This work aimed to develop a calcium alginate hydrogel as a pH responsive delivery system for polymyxin B (PMX) sustained-release through the vaginal route. Two samples of sodium alginate from different suppliers were characterized. The molecular weight and M/G ratio determined were, approximately, 107?KDa and 1.93 for alginate_S and 32?KDa and 1.36 for alginate_V. Polymer rheological investigations were further performed through the preparation of hydrogels. Alginate_V was selected for subsequent incorporation of PMX due to the acquisition of pseudoplastic viscous system able to acquiring a differential structure in simulated vaginal microenvironment (pH 4.5). The PMX-loaded hydrogel (hydrogel_PMX) was engineered based on polyelectrolyte complexes (PECs) formation between alginate and PMX followed by crosslinking with calcium chloride. This system exhibited a morphology with variable pore sizes, ranging from 100 to 200?μm and adequate syringeability. The hydrogel liquid uptake ability in an acid environment was minimized by the previous PECs formation. In vitro tests evidenced the hydrogels mucoadhesiveness. PMX release was pH-dependent and the system was able to sustain the release up to 6 days. A burst release was observed at pH 7.4 and drug release was driven by an anomalous transport, as determined by the Korsmeyer–Peppas model. At pH 4.5, drug release correlated with Weibull model and drug transport was driven by Fickian diffusion. The calcium alginate hydrogels engineered by the previous formation of PECs showed to be a promising platform for sustained release of cationic drugs through vaginal administration.  相似文献

This research assessed the economic impact and construction costs of buildings with solar chimneys in Thailand, called bioclimatic houses. A solar chimney uses a combination of air gaps between walls and roof, and effective ventilation to lower the indoor temperature and reduce the need for air conditioning. The study looked at small, medium, and large size houses and studied construction techniques, the materials used, and the time needed to build a residential structure. It also polled owners of bioclimatic houses about their satisfaction. The techniques employed in building bioclimatic houses take slightly longer and cost slightly more than those used in standard construction. However, the study found that bioclimatic houses use approximately 10–20% less electricity for air conditioning require less maintenance and have a payback period of 6–13 years, depending on the size. The lifespan of insulation material used in standard houses is about 20–25 years, while the lifespan of a solar chimney is about 40–50 years. The solar chimney concept can also be applied to standard houses with minimal renovation. The study found that this type of house was suitable for use in Thailand, and with increased consumer awareness, had a high probability for adoption.  相似文献

Heat transfer enhancement of natural convection inside the inclined solar chimneys is investigated using electrohydrodynamic technique. The interactions between electric field, flow field, and temperature field are analyzed. The ranges of parameters considered are 104Ra107, 7.5 kVV017.5 kV, 30°θ120°, and 2aspect ratio14. Flow and heat transfer enhancements are significantly influenced at low Rayleigh number. The optimum inclined angle which obtains maximum volume flow rate and heat transfer is found to be at θ=60°. A maximum volume flow rate enhancement is expressed in relation with the number of electrodes. The relation between aspect ratio of chimney and number of electrodes that performs the optimum condition between efficiency and economy is analyzed incorporating with all concerning parameters.  相似文献

The interactions between electric field, flow field, and temperature field are numerically determined. Flow and heat transfer enhancements are significantly influenced at low Rayleigh number. The effect of electrode arrangement and number of electrodes to the average velocity and Nusselt number are expressed. An optimum inclined angle of the channel is recommended. Saccharomyces strains capable of fermenting maltose contain any one of five telomere-associated MAL loci. Each MAL locus is a complex of three genes encoding the three functions required to ferment maltose: maltose permease (GENE 1), maltase (GENE 2) and the MAL trans-activator (GENE 3). All five loci have been cloned and all are highly sequence homologous over at least a 9.0 kbp region containing these GENEs (Charron et al., Genetics 122, 307-331, 1989). Our initial studies of strains carrying the MAL3 locus indicated the presence of linked, repeated MAL-homologous sequences (Michels and Needleman, Mol. Gen. Genet. 191, 225-230, 1983). Here we report our analysis of the centromere-proximal MAL3-linked sequences and show that the complete MAL3 locus spans approximately 40 kbp and consists of tandemly arrayed, partial repeats of the three GENE sequences described above. In addition, the structure of the MAL3 locus is compared to that of three partially functional alleles of MAL3. These alleles were shown to contain only MAL31 and MAL32 and their structure suggests that they resulted from MAL3 deletions removing the sequences centromere-proximal to MAL31. The amplification and rearrangement of the telomere-linked MAL3 sequences are discussed in the context of studies on other telemere-associated sequences from yeast and other species.  相似文献

A high‐content screening method to characterize multifunctional multilayer films that combine mechanical adhesion and favorable biological response is reported. Distinct combinations of nanostructured films are produced using layer‐by‐layer methodology and their morphological, physicochemical, and biological properties are analyzed in a single microarray chip. Inspired by the composition of the adhesive proteins in mussels, thin films containing dopamine‐modified hyaluronic acid are studied. Flat biomimetic superhydrophobic patterned chips produced by a bench‐top methodology are used for the build‐up of arrays of multilayer films. The wettability contrasts imprinted onto the chips are allowed to produce individual, position controlled, multilayer films in the wettable regions. The flat configuration of the chip permits to perform a series of nondestructive measurements directly on the individual spots. In situ adhesion properties are directly measured in each spot, showing that nanostructured films richer in dopamine promote the adhesion. In vitro tests show an enhanced cell adhesion for the films with more catechol groups. The advantages presented by this platform include ability to control the uniformity and size of the multilayers films, its suitability to be used as a new low cost toolbox and for high‐content cellular screening, and capability for monitoring in situ a variety of distinct material properties.  相似文献
