The proposed rapid small‐scale starch isolation technique in the laboratory was a combination of dry grinding of grain, suspension of the resulting flour in extraction buffer, application of ultrasonic sonication, then separation by sucrose density centrifugation. Light microscopy of separated fractions showed intact starch granules in the pellet and proteins and damaged starch in the top layer. The extraction method yielded 61% starch from sorghum and 63% from maize. The isolated starch showed lower starch damage and proteins content than by the conventional method. The gelatinization enthalpy of isolated starch was slightly higher than by wet grinding conventional method. In addition to low amount of starting flour (100 mg) the new starch isolation method was performed in less than 2 h from dry grinded seed to dried starch. Thus, it could be a useful method for cereal chemists and plant genetists.  相似文献

This study evaluated the effects of two commonly used microbial inoculants (Lactobacillus buchneri (LB) and Lactobacillus plantarum (LP)), formic acid (FA) and quebracho tannins (QT) on the fermentation quality, aerobic stability and in situ rumen degradation of pea/wheat bi‐crop forages. Precision‐chopped spring pea (Pisum sativum, var Magnus) and wheat (Triticum aestivum, var Axona) bi‐crops (3:1 pea/wheat ratio) harvested at a combined dry matter (DM) content of 301 g kg?1 were used for the study. The bi‐crops were conserved without (Control) or with inoculants based on lactic acid bacteria (LB (105 CFU g?1 fresh weight (FW)) or LP (106 CFU g?1 FW)), QT (16 g kg?1 FW) or FA (2.5 g kg?1 FW) in laboratory silos of 1.5 kg capacity, with each treatment being replicated six times. The pH, chemical composition, aerobic stability and in situ rumen degradation of DM, nitrogen (N) and neutral detergent fibre (NDF) after 112 days of ensilage were measured. The average pH at silo opening was 4.0, suggesting that the silages were well fermented. There were no significant effects of additive treatment on water‐soluble carbohydrate, total N, soluble N, ammonia N and NDF. Lactic acid and acetic acid were the main fermentation products. High concentrations of acetic acid were found in all the treatments, indicating a heterofermentative pathway. Although FA treatment gave the most aerobically stable silage, the Control and QT‐treated silages did not heat up by more than 1 °C until after 6 days of exposure to air. There were no effects of additives on DM degradation characteristics. However, the inoculants increased the rate of N and NDF degradation in the rumen, and both FA and QT reduced the effective and potential degradation of N. © 2001 Society of Chemical Industry  相似文献

Inhibition of Escherichia coli O157:H7, Salmonella Typhimurium, and Listeria monocytogenes by grape seed extract (ActiVin) and pine bark extract (Pycnogenol) and the effect of these natural extracts on the oxidative stability of raw ground beef were studied. In an agar dilution test, the MICs of ActiVin and Pycnogenol were determined to be 4.0 mg/ml for 4.43 log CFU per plate of E. coli O157:H7 and 4.0 mg/ml for 4.38 log CFU per plate of L. monocytogenes. In an inhibition curve test, populations of E. coli O157:H7, Salmonella Typhimurium, and L. monocytogenes fell to below the detection limit (10 CFU/ml) after 16 h of incubation. The numbers of E. coli O157:H7, L. monocytogenes, and Salmonella Typhimurium declined by 1.08, 1.24, and 1.33 log CFU/g, respectively, in raw ground beef treated with 1% Pycnogenol after 9 days of refrigerated storage. ActiVin (1%) and oleoresin rosemary (1%) resulted in an approximately 1-log CFU/g reduction in the populations of all three pathogens after 9 days. The addition of 1% ActiVin and Pycnogenol contributed to the maintenance of an acidic pH of 5.80 and 5.58, respectively, in raw ground beef. Compared to the control, all treatments increased in L* (lightness), with the exception of ActiVin. ActiVin and oleoresin rosemary had the highest a* (redness) and b* (yellowness) values, respectively. ActiVin most effectively retarded lipid oxidation, followed by Pycnogenol. The results suggest that these natural extracts have potential to be used with other preservative methods to reduce pathogenic numbers, lipid oxidation, and color degradation in ground beef.  相似文献

