Detailed behaviors of ignition kernel(s) in a uniform stoichiometric methane/air mixture under the temperature gradient were investigated numerically by using a fundamental system of microcombustion. Bifurcation of the heat release rate peak in an ignition phase at the high wall temperature side, which has been observed in previous experimental and theoretical studies, was successfully reproduced by the present computation. The bifurcated heat release rate peak exhibited negative propagation speed relative to the local flow velocity by consuming a separated methane/air mixture in the downstream side of the boundary zone between an incoming fresh mixture and burned gas. CH4 was completely consumed at the main peak, whereas CO remained unreacted in the wide region behind the main peak. In a weak reaction phase at the low wall temperature side, two bifurcations of heat release rate peak were newly captured. By the two bifurcations, three heat release rate peaks, namely, a main and two bifurcated peaks appeared. The two bifurcations were caused by remaining intermediates such as CH3, CO, H, and OH in the downstream side of the boundary zone. The main and one bifurcated peak disappeared, whereas the other bifurcated peak remained and flowed downstream. The main and two bifurcated peaks exhibited negative propagation speeds relative to local flow velocity by consuming the remaining intermediates. CO which formed in the middle of the boundary zone in the weak reaction phase remained unreacted and kept on flowing downstream, but did not flow out since the next cycle of the ignition phase was initiated there.  相似文献

A novel all-optical modulation-format conversion from nonreturn-to-zero on-off keying (NRZ-OOK) to return-to-zero binary phase-shift keying (RZ-BPSK) is proposed. A Mach-Zehnder interferometric (MZI) wavelength converter consisting of integrated semiconductor optical amplifiers (SOAs) is utilized to implement the proposed format conversion. Error-free operation at a bit rate of 10.7 Gb/s is experimentally demonstrated in order to show the feasibility of the proposed scheme. The received sensitivity of the converted signal is improved by 2.9 dB compared with a back-to-back NRZ-OOK signal at BER=10/sup -9/. In addition, a reasonable dispersion tolerance of the converted signal up to /spl plusmn/500 ps/nm is observed. The numerical simulation based upon the carrier-rate equation verifies the experimental results.  相似文献

An affinity purified antibody specific for midkine (MK) stained senile plaques in the brain of Alzheimer disease (AD) patients. After formic acid treatment, plaque staining was dramatically enhanced, and almost all beta-amyloid protein (BAP) deposits were also immunoreactive for MK. MK-immunoreactivity was not observed in normal cellular components nor in other pathological lesions including tangles in AD brain. Control brain sections were not immunoreactive for MK. The presence of MK in AD brain but not in control brain was confirmed by Western blotting. MK appears to be involved in the pathological process leading to senile plaque formation.  相似文献

Bone marrow specimens obtained from 54 patients with de novo AML and 7 patients with AML evolving from MDS were retrospectively examined for the presence of micromegakaryocytes defined as cells of less than 30 microns in diameter with one or two nuclei. At least 25 megakaryocytes were counted in each patient. Micromegakaryocytes were found in 17 cases (31%), M1:1/11, M2:5/18, M3:0/4, M4:5/12, M5:1/4, M6:4/4, M7:1/1. The median age of the patients was higher in de novo AML with micromegakaryocytes (57 years) than in de novo AML without micromegakaryocytes (41 years) (p = 0.014). Chromosomal analysis revealed that deletion of 5 or 5q-, 7 or 7q- was recognized only in the group of de novo AML with micromegakaryocytes and that t(15;17), t(8:21) and inv (16) were not recognized in this group. Micromegakaryocytes were identified in each bone marrow specimen obtained from 9 of 10 patients with de novo AML with trilineage myelodysplasia. The complete remission rate was significantly lower in de novo AML with micromegakaryocytes (33%) than in de novo AML without micromegakaryocytes (86%) (p = 0.001). The duration of survival of the patients with de novo AML with micromegakaryocytes was shown to be shorter than that of the patients with de novo AML without micromegakaryocytes (p = 0.017). Micromegakaryocytes were recognized in all of 7 patients with AML evolving from MDS. The presence of micromegakaryocytes in bone marrow of the patients with AML indicates a subset of AML with poor prognosis that may be closely associated with myelodysplastic syndrome.  相似文献

