Cytoskeleton localization in the sperm head prior to fertilization   总被引:2,自引:0,他引:2  
Three major cytoskeletal proteins, actin, tubulin and spectrin, are present in the head of mammalian spermatozoa. Although cytoskeletal proteins are implicated in the regulation of capacitation and the acrosome reaction (AR), their exact role remains poorly understood. The aim of this study was to compare the distribution of the sperm head cytoskeleton before and after the AR in spermatozoa representing a range of acrosome size and shape. Spermatozoa from the human and three rodents (rat, hamster and grey squirrel) were fixed before and after the AR in appropriate medium in vitro. Indirect immunofluorescent localization of cytoskeletal proteins was undertaken with antibodies recognizing actin, spectrin and alpha-tubulin. Preparations were counterstained with propidium iodide and examined by epifluorescent and confocal microscopy. Our results clearly demonstrated changes in localization of cytoskeleton during the AR, mainly in the apical acrosome with further changes to the equatorial segment and post-acrosomal regions. The pattern of cytoskeletal proteins in the sperm head of all the species was similar in respect to various sub-compartments. These observations indicated that the sperm head cortical cytoskeleton exhibits significant changes during the AR and, therefore, support the image of cytoskeletal proteins as highly dynamic structures participating actively in processes prior to fertilization.  相似文献

The cellular and subcellular organization of thyroid follicular cells in peripubertal (6-month-old) male pigs treated with estradiol-dipropionate (Oe) plus progesterone (Pr) in combination on the first postnatal day was studied. A stereological method was used for morphometric determinations of the relative volume densities of the follicular epithelium, colloid and interstitium, and for establishing the epithelial height and index of activation rate. Statistically significant differences of the examined parameters between the control and Oe+Pr -treated groups were determined by Student's t-test. The subcellular organization of thyroid follicular cells was studied using transmission electron microscopy (TEM). When compared with the control group, in the Oe+Pr -treated pigs, thyroid follicles possessed a noticeably higher follicular epithelium when compared with the control animals. The observed changes were quantified and comparison between the experimental groups showed that the height, volume density of follicular epithelium, and index of activation rate were significantly (P < 0.05) increased, whereas the relative volume density of the colloid was significantly (P < 0.05) decreased. At the subcellular level the thyroid follicular cells of Oe+Pr -treated pigs were characterized by increased number of polysomes and dense bodies and extensive endoplasmatic reticulum. It was concluded that a single neonatal treatment with female gonadal steroids exerted a prolonged effect on the pig's thyroid, characterized by increased biosynthesis and reabsorption of the colloid by the follicular cells.  相似文献

Causes and effects of a few real faults in a hydronic heating system are explained in this paper. Since building energy management system (BEMS) has to be utilized in fault detection and diagnosis (FDD), practical explanations of faults and their related effects are important to building caretakers. A simple heat balance model is used in this study. The model is calibrated using the optimization tool. Site data from the BEMS of a real building are calibrated against the model. Desired and real data are compared, so that the effects of the following faults are analyzed: faults in an outdoor air temperature sensor, fault in the time schedule, and a water flow imbalance problem. This paper presents an overview of the real causes of the faults and their effects both on the energy consumption and on the indoor air temperature. In addition, simple instructions for the building caretakers for fault detection in the hydronic heating systems are given.  相似文献

The aim of the study was to develop improved measurements on heat pump performance. A model-based approach was combined with data fusion technique to estimate performance of a heat pump. These improved heat pump performance measurements implied fused measurement between direct and indirect measurements of compressor power and condenser load. Developed models of compressor power and condenser load used input data from building energy management system to produce measurements on the heat pump performance. The direct measurements were obtained by using the temperature and pressure measurements, while the indirect measurements were obtained using the electrical signal of the heat pump part load. The results showed a big need for use of different data sources to define real energy performance metrics. Further, the analysis on obtained measurements showed that indirect and fused measurements were more reliable than only direct measurement, particularly for the condenser load. Use of only direct measurements could result in higher estimation of electricity and heating energy consumption than the real consumption is. The analysis of fused measurements on the heat pump performance showed that such improved measurement could enhance heat pump performance verification.  相似文献

This paper reviews the applicability and drawbacks of available European and international standards related to lifetime commissioning, by structuring them into Norwegian commissioning procedures. The work describes research on lifetime commissioning that proposes a generic framework on building performances. The generic framework describes a component in HVAC system by performances. The results of the standard review show that there is a need for measurement and testing standards in hydronic systems. In addition, there is no generic framework on the definition of energy-efficiency measure in the reviewed standards. Findings on a case study, where lifetime commissioning procedures were tested, are presented. After the performances of the case study were defined by the generic framework, it was found that 20% of all the performances can be monitored by a building energy management system. Due to good operation, the building managed to achieve 3% lower energy consumption than design a year after the building was taken in use. In addition, results show that energy signature curves can be used only in a modified way to predict heating consumption, while electricity consumption cannot be described in that way.  相似文献

