The direct metal deposition (DMD) with laser is a free-form metal deposition process for manufacturing dense pieces, which allows generating a prototype or small series of near net-shape structures. One of the most critical issues is that produced pieces have a deleterious surface finish which systematically requires post machining steps. This problem has never been fully addressed before.The present work describes investigations on the DMD process, using an Yb-YAG disk laser, and a widely used titanium alloy (Ti–6Al–4V) to understand the influence of the main process parameters on the surface finish quality. The focus of our work was: (1) to understand the physical mechanisms responsible for deleterious surface finishes, (2) to propose different experimental solutions for improving surface finish.In order to understand the physical mechanisms responsible for deleterious surface finishes, we have carried out: (1) a precise characterization of the laser beam and the powder stream; (2) a large number of multi-layered walls using different process parameters (P(W), V(m/min), Dm (g/min), Gaussian or uniform beam distribution); (3) a real time fast camera analysis of melt pool dynamics and melt-pool – powder stream coupling; (4) a characterization of wall morphologies versus process parameters using 2D and 3D profilometry.The results confirm that surface degradation depends on two distinct aspects: the sticking of non-melted or partially melted particles on the free surfaces, and the formation of menisci with more or less pronounced curvature radii. Among other aspects, a reduction of layer thickness and an increase of melt-pool volumes to favor re-melting processes are shown to have a beneficial effect on roughness parameters. Last, a simple analytical model was proposed to correlate melt-pool geometries to resulting surface finishes.  相似文献

The catalytic contribution of His48 in the active site of porcinepancreatic phospholipase A2 was examined using site-directedmutagenesis. Replacement of His48 by lysine (H48K) gives riseto a protein having a distorted lipid binding pocket. Activityof this variant drops below the detection limit which is 107-foldlower than that of the wild-type enzyme. On the other hand,the presence of glutamine (H48Q) or asparagine (H48N) at thisposition does not affect the structural integrity of the enzymeas can be derived from the preserved lipid binding propertiesof these variants. However, the substitutions H48Q and H48Nstrongly reduce the turnover number, i.e. by a factor of 105.Residual activity is totally lost after addition of a competitiveinhibitor. We conclude that proper lipid binding on its ownaccelerates ester bond hydrolysis by a factor of 102. With theselected variants, we were also able to dissect the contributionof the hydrogen bond between Asp99 and His48 on conformationalstability, being 5.2 kJ/mol. Another hydrogen bond with His48is formed when the competitive inhibitor (R)-2-dodecanoylamino-hexanol-1-phosphoglycolinteracts with the enzyme. Its contribution to binding of theinhibitor in the presence of an interface was found to be 5.7kJ/mol.  相似文献

Hydroxyapatite (HA, Ca10(PO4)6(OH)2) is a bioactive material being frequently used as a coating onto implants. The typical coating technology is air plasma spraying with the use of coarse powder. The resulting coatings are relatively thick (about 200-400 µm) and porous. Much thinner coatings being 5-40 µm thick, can be obtained by emerging technology of suspension plasma spraying with the use of powder particles having the diameters ranging from a few submicrometers to a few micrometers. The paper describes the way of synthesizing and preparing such fine powder starting from an aqueous solution of ammonium phosphate (H2(PO4)NH4) and calcium nitrate (Ca(NO3)·4H2O) using statistical design of experiments (DOE). The design was made using composite matrix including a full factorial plan, star points and 3 experiments in the centre. The crystal phases purity and the mass of powder batch were the optimized responses of the powder synthesis and the concentration of calcium ions and volume of ammonium hydroxide were the experimental variables. The synthesized material was characterized by X-ray diffraction (XRD). The powder was calcined and crushed using a milling machine with zirconia balls and resulting morphology and size of fine particles was characterized using scanning electron microscope (SEM) and laser sizer correspondingly. The powder was then formulated into water and alcohol based suspension and the zeta potential was determined to understand its capacity of agglomeration. It was found out that the formulation of the suspension with the use of ethanol slightly favours dispersion of solid particles in the suspension. The initial tests of water based suspension plasma spraying onto titanium substrate were also carried out and the XRD phase analysis of obtained coatings was carried out the presence of HA and its phases of decomposition.  相似文献

