The spectrophotometric assay of 2,4-dienoyl coenzyme A (CoA) reductase (EC 1.1.1.34) was modified to improve the linearity and sensitivity of this method. 5-Phenyl-2,4-pentadienoyl-CoA, which has an absorbance maximum at 340 nm with an extinction coefficient of 44,300 M−1 cm−1, was synthesized and used as substrate. This compound is reduced by nicotinamide adenine dinucleotide phosphate (NADPH)-dependent 2,4-dienoyl-CoA reductase to 5-phenyl-3-pentenoyl-CoA. When a tissue homogenate serves as an enzyme source, the product is further metabolized by Δ3Δ2-enoyl-CoA isomerase (EC 5.3.3.8) to 5-phenyl-2-pentenoyl-CoA, which is hydrated to 5-phenyl-3-hydroxypentanoyl-CoA by enoyl-CoA hydratase (EC 4.2.1.17). The modified assay method, which measures the decrease in absorbance at 340 nm due to the reduction of 5-phenyl-2,4-pentadienoyl-CoA and the oxidation of NADPH, is linear for a longer period of time and is twice as sensitive as the conventional assay with 2,4-decadienoyl-CoA as substrate.  相似文献

Co-ground powders of the poorly water-soluble drug nifedipine and a hydrophilic carrier, [partially hydrolyzed gelatin (PHG), polyvinylpyrrolidone (PVP), sodium dodecyl sulfate (SDS), hydroxypropyl methylcellulose (HPMC), polyethylene glycol (PEG), urea or Pluronic F108] were prepared in order to improve the dissolution rate of nifedipine. The effects of type of grinding equipment, grinding time, and type of hydrophilic carrier on the crystallinity of nifedipine (x-ray diffraction and differential scanning calorimetry) on the interaction between drug and carriers (differential scanning calorimetry), on the particle size and appearance (scanning electron microscopy), on the wettability (contact angle measurements), and on the drug release were investigated. Grinding nifedipine together with these carriers improved the dissolution rate. PHG-ground mixtures resulted in the fastest dissolution rate followed by PVP, SDS, HPMC, Pluronic, urea, and PEG. A technoeconomic analysis and optimization of wind turbine size and layout are performed using WAsP software. A case study of a 100‐MW wind farm located in Egypt is considered. Wind atlas for Egypt was used as the input data of the WAsP software. Two turbine models of powers 52 and 80 MW are considered for this project. The wind turbine size and distributions are selected based on the technoeconomic optimization, namely minimum wake effect, maximum annual energy production (AEP) rate, optimum cash flow, and payback period. The future worth method is adopted in economic comparison between the two alternatives, and the cash flow diagram provided the payback period and future worth after the lifetime of the plant. The results showed that (1) the AEP dramatically decreases for a wind farm area less than 15 km2; (2) the turbine spacing, spacing‐to‐diameter ratio, and the setback distances decrease and the wind turbine density and wake losses increase with decreasing the wind turbines size; (3) the total net AEP using G52 is lower than that of using G80 by about 16%; (4) the technoeconomic analysis recommended using G80 as it has higher profit than those of G52 by about $20 million.  相似文献

Pulsed laser deposition of Al2O3 onto porous silicon (PS) is shown to provide excellent passivation of multi-crystalline silicon surfaces intended for solar cells applications. Surface passivation and reflectivity are investigated before and after the deposition of various nominal thicknesses of Al2O3 ranging from isolated nanoparticles to ~80 nm-thick films. The level of surface passivation is determined by techniques based on photoconductance and FTIR. As a result, the effective minority carrier lifetime increase from 1 to 130 μs at a minority carrier density (Δn) of 1?×?1013 cm?3. However, passivation scheme provide a significant decrease in the reflectivity; it’s reduced from 28% to about 5% after Al2O3/PS coating.  相似文献

Video compression in medical video streaming is one of the key technologies associated with mobile healthcare. Seamless delivery of medical video streams over a resource constrained network emphasizes the need of a video codec that requires minimum bitrates and maintains high perceptual quality. This paper presents a comparative study between High Efficiency Video Coding (HEVC) and its potential successor Versatile Video Coding (VVC) in the context of healthcare. A large-scale subjective experiment comprising of twenty-four non-expert participants is presented for eight different test conditions in Full High Definition (FHD) videos. The presented analysis highlights the impact of compression artefacts on the perceptual quality of HEVC and VVC processed videos. Our results and findings show that VVC clearly outperforms HEVC in terms of achieving higher compression, while maintaining high quality in FHD videos. VVC requires upto 40% less bitrate for encoding an FHD video at excellent perceptual quality. We have provided rate-quality curves for both encoders and a degree of overlap across both codecs in terms of perceptual quality. Overall, there is a 71% degree of overlap in terms of quality between VVC and HEVC compressed videos for eight different test conditions.  相似文献

In this paper, the design and analysis of the all-optical up- and down-wavelength converter based on four-wave mixing (FWM) effect of semiconductor optical amplifier Mach–Zehnder interferometer (SOA-MZI) have been presented. The return-to-zero (RZ) modulated data signal at a bit rate of 60 Gbps has been evaluated for error-free operation to show the feasibility of proposed system at different pump wavelength. The converted signal power and quality factor are investigated as the function of variable signal power and pump power. The optimized operating input signal power of ?5 dBm with Q-factor of \(\sim \)28 dB for RZ modulated signal by using SOA-MZI structure with enhanced FWM effect. The important contribution of these investigations that it is possible to expand the optical network with limited available channel bandwidth by utilizing the wavelength converter and gives an approach to implement wavelength converter for future hybrid optical access networks.  相似文献

17β‐Hydroxysteroid dehydrogenase (17β‐HSDcl) from the filamentous fungus Curvularia lunata (teleomorph Cochliobolus lunatus) catalyzes NADP(H)‐dependent oxidoreductions of androgens and estrogens. Despite detailed biochemical and structural characterization of 17β‐HSDcl, its physiological function remains unknown. On the basis of amino acid sequence alignment, phylogenetic studies, and the recent identification of the physiological substrates of the homologous MdpC from Aspergillus nidulans and AflM from Aspergillus parasiticus, we propose an anthrahydroquinone as the physiological substrate of 17β‐HSDcl. This is also supported by our analysis of a secondary metabolite biosynthetic gene cluster in C. lunata m118, containing 17β‐HSDcl and ten other genes, including a polyketide synthase probably involved in emodin formation. Chemoenzymatic reduction of emodin by 17β‐HSDcl in the presence of sodium dithionite verified this hypothesis. On the basis of these results, the involvement of a 17β‐HSDcl in the biosynthesis of other anthrahydroquinone‐derived natural products is proposed; hence, 17β‐HSDcl should be more appropriately referred to as a polyhydroxyanthracene reductase (PHAR).  相似文献