Engineered nanoparticles (ENPs), such as metallic or metallic oxide nanoparticles (NPs), have gained much attention in recent years. Increasing use of ENPs in various areas may lead to the release of ENPs into the environment and cause the contamination of agricultural and food products by ENPs. In this study, we selected two important ENPs (zinc oxide [ZnO] and silver [Ag] NPs) as potential food contaminants and investigated their toxicity via an in vitro model using Caco‐2 cells. The physical properties of ENPs and their effects on Caco‐2 cells were characterized by electron microscopy and energy dispersive X‐ray spectroscopic (EDS) techniques. Results demonstrate that a significant inhibition of cell viability was observed after a 24‐h of exposure of Caco‐2 cells to 3‐, 6‐, and 12‐mM ZnO NPs or 0.5‐, 1.5‐, and 3‐mM Ag NPs. The noticeable changes of cells include the alteration in cell shape, abnormal nuclear structure, membrane blebbing, and cytoplasmic deterioration. The toxicity of ZnO NPs, but not that of Ag NPs after exposure to simulated gastric fluid, significantly decreased. Scanning transmission electron microscopy shows that ZnO and Ag NPs penetrated the membrane of Caco‐2 cells. EDS results also confirm the presence of NPs in the cytoplasm of the cells. This study demonstrates that ZnO and Ag NPs have cytotoxic effects and can inhibit the growth of Caco‐2 cells.  相似文献

In this article, an analytical approach, for optimum nonlinear circuit analysis is described. At a given input frequency, the method consists in determining, for each nonlinear component of the circuit, a closed volume including all the allowed powers at its terminal ports. The boundary of this volume, called “characteristic surface,” allows extremum powers of nonlinear devices to be predicted and then optimum performance of the circuit to be deduced. Applied to various microwave transistor circuits, this procedure has shown good agreement between computed and measured or previous published results. © 1996 John Wiley & Sons, Inc.  相似文献

This work aims to study the effects of date stone flour (DSF) on morphology, thermal, and mechanical properties of polypropylene (PP) composites in the absence and presence of ethylene‐butyl acrylate‐glycidyl methacrylate (EBAGMA) used as the compatibilizer. DSF was added to the PP matrix at loading rates of 10, 20, 30, and 40 wt %, while the amount of compatibilizer was fixed to the half of the filler content. The study showed through scanning electron microscopy analysis that EBAGMA compatibilizer improved the dispersion and the wettability of DSF in the PP matrix. Thermogravimetric analysis (TGA) indicated a slight decrease in the decomposition temperature at onset (Tonset) for all composite materials compared to PP matrix, whereas the thermal degradation rate was slower. Differential scanning calorimetry (DSC) data revealed that the melting temperature of PP in the composite materials remained almost unchanged. The nucleating effect of DSF was however reduced by the compatibilizer. Furthermore, the incorporation of DSF resulted in the increase of stiffness of the PP composites accompanied by a significant decrease in both the stress and strain at break. Papermaking sludge (PS), a waste residue from the pulp and paper processing, has brought great pressure on the environment because of large quantities that are produced in paper mills. This work was carried out to explore the possibility of making PS/cement composite products using solid waste of PS. Boards measuring 350 × 270 × 12 mm3 were manufactured using PS contents of 40, 50, and 60 wt%, adhesive dosages of 0, 10, and 15 wt%, and 0 and 5 wt% of calcium chloride as an accelerator. At least three replications were fabricated for each treatment, and some mechanical and physical properties of the boards were evaluated. Test results showed that the bending and internal strengths of the specimens decreased with an increase in the PS content, and the maximum values were obtained at PS loading of 40 wt%. The negative influence of PS content on the mechanical properties can be explained by the reduced bonding ability because of weaker PS compared with cement. Screw withdrawal values were up to 22.7 kPa. Water absorption and thickness swelling of cement mortar considerably increased with increased content of PS, with a corresponding reduction of bulk density. In general, all properties of the boards were improved when the adhesive and calcium chloride contents were increased. The results showed that an increase in board density improved the mechanical and physical properties. The aim of this study was to investigate the performance of UHMWPE/HDPE-reinforced kenaf, basalt and hybrid kenaf/basalt composites. Mechanical testing of these samples was carried out such as tensile, flexural (three-point bending) and an impact test (Charpy). Pure resin (UHMWPE/HDPE) samples were tested and compare with reinforced 10% weight fraction of kenaf, basalt and hybrid kenaf/basalt samples to identifying their contribution and potential in this new composite material. UHMWPE/ HDPE sample was produced in constant ratio 60:40 respectively via extrusion process. Basalt reinforced UHMWPE/HDPE generates the highest elastic modulus result compared to kenaf and hybrid kenaf/basalt as a reinforcement material. The tensile results of kenaf reinforcement UHMWPE/HDPE samples are significantly higher (20%) than pure blend resin, which is an indication for good performance of kenaf, basalt and hybrid kenaf/basalt to be used in UHMWPE/HDPE-blend polymers. The flexural and Charpy strengths show the drawback results, where performance of polymer is reduced 5% with the absence of kenaf. It can be concluded that kenaf, basalt and hybrid kenaf/basalt fiber successfully increase the UHMWPE/HDPE blends performance especially under tensile loading.  相似文献
