The authors describe a new technique for extracting the intrinsic laser-diode dynamic properties accurately. This simple technique eliminates the need for accurate microwave calibration of the test equipment and problems of microwave reflections, nonideal frequency response of laser mount, and detector. The effect of the parasitic components of the laser diode are also eliminated from the results so that measurements of important dynamic properties of the laser can be found up to high frequencies (10-20 GHz) on standard laser diodes. The techinque being used to measure variations of resonance peak and damping factor at different bias levels for a standard bulk active region 1.3 μm laser diode is shown  相似文献

The authors examined over time the relationship between characteristics of the family caregiving system for older persons with Alzheimer's disease and the health of 136 adult offspring who were involved in care. The goal was to determine if family and offspring health relationships found at point of initial patient evaluation (Tl) remained consistent over a 12-month period (T2) and if qualities of the family environment assessed at Tl predicted a change in offspring health and well-being as the demands for care increased over time. Offspring carehours, gender, education, distress, and patient severity (Step 1); measures of family functioning (Step II); and family by distress interaction terms (Step III) were used to predict offspring health and well-being at T2. Findings indicated that (a) family characteristics associated with offspring health at Tl displayed consistent predictions of health over time and (b) family emotion management variables predicted a reduction in offspring health scores by T2. These data document the ongoing effects of the family context of care and suggest the family setting as a target for intervention. The interaction of the Fyn SH3 domain with the p85 subunit of PI3-kinase is investigated using structural detail and thermodynamic data. The solution structure complex of the SH3 domain with a proline-rich peptide mimic of the binding site on the p85 subunit is described. This indicates that the peptide binds as a poly(L-proline) type II helix. Circular dichroism spectroscopic studies reveal that in the unbound state the peptide exhibits no structure. Thermodynamic data for the binding of this peptide to the SH3 domain suggest that the weak binding (approximately 31 microM) of this interaction is, in part, due to the entropically unfavorable effect of helix formation (delta S0 = -78 J.mol-1.K-1). Binding of the SH3 domain to the intact p85 subunit (minus its own SH3 domain) is tighter, and the entropic and enthalpic contributions are very different from those given by the peptide interaction (delta S0 = +252 J.mol-1.K-1; delta H0 = +44 kJ.mol-1). From these dramatically different thermodynamic measurements we are able to conclude that the interaction of the proline-rich peptide does not effectively mimic the interaction of the intact p85 subunit with the SH3 domain and suggest that other interactions could be important.  相似文献

BACKGROUND: The authors evaluated the effect of postoperative radiation therapy on freedom from biochemical failure (bNED) in men with prostate carcinoma who had pathologic seminal vesicle invasion after radical prostatectomy and negative pelvic lymph node dissection (pT3cN0). METHODS: Between 1989 and 1995, 375 men underwent radical prostatectomy at Thomas Jefferson University Hospital. Fifty-three men (13%) had pT3cN0 prostate carcinoma and were the subject of this analysis. Men in whom prostate specific antigen (PSA) could not be detected were deemed free of biochemical failure. RESULTS: Of the 53 men with pT3cN0 prostate carcinoma, 18 had an elevated PSA immediately after surgery and received salvage radiation therapy (RT). The 3-year bNED rate for this group was only 38%. At 3 months, PSA could not be detected in the other 35 men. Fifteen of those 35 men underwent early adjuvant RT, and the other 20 were observed for biochemical failure. The 3-year bNED rate for the 15 patients treated with immediate adjuvant RT was 86%, compared with 48% for the 20 men who were observed (P = 0.01). CONCLUSIONS: These data suggest that early adjuvant RT for men with pT3cN0 prostate carcinoma and no detectable PSA postoperatively reduces the likelihood of future biochemical failure. Men with pT3cN0 prostate carcinoma and a persistently elevated postoperative PSA level are less likely to benefit from RT and should be considered for systemic therapy.  相似文献

Interleukin 4 (IL-4) inhibits carcinoma cell growth and promotes expression of differentiation-associated products by normal and malignant epithelial cells. The effects of IL-4 and IL-13 on expression of the CD44 transmembrane adhesion receptor were examined in human epithelial cell lines of colonic (HT-29, CaCo-2, DLD-1, T84), breast (MCF-7, ZR75-1) and liver (Hep-G2, PLC/PRF/5) origins as well as mitogen-activated and resting peripheral blood lymphocytes (PBL) and T cell lines (Jurkat, HUT78). Liver and Jurkat cells were negative for CD44. Colonic, breast and HUT78 cells expressed CD44 constitutively and all except DLD-1 and HUT78 also expressed CD44 splice variant (CD44v) epitopes. All cell lines expressed IL-4 receptors, but IL-4 and IL-13 induced upregulation of CD44 only in the colonic cell lines. CD44v was also upregulated, but there was no de novo induction of CD44v in variant-negative cells and no de novo expression of CD44 in the CD44(-) lines. CD44 upregulation in mitogen-activated PBL was not increased by IL-4 and IL-13 and was not inhibited by neutralizing antibodies. Other cytokines tested [interferon gamma (IFN-gamma, tumour necrosis factor alpha (TNF-alpha), transforming growth factor beta1 (TGF-beta1) and IL-6] did not affect CD44 core epitope expression in the cell lines tested.  相似文献

