique. The crystal structure of calcined powders was characterized by x-ray diffraction. The single phase of Ca3Co4-x Ga x O9+δ was obtained. The scanning electron micrograph illustrated the grain alignment perpendicular to the direction of the pressure in the sintering process. The evidence from x-ray absorption near edge spectra were used to confirm the oxidation state of the Ga dopant. The thermoelectric properties of the misfit-layered of Ca3Co4-x Ga x O9+δ were investigated. Seebeck coefficient tended to decrease with increasing Ga content due to the hole-doping effect. The electrical resistivity and thermal conductivity were monotonically decreased with increasing Ga content. The Ga doping of x = 0.15 showed the highest power factor of 3.99 × 10-4 W/mK2 at 1,023 K and the lowest thermal conductivity of 1.45 W/mK at 1,073 K. This resulted in the highest ZT of 0.29 at 1,073 K. From the optical absorption spectra, the electronic structure near the Fermi level show no significant change with Ga doping.  相似文献

Dolichol-phosphate-mannose (Dol-P-Man) serves as a donor of mannosyl residues in major eukaryotic glycoconjugates. It donates four mannosyl residues in the N-linked oligosaccharide precursor and all three mannosyl residues in the core of the glycosylphosphatidylinositol anchor. In yeasts it also donates one mannose to the O-linked oligosaccharide. The yeast DPM1 gene encodes a Dol-P-Man synthase that is a transmembrane protein expressed in the endoplasmic reticulum. We cloned human and mouse homologues of DPM1, termed hDPM1 and mDPM1, respectively, both of which encode proteins of 260 amino acids, having 30% amino acid identity with yeast Dpm1 protein but lacking a hydrophobic transmembrane domain, which exists in the yeast synthase. Human and mouse DPM1 cDNA restored Dol-P-Man synthesis in mouse Thy-1-deficient mutant class E cells. Mouse class E mutant cells had an inactivating mutation in the mDPM1 gene, indicating that mDPM1 is the gene for class E mutant. In contrast, hDPM1 and mDPM1 cDNA did not complement another Dol-P-Man synthesis mutant, hamster Lec15 cells, whereas yeast DPM1 restored both mutants. Therefore, in contrast to yeast, mammalian cells require hDPM1/mDPM1 protein and a product of another gene that is defective in Lec15 mutant cells for synthesis of Dol-P-Man.  相似文献

The microstructural properties of the β-FeSi2/FeSi structure prepared from a molten salt have been characterized using transmission electron microscopy (TEM). The β-FeSi2 films were grown on FeSi substrates at the heat treatment temperature of 900 °C from 1 min to 24 h using the molten salt technique. It is found that the films consisted of a thin surface layer and a thick underlying layer with columnar-shaped domains. The crystallographic directions of the domains are mostly randomly oriented. The β-FeSi2 domains in the film, however, have specific crystallographic orientation relationships with the adjoining domains and the FeSi substrate. A high density of the stacking faults on the β-FeSi2 (100) planes was also observed through the films. Moreover, the growth evolution of the β-FeSi2 domains, the defect characteristics and the formation mechanism of the defects are discussed.  相似文献

Intravenous (I.V.) cocaine (0.03-3 mg/kg) produced dose-dependent, rapid, and brief increases in blood pressure (BP) in conscious rats pretreated with the dopamine receptor antagonist, SCH 23390. Monoamine uptake inhibitors structurally analogous to cocaine (cocaethylene, CFT, betaCIT, CPT, (+)-cocaine, norcocaine, and benztropine) also produced this rapid pressor response, whereas structurally unrelated uptake inhibitors with diverse monoamine transporter selectivities (BTCP, indatraline, GBR 12935, mazindol, nomifensine, and zimeldine) either did not produce a rapid pressor response or produced only a small pressor response. At nonconvulsant doses, the sodium channel blockers acetylprocainamide, dibucaine, dyclonine, prilocaine, proparacaine, quinidine, and tetracaine produced a small pressor response or no increase in BP. In rats implanted with telemetric devices, cocaine and its analog, CFT, produced a biphasic pharmacological response that consisted of an initial brief and abrupt behavioral arousal associated with a rapid, large increase in BP followed by prolonged, parallel increases in BP and locomotor activity. Pretreatment with SCH 23390 prevented the prolonged but not the initial rapid and brief pressor and activity responses to both cocaine and CFT administration. The present data suggest that the inhibition of dopamine, norepinephrine, or serotonin transporter functions, either alone or in combination, does not mediate the rapid pressor response to cocaine. The sodium channel-blocking action of cocaine per se does not appear to be involved in the rapid pressor response to cocaine. Finally, the present results confirm previous findings that dopaminergic mechanisms mediate the prolonged increases in BP and locomotor activity produced by cocaine.  相似文献