Cereals are an important source of sterols and stanols in the human diet. The present study underlines the effect of genotype and weather conditions in bread wheat, on total sterol and stanol content (TSS), agronomic traits, proteins and ash content under organic conditions. Variations in TSS as well as other characters between two sowing dates were observed. A broad genotypic variability was also reported since extreme genotypes differed by more than 30 mg 100 g−1 DW for TSS, with total stanol content varying twofold. Moreover, two groups of genotypes that differed in agronomic production, ash and protein content were depicted, based on their response to an increase in temperature. This result suggests that the genotypic factor prevails over the sowing date factor for determining sterol and stanol traits in wheat cultivated under organic conditions. Nevertheless, a strong interaction exists between the two factors, which can be used to drive bioaccumulation of these molecules.  相似文献

BACKGROUND: Feeding conjugated linoleic acid (CLA) to pigs leads to CLA‐enriched meat products, which are of current interest owing to the potential health properties of CLA. However, dietary CLA increases the ratio of saturated to monounsaturated fatty acids (MUFA) in pig tissues. Combining CLA with high MUFA levels in pig diets may counteract this effect. Owing to the modification of fatty acid composition, variation in the volatile profile of derived meat products might be expected. The aim of the present work was to study the volatile profile, and its change during storage, of experimental liver pâté manufactured from pigs fed different levels of CLA and MUFA. RESULTS: Regardless of the CLA or MUFA level, 98 compounds were identified. Lower area units were observed for most branched aldehydes, ketones and alcohols and some furans, sulfur compounds and nitrogen compounds with increasing CLA and MUFA contents. Levels of nonanal, propan‐2‐one, dodecane, 2‐pentylfuran and ethylbenzene were higher in CLA pâtes. The effect of experimental diets on the change in volatiles during storage was not significant. CONCLUSION: Supplementation of pig diets with CLA isomers in combination with low or high MUFA levels seems to have a significant effect on the volatile profile of pork liver pâté. Fracture toughness of ZrO2-toughened alumina could he increased by macroscopic interfaces, such as those existing in laminated composites. In this work, tape casting was used to produce A/A or A/B laminates, where A and B can be Al2O3, Al2O3/5 vol% ZrO2, and Al2O3/l0 vol% ZrO2. ABSTRACT: Wild lowbush blueberry fruit extract was fractionated using vacuum chromatography and analyzed for chemopreventive potential using bioassays that test the ability of compounds to inhibit the initiation, promotion, and progression stages of carcinogenesis. A fraction containing phytosterols was active against the initiation stage (quinone reductase assay). However, more polar compounds were inhibitors of later stages of carcinogenesis; a fraction containing flavan-3-ols and fractions containing mainly anthocyanins, phenolic acids, flavan-3-ols, and some proanthocyanidin dimers demonstrated activity against the promotion stage (cyclooxygenase and ornithine decarboxylase assays, respectively), and a proanthocyanidin-rich fraction demonstrated antiproliferation activity (inhibition of cancerous murine hepatocyte proliferation is associated with the progression stage). These results indicate that lowbush blueberries contain a range of compounds that have bioactivity against multiple stages of carcinogenesis, and different types of phenolic compounds are active at different stages.  相似文献

A paced search of a visual display was made in auditory conditions containing bursts of noise at either 68 or 105 db. Each selected visual item required 2 types of response, crossing out and counting. The preferred activity in the quieter condition was counting. When the test instructions emphasized this aspect of the task, attention shifted further toward the preferred activity during loud noise. When the instructions emphasized searching, there were no significant differences between noise and quiet. It appears that although noise will not always induce a redistribution of the attention needed to respond equally often in 2 paced activities, when it does so, the preferred activity gains. Thymidine monophosphate kinase (TMPK) from Mycobacterium tuberculosis (TMPKmt) is an attractive target for the design of specific inhibitors. This fact is the result of its key role in the thymidine pathway and of unique structural features in the active site observed by X-ray crystallography, especially in comparison to its human counterpart (TMPKh). Different 5-modified thymidine derivatives, as well as purine and pyrimidine analogues or C-nucleosides were tested on TMPKmt and TMPKh, and the results were rationalized by docking studies. 5-Halogenated 2'-deoxyuridines are the best inhibitors of TMPKmt found and present the highest selectivity indexes in favor of TMPKmt.  相似文献