From considerations of the pull-out behaviour of individual inclined wires from model composites, the work of fracture and post-cracking stress of random fibre reinforced composites have been estimated. Results are presented for cement composites reinforced with short, high-strength steel wires. It was found that when the fibres were short and/or the interfacial shear stress low, a substantial part of the composite work of fracture was due to plastic deformation of those fibres which were not perpendicular to the matrix crack face. Under such conditions random fibre composites showed superior properties than aligned composites of the same volume fraction of reinforcement.
Résumé On a évalué le travail de rupture et la contrainte après fissuration de matériaux renforcés de fibres à répartition aléatoire à partir de l'étude de modèles de composites dans lesquels on a considéré le comportement à l'arrachement de filaments individuels inclinés. Les résultats donnés ici se rapportent à des composites à base de ciment renforcé avec de courts filaments d'acier de haute résistance. On constate que lorsque les fibres sont courtes et/ou la contrainte de cisaillement à l'interface faible, une part substantielle du travail de rupture est due à la déformation plastique des fibres qui ne sont pas perpendiculaires à la surface fissurée. Dans de telles conditions, les composites à fibres réparties au hasard montrent des proprietés meilleures que les composites à fibres alignées pour le même rapport de volume de renforcement.

A petrographical investigation of the sandstones of the Brent Group (Middle Jurassic) from the Murchison field has produced evidence for the existence of two transgressive events within the sequence, one occurring immediately above the Etive Fm., the other within the upper part of the Ness Fm. These transgressions have not been previously recorded in Murchison and their discovery demands that some modification be made to the existing depositional model for the Brent Group of this area. A volcanic ash horizon has been found in a similar stratigraphic position to that recorded from Statfjord, and if this, and the two transgressive horizons noted above, are of more than local extent they could prove to be useful stratigraphic markers.
Although the diagenesis of the Brent Group sandstones in Murchison differs little from that recorded elsewhere in the Brent province, the interpretation of the timing of the development of certain phases differs somewhat from those of other authors. In particular, kaolinite development appears to have taken place soon after burial in response to periods of emergence and weathering during Brent Group deposition, rather than through groundwater circulation during the Kimmerian tectonic phase.  相似文献

The kinetics of protein adsorption/desorption onto peptide microarrays was studied using real-time surface plasmon resonance (SPR) imaging. S protein binding interactions were examined using an array composed of five different peptides: N terminal and C terminal immobilized wild-type S peptide (S1 and S2), an alternate binding sequence derived by phage display (LB2), an NVOC-protected S peptide, and a FLAG peptide control sequence (F). Kinetic measurements of the S protein-S1 peptide interaction were analyzed to determine a desorption rate constant (k(d)) of 1.1 (+/-0.08) x 10(-2) s(-1), an adsorption rate constant (k(a)) of 1.9 (+/-0.05) x 10(5) M(-1) s(-1), and an equilibrium adsorption constant (K(Ads)) of 1.7 (+/-0.08) x 10(7) M(-1). SPR imaging equilibrium measurements of S protein to S1 peptide were performed to independently confirm the kinetically determined value of K(Ads). Rate constants for the S2 and LB2 peptides on the array were measured as follows: 1.6 (+/-0.04) x 10(5) M(-1) s(-1) (k(a)) and 1.1 (+/-0.07) x 10(-2) s(-1) (k(d)) for S2, 1.2 (+/-0.05) x 10(5) M(-1) s(-1) (k(a)) and 1.1 (+/-0.03) x 10(-2) s(-1) (k(d)) for LB2. In addition to S protein adsorption/desorption, real-time SPR imaging of peptide arrays was applied to study the surface enzymatic activities of the protease factor Xa. Enzymatic cleavage of the substrate peptide (P1) was shown to follow first-order kinetics and proceed at a rate 10 times faster than that of the mutant peptide (P2), with cleavage velocities of 5.6 (+/-0.3) x 10(-4) s(-1) for P1 and 5.7 (+/-0.3) x 10(-5) s(-1) for P2.  相似文献

Good performance of optical coatings depends on the appropriate combination of optical and mechanical properties. Therefore, successful applications require good understanding of the relationship between optical microstructural and mechanical characteristics and film stability. In addition, there is a lack of standard mechanical tests that allow one to compare film properties measured in different laboratories. We give an overview of the methodology of mechanical measurements suitable for optical coatings; this includes depth-sensing indentation, scratch resistance, friction, abrasion and wear testing, and stress and adhesion evaluation. We used the techniques mentioned above in the same laboratory to systematically compare the mechanical behavior of frequently used high- and low-index materials, namely, TiO2, Ta2O5, and SiO2, prepared by different complementary techniques. They include ion-beam-assisted deposition by electron-beam evaporation, magnetron sputtering, dual-ion-beam sputtering, plasma-enhanced chemical-vapor deposition, and filtered cathodic arc deposition. The mechanical properties are correlated with the film microstructure that is inherently related to energetic conditions during film growth.  相似文献
