By its functional interaction with a RecA polymer, the mutagenic UmuD'C complex possesses an antirecombination activity. We show here that MucA'B, a functional homolog of the UmuD'C complex, inhibits homologous recombination as well. In F- recipients expressing MucA'B from a Ptac promoter, Hfr x F- recombination decreased with increasing MucA'B concentrations down to 50-fold. In damage-induced pKM101-containing cells expressing MucA'B from the native promoter, recombination between a UV-damaged F lac plasmid and homologous chromosomal DNA decreased 10-fold. Overexpression of MucA'B together with UmuD'C resulted in a synergistic inhibition of recombination. RecA[UmuR] proteins, which are resistant to UmuD'C inhibition of recombination, are inhibited by MucA'B while promoting MucA'B-promoted mutagenesis efficiently. The data suggest that MucA'B and UmuD'C contact a RecA polymer at distinct sites. The MucA'B complex was more active than UmuD'C in promoting UV mutagenesis, yet it did not inhibit recombination more than UmuD'C does. The enhanced mutagenic potential of MucA'B may result from its inherent superior capacity to assist DNA polymerase in trans-lesion synthesis. In the course of this work, we found that the natural plasmid pKM101 expresses around 45,000 MucA and 13,000 MucB molecules per lexA(Def) cell devoid of LexA. These molecular Muc concentrations are far above those of the chromosomally encoded Umu counterparts. Plasmid pKM101 belongs to a family of broad-host-range conjugative plasmids. The elevated levels of the Muc proteins might be required for successful installation of pKM101-like plasmids into a variety of host cells.  相似文献

The Bayesian approach to uncertainty evaluation is a classical example of the fusion of information from different sources. Basically, it is founded on both the knowledge about the measurement process and the influencing quantities and parameters. The knowledge about the measurement process is primarily represented by the so-called model equation, which forms the basic relationship for the fusion of all involved quantities. The knowledge about the influencing quantities and parameters is expressed by their degree of belief, i.e. appropriate probability density functions that usually are obtained by utilizing the principle of maximum information entropy and the Bayes theorem. Practically, the Bayesian approach to uncertainty evaluation is put into effect by employing numerical integration techniques, preferably Monte-Carlo methods. Compared to the ISO-GUM procedure, the Bayesian approach does not have any restrictions with respect to nonlinearities and calculation of confidence intervals.  相似文献

The authors investigated effects of task and overlapping processing load on semantic processing. In 3 experiments the brain potential component N400 was elicited by synonymous and nonsynonymous spoken noun pairs that were to be classified according to semantic relatedness. The time course of the N400 component to the nouns was delayed, and its amplitude was reduced when additional tasks had to be performed. The delay increased with temporal overlap and was more pronounced for language-related than for spatial additional tasks. Delays of N400 were also caused by a compatibility manipulation in the additional tasks. Theoretical accounts of the observed interference of overlapping tasks with language perception in terms of attentional shifts were discarded. Today's civil engineering NDT needs are for quantitative, fast to apply and easy to interpret methods. They are required on site both for evaluation of existing structures/infrastructure and for quality control in new construction. With this purpose, BAM has carried out in-situ feasibility studies, using radar, impact-echo and ultrasonic-echo methods. Two examples are given of their combined application on different railway slab–track constructions, using high frequency radar, scanning impact-echo and ultrasonic array methods. The aim was to evaluate limitations and advantages, comparability and complementarity of these methods. The on-site measurements demonstrated the possibility to investigate, with good accuracy via NDT, the bonding conditions between sleepers and slab, to locate voids and measure layer thickness. The acoustic and electromagnetic methods used, were found to be complementary.  相似文献

While the quality and variety of particle size measuring instruments have improved dramatically in recent years, both in their accuracy and in their speed of measurement, the representation of the measured data — particularly the density distribution — has generally received little attention from the manufacturers. The depiction of a continuous density distribution by a percentage value on the ordinate makes no sense. Although logarithmic density distributions serve a purpose with distinctly separated bi‐ or multimodal distributions (by analogy with analytical chemical spectra), they can lead to serious interpretation errors in the case of the monomodal distribution. Thus for monomodal distributions either the density distribution qr(x) should be used or no representation of the density distribution attempted.  相似文献

Biogenic emissions of methane (CH4) and nitrous oxide (N2O) from animal manure are stimulated by the degradation of volatile solids (VS) which serves as an energy source and a sink for atmospheric oxygen. Algorithms are presented which link carbon and nitrogen turnover in a dynamic prediction of CH4 and N2O emissions during handling and use of liquid manure (slurry). A sub-model for CH4 emissions during storage relates CH4 emissions to VS, temperature and storage time, and estimates the reduction in VS. A second sub-model estimates N2O emissions from field-applied slurry as a function of VS, slurry N and soil water potential, but emissions are estimated using emission factors. The model indicated that daily flushing of slurry from cattle houses would reduce total annual CH4 + N2O emissions by 35% (CO2 eq.), and that cooling of pig slurry in-house would reduce total annual CH4 + N2O emissions by 21% (CO2 eq.). Anaerobic digestion of slurry and organic waste produces CH4 at the expense of VS. Accordingly, the model predicted a 90% reduction of CH4 emissions from outside stores with digested slurry, and a >50% reduction of N2O emissions after spring application of digested as opposed to untreated slurry. The sensitivity of the model towards storage temperature and soil water potential was examined. This study indicates that simple algorithms to account for ambient climatic conditions may significantly improve the prediction of CH4 and N2O emissions from animal manure.  相似文献