Traditional fermented foods are the best source for the isolation of strains with specific traits to act as functional starters and to keep the biodiversity of the culture collections. Besides, these strains could be used in the formulation of foods claimed to promote health benefits, i.e. those containing probiotic microorganisms. For the rational selection of strains acting as probiotics, several in vitro tests have been proposed. In the current study, we have characterized the probiotic potential of the strain Lactobacillus paraplantarum BGCG11, isolated from a Serbian soft, white, homemade cheese, which is able to produce a "ropy" exopolysaccharide (EPS). Three novobiocin derivative strains, which have lost the ropy phenotype, were characterized as well in order to determine the putative role of the EPS in the probiotic potential. Under chemically gastrointestinal conditions, all strains were able to survive around 1-2% (10(6)-10(7)cfu/ml cultivable bacteria) only when they were included in a food matrix (1% skimmed milk). The strains were more resistant to acid conditions than to bile salts and gastric or pancreatic enzymes, which could be due to a pre-adaptation of the parental strain to acidic conditions in the cheese habitat. The ropy EPS did not improve the survival of the producing strain. On the contrary, the presence of an EPS layer surrounding the strain BGCG11 hindered its adhesion to the three epithelial intestinal cell lines tested, since the adhesion of the three non-ropy derivatives was higher than the parental one and also than that of the reference strain Lactobacillus rhamnosus GG. Aiming to propose a potential target application of these strains as probiotics, the cytokine production of peripheral blood mononuclear cells (PBMC) was analyzed. The EPS-producing L. paraplantarum BGCG11 strain showed an anti-inflammatory or immunosuppressor profile whereas the non-ropy derivative strains induced higher pro-inflammatory response. In addition, when PBMC were stimulated with increasing concentrations of the purified ropy EPS (1, 10 and 100μg/ml) the cytokine profile was similar to that obtained with the EPS-producing lactobacilli, therefore pointing to a putative role of this biopolymer in its immune response.  相似文献

In order for mammalian sperm to obtain a fertilizing ability, they must undergo a complex of molecular changes, called capacitation. During capacitation, steroidal compounds can exert a fast nongenomic response in sperm through their interaction with plasma membrane receptors, and activate crucial signaling pathways leading to time-dependent protein tyrosine phosphorylation (TyrP). Estrogen receptor beta was detected in epididymal mouse sperm; therefore, the effect of 17B-estradiol, estrone, estriol, and 17A-ethynylestradiol on mouse sperm capacitation in vitro was investigated. The effect was evaluated by positive TyrP in sperm heads and in the whole sperm lysates. Simultaneously, the state of the acrosome after the calcium ionophore-induced acrosome reaction was assessed. Generally, estrogens displayed a time and concentration-dependent stimulatory effect on sperm TyrP during capacitation. In contrast, the number of sperm that underwent the acrosome reaction was lower in the experimental groups. It has been demonstrated that both natural and synthetic estrogens can modify the physiological progress of mouse sperm capacitation. The potential risk in the procapacitation effect of estrogens can also be seen in the decreased ability of sperm to undergo the acrosome reaction. In conclusion, the capacitating ability of sperm can be significantly lowered by increasing the level of estrogens in the environment.  相似文献

Crosslinking enzymes are frequently used in bioprocessing of dairy products. The aim of this study was to examine the effects of enzymatic crosslinking on IgE binding, allergenicity and digestion stability of β‐casein (CN). β‐CN was crosslinked by transglutaminase, tyrosinase, mushroom tyrosinase/caffeic acid and laccase/caffeic acid. The IgE binding to β‐CN was compared in vitro by CAP inhibition assay, ELISA inhibition as well as ex vivo by basophil activation assay. Crosslinked CNs were digested by simulated gastric fluid for 15 and 60 min and obtained digests analyzed for their ability to inhibit IgE binding by CAP inhibition assay and SDS‐PAGE. The ability of crosslinked CNs to activate basophils was significantly reduced in seven patients in the case of CN crosslinked by laccase and moderately reduced in the case of tyrosinase/caffeic acid crosslinked CN (in two cow's milk allergy patients tested with different allergen concentrations). The response to various crosslinked CNs differed individually among patients' sera tested by ELISA inhibition assay. The presence of caffeic acid hampered digestion by pepsin, and this effect was most pronounced for the tyrosinase/caffeic acid crosslinked CN. The laccase/caffeic acid and mushroom tyrosinase/caffeic acid had the highest potential in mitigating IgE binding and allergenicity of the β‐CN out of all investigated enzymes. BACKGROUND: The major peanut allergens are Ara h 1, Ara h 2 and Ara h 6. Proteolytic processing has been shown to be required for the maturation process of Ara h 6. The aim of this study was to examine whether Ara h 2 undergoes proteolytic processing and, if so, whether proteolytic processing influences its ability to bind human immunoglobulin E (IgE). RESULTS: Ara h 2 isolated from peanut extract under conditions of protease inhibition revealed a single additional peak for its two known isoforms (Ara h 2.01 and Ara h 2.02), corresponding to a C‐terminally truncated form lacking a dipeptide (RY). Ara h 2 isolated in the absence of protease inhibition, however, yielded two additional peaks, identified as C‐terminally truncated forms lacking either a dipeptide (RY) or a single tyrosine residue. The IgE‐binding capacity of the Ara h 2 truncated forms was not altered. CONCLUSION: Ara h 2 undergoes proteolytic processing by peanut proteases that involves C‐terminal removal of a dipeptide. Hence Ara h 2 isolated from peanut extract is a complex mixture of two isoforms expressed by different genes, Ara h 2.01 and Ara h 2.02, as well as truncated forms generated by the proteolytic processing of these isoforms. In this paper, the authors present a coupled thermal and electromechanical model for squirrel-cage induction motor simulation and analyses. The effect of iron saturation, rotor parameter variation due to current displacement (skin-effect) and temperature variations of both stator and rotor resistances are taking into account simultaneously. A new device for the temperature measurement in the stationary as well as rotating parts of electric machines is built. The unique construction of the device for continuous rotor temperature monitoring is briefly described. This device is a part of a complete acquisition system, which is used for precise testing and investigating of a coupled thermal and electromechanical phenomena. An original thermal observer based on the direct stator temperature measurement under normal running condition with no additional sensors is proposed  相似文献
