Bacteria, plants, and higher and lower animals have evolved an innate immune system as a first line of defense against microbial invasion. Some of these organisms produce antimicrobial peptides (AMPs) as a part of this chemical immune system. AMPs exert their antimicrobial activity by binding to components of the microbe's surface and disrupting the membrane. The overall goal of this study was to apply the AMP magainin I as a recognition element for Escherichia coli O157:H7 and Salmonella typhimurium detection on an array-based biosensor. We immobilized magainin I on silanized glass slides using biotin-avidin chemistry, as well as through direct covalent attachment. Cy5-labeled, heat-killed cells were used to demonstrate that the immobilized magainin I can bind Salmonella with detection limits similar to analogous antibody-based assays. Detection limits for E. coli were higher than in analogous antibody-based assays, but it is expected that other AMPs may possess higher affinities for this target. The results showed that both specific and nonspecific binding strongly depend on the method used for peptide immobilization. Direct attachment of magainin to the substrate surface not only decreased nonspecific cell binding but also resulted in improved detection limits for both Salmonella and E. coli.  相似文献

Poly(ε-caprolactone)/polyvinylpyrrolidone (PCL/PVP) scaffolds with various composition were fabricated from 1,1,1,3,3,3-hexafluoro-2-propanol (HFIP) solution using the same electrospinning parameters in order to reveal the effect of polymer ratio on the material properties. The obtained materials were characterized using scanning electron microscopy, contact angle measurements, X-ray diffraction, Fourier-transformed infrared spectroscopy, and tensile testing. The strengthening effect of PVP was observed: Young modulus of PCL/PVP scaffold with 50/50 polymer ratio was found at 105.4 ± 8.4 MPa which is six times higher comparing to those of PCL scaffold. PVP-containing scaffolds were extremely hydrophilic with PVP concentration of 5 wt% (vs. 25 wt% in previous reports) leading to full wetting of the material. in vitro studies showed an improved viability of HeLa cells cultured with the composites containing higher concentrations of PVP. Owing to the application of HFIP, PCL-based materials were loaded with cyclophosphamide for the first time and the PVP-containing materials demonstrated the intensified initial release of the model compound. Utilizing HFIP for the fabrication of PCL/PVP scaffolds significantly widens their application for drug delivery systems due to a good solubility of proteins, drugs, and other biologically active compounds in this solvent.  相似文献

We report the operation of a discharge-produced argon (Ar) plasma waveguide in an alumina (Al(2)O(3)) capillary to guide a 10(16)-W/cm(2) ultrashort laser pulse for shorter wavelength light sources at high repetition rate operation. The electron density in the plasma channel was measured to be 1 × 10(18) cm(-3). Modeling with a one-dimensional magnetrohydrodynamic code was used to evaluate the degree of ionization of Ar in the preformed plasma channel. The observed spectrum of the laser pulse after propagation in the argon plasma waveguide was not modified and was well reproduced by a particle in cell simulation.  相似文献

We describe for the first time a method for synthesis of boron suboxide under vacuum from a B2O3 + B mixture. We show that at 1420°C, under vacuum a boron suboxide phase of composition B13O2 is formed. The product is a highly disperse powder which, according to electron microscopy and crystal optics analysis, consists of agglomerates of various sizes with single-crystal particles mainly no larger than 2 m. We have used differential thermal analysis for the first time to study the reaction of crystalline boron with boric anhydride under vacuum and also in helium. We have shown that the temperature of onset of the reaction (B + B2O3) depends on the heating rate of the reagent mixture, and is practically independent of the reaction medium (1420°C). Compared with the original boron, the B13O2 powder has higher chemical stability in liquid reagents and thermal stability in air for isothermal and nonisothermal heating conditions. We wetted hot-pressed B13O2 samples with iron, nickel, copper, and tin melts. We have shown that B13O2 is readily wetted by iron and nickel at temperatures of 1200-1300°C, while it is practically unwetted by copper and tin at 1200°C and 900°C.  相似文献

A two-stage pressure-and-temperature treatment of the C60 and C70 fullerites was carried out. C60 and C70 molecules collapsed at the first-stage hot-isostatic-pressing (HIP; 220 MPa, argon) and transformed into some fullerene-related form in the 900–1750°C temperature range. These materials were used at the second stage of the high-pressure-high-temperature (HPHT; 7.7 GPa/1400°C) treatment to produce the bulk samples that had: a specific weight of about 2.0 g/cm3, 40/110 GPa Young modulus, 6.0/12.5 GPa, and elastic recovery above 81%. Transformations under the treatments were investigated with the X-ray and transmission electron microscopy techniques. The mechanism of the pressure-and-temperature transformations is discussed.  相似文献

In several Russian northern lakes and rivers, Arctic cisco Coregonus autumnalis, least cisco C. sardinella, peled C. peled, tugun C. tugun, broad whitefish C. nasus, whitefish C. lavaretus and vendace C. albula were sampled in periods of officially permitted commercial fishery. Special attention was paid to contents (mg g?1 of wet weight) of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) in muscle tissues (filets), which are essential for human nutrition. The highest values of EPA + DHA content in semi‐anadromous fish and freshwater fish were recorded for C. autumnalis from the Yenisei River, 17.60 mg g?1 wet weight, and for C. lavaretus from the Sobachye Lake, 16.61 mg g?1 wet weight, respectively. Intra‐genus variations of EPA + DHA contents of Coregonus species were from 1.87 to 17.60 mg g?1 wet weight. Since the congeneric species were genetically close to each other, the variations in EPA and DHA contents were thought to be caused primarily by ecological factors: migrational capability, type of feeding and trophic status of aquatic ecosystems. In general, the majority of studied species appeared to be of a high nutritive value for humans, although unfavorable environmental conditions could considerably diminish this value.  相似文献

Interferon-β (IFN-β) is a pleiotropic cytokine used for therapy of multiple sclerosis, which is also effective in suppression of viral and bacterial infections and cancer. Recently, we reported a highly specific interaction between IFN-β and S100P lowering IFN-β cytotoxicity to cancer cells (Int J Biol Macromol. 2020; 143: 633–639). S100P is a member of large family of multifunctional Ca2+-binding proteins with cytokine-like activities. To probe selectivity of IFN-β—S100 interaction with respect to S100 proteins, we used surface plasmon resonance spectroscopy, chemical crosslinking, and crystal violet assay. Among the thirteen S100 proteins studied S100A1, S100A4, and S100A6 proteins exhibit strictly Ca2+-dependent binding to IFN-β with equilibrium dissociation constants, Kd, of 0.04–1.5 µM for their Ca2+-bound homodimeric forms. Calcium depletion abolishes the S100—IFN-β interactions. Monomerization of S100A1/A4/A6 decreases Kd values down to 0.11–1.0 nM. Interferon-α is unable of binding to the S100 proteins studied. S100A1/A4 proteins inhibit IFN-β-induced suppression of MCF-7 cells viability. The revealed direct influence of specific S100 proteins on IFN-β activity uncovers a novel regulatory role of particular S100 proteins, and opens up novel approaches to enhancement of therapeutic efficacy of IFN-β.  相似文献

We studied the fatty acid (FA) content and composition of ten zoobenthic species of several taxonomic groups from different freshwater bodies. Special attention was paid to essential polyunsaturated fatty acids, eicosapentaenoic acid (EPA, 20:5n-3), docosahexaenoic acid (DHA, 22:6n-3), and arachidonic acid (ARA, 20:4n-6); and the n-3/n-6 and DHA/ARA ratios, which are important for consumers of higher trophic levels, i.e., fish. The content and ratios of these FA varied significantly in the studied zoobenthic species, consequently, the invertebrates were of different nutritional quality for fish. Eulimnogammarus viridis (Crustacea) and Dendrocoelopsis sp. (Turbellaria) had the highest nutrition value for fish concerning the content of EPA and DHA and n-3/n-6 and DHA/ARA ratios. Using canonical correspondence analysis we compared the FA profiles of species of the studied taxa taking into account their feeding strategies and habitats. We gained evidence that feeding strategy is of importance to determine fatty acid profiles of zoobenthic species. However, the phylogenetic position of the zoobenthic species is also responsible and may result in a similar fatty acid composition even if species or populations inhabit different water bodies or have different feeding strategies.  相似文献

A comparative investigation of the induction of double-strand DNA breaks (DSBs) in the Chinese hamster V79 cells by γ-radiation at dose rates of 1, 10 and 400 mGy/min (doses ranged from 0.36 to 4.32 Gy) was performed. The acute radiation exposure at a dose rate of 400 mGy/min resulted in the linear dose-dependent increase of the γ-H2AX foci formation. The dose-response curve for the acute exposure was well described by a linear function y = 1.22 + 19.7x, where “y” is an average number of γ-H2AX foci per a cell and “x” is the absorbed dose (Gy). The dose rate reduction down to 10 mGy/min lead to a decreased number of γ-H2AX foci, as well as to a change of the dose-response relationship. Thus, the foci number up to 1.44 Gy increased and reached the “plateau” area between 1.44 and 4.32 Gy. There was only a slight increase of the γ-H2AX foci number (up to 7) in cells after the protracted exposure (up to 72 h) to ionizing radiation at a dose rate of 1 mGy/min. Similar effects of the varying dose rates were obtained when DNA damage was assessed using the comet assay. In general, our results show that the reduction of the radiation dose rate resulted in a significant decrease of DSBs per cell per an absorbed dose.  相似文献

Polytetrafluoroethylene/cellulose composite aerogels containing PTFE from 0 to 40 wt.% were obtained by drying the corresponding suspension mixtures at a temperature of 50°C. Cellulose isolated from the stem of Sosnowski hogweed was used as the main component of the aerogels. In present work, the composite aerogels were heated in an inert atmosphere at a temperature of 370°C to partially hydrophobize cellulose and remove surfactants that were present in the initial PTFE suspension. It was found that at this temperature, the thermal degradation of cellulose proceeded differently in the presence and absence of PTFE. The maximum water contact angle (156°) was obtained for the sample with 10 wt.% PTFE. Hydrophobized samples of aerogels were studied with the help of infrared spectroscopy, scanning electron microscopy, and powder x-ray diffractometry and tested as sorbents for various solvents. It was found that the volume of solvent absorbed by the aerogel, in the case of a sample with a PTFE content of 40 wt.%, can even exceed the initial volume of the absorber.  相似文献