Several distinct Ras GTPase activating proteins (GAPs) from mammals, including Ras GAP of 120 kDa (GAP1) and NF1, stimulate the intrinsic GTPase activity of normal Ras, but not oncogenic Ras mutants (Trahey and McCormick, 1987). That is the reason why normal Ras remains predominantly in the inactive GDP-bound form (D-Ras), whereas oncogenic Ras remains constitutively in the active GTP-bound form (T-Ras). NF1 is a tumor suppressor of 2818 amino acids whose disruption or deletion causes brain tumors called neurofibromatosis type 1 by elevating the T-Ras level. T-Ras activates several distinct oncogenic effectors, including Ser/Thr kinase Raf, GAP1, P1-3 kinase, PKC-zeta and Ra1 GDS. Interestingly, the binding of T-Ras to either GAPs or these oncogenic effectors requires the same effector domain I (residues 32-40) of T-Ras molecule. In other words, these GAPs and effectors compete for binding to T-Ras. Using a series of N- and C-terminal deletion mutants of NF1, we identified a 78 amino acid fragment (NF78, residues 1441-1518) as the minimum GAP domain, and a 56 amino acid fragment (NF 56, residues 1441-1496) as the minimum Ras-binding domain. Furthermore, we identified the Raf fragment of 81 amino acids (Raf81, residues, 51-131) as the minimum Ras-binding domain with a high affinity. We found that (i) these NF1 fragments and Raf81 compete for binding to T-Ras, and that (ii) over-expression of these NF1 or Raf fragments strongly suppresses the malignant transformation caused by oncogenic Ras mutants. Thus, these agents offer a unique opportunity to control the proliferation of T-Ras-associated tumors that represent more than 30% of all human carcinomas including neurofibromatosis type 1.  相似文献

A possibility of simultaneous formation of two chemical reaction fronts during nonstationary combustion of a gas in a microchannel with a temperature gradient in the walls is demonstrated. Combustion in a straight tube and in a gap between two disks with radial fuel injection is considered. In both cases, the characteristic transverse size of the channel is smaller than the critical diameter determined for the ambient temperature, and gas combustion occurs in the region where the wall temperature is higher than the ambient temperature. A numerical study of flame repetitive extinction/ignition (FREI) demonstrated a possibility of simultaneous formation of two chemical reaction fronts in the hot region of the channel. One front corresponds to conventional flame propagating upstream from the hot to the cold part of the channel, and the other front moves in the downstream direction and decays as the fuel burns out. A dynamic routing and wavelength allocation technique with an interplay between physical and network layer parameters encompassing Four-wave mixing (FWM) awareness and teletraffic performance of wavelength-routed optical networks has previously been proposed for a distributed approach. In this article, we present a fast computational algorithm for our routing and wavelength assignment (RWA) encompassing FWM-induced crosstalk. The objective is to minimize the time of establishing a dynamic lightpath. For this purpose, a precomputed matrix of FWM crosstalk products is used in an adapted version of the FWM-aware dynamic RWA algorithm. The approach is validated through simulations showing improvement up to 30–50% on the provisioning time of lightpaths for different network topologies compared to an online full computational scheme.

The Kerr effect in fiber is expected to play a major role in ultrafast all-optical signal processing, which can overcome the obstacles of inherently limited operating speed of electronic devices. In this paper, we propose novel ultrafast and polarization-independent all-optical analog-to-digital conversion schemes. The proposed schemes are realized by filtering the broadened and split spectrum induced by the soliton effect or self-phase modulation in fiber with an arrayed waveguide grating. We conducted proof-of-principle experiments and successfully demonstrated two-bit all-optical analog-to-digital conversions with Gray code output, which shows the feasibility of the proposed scheme.  相似文献

Silk fibroin exhibits high biocompatibility and biodegradability, making it a versatile biomaterial for medical applications. However, contaminated silkworm-derived substances in remnant sericin from the filature and degumming process can result in undesired immune reactions and silk allergy, limiting the widespread use of fibroin. Here, we established transgenic silkworms with modified middle silk glands, in which sericin expression was repressed by the ectopic expression of cabbage butterfly-derived cytotoxin pierisin-1A, to produce cocoons composed solely of fibroin. Intact, nondegraded fibroin can be prepared from the transgenic cocoons without the need for sericin removal by the filature and degumming steps that cause fibroin degradation. A wide-angle X-ray diffraction analysis revealed low crystallinity in the transgenic cocoons. However, nondegraded fibroin obtained from transgenic cocoons enabled the formation of fibroin sponges with varying densities by using 1–5% (v/v) alcohol. The effective chondrogenic differentiation of ATDC5 cells was induced following their cultivation on substrates coated with intact fibroin. Our results showed that intact, allergen-free fibroin can be obtained from transgenic cocoons without the need for sericin removal, providing a method to produce fibroin-based materials with high biocompatibility for biomedical uses.  相似文献

B16-F1 melanoma cells have often been used as a model to investigate melanogenesis, but the evidence that melanosome biogenesis and transport occur by the same mechanisms in normal melanocytes and B16-F1 cells is insufficient. In this study, we established knockout B16-F1 cells for each of several key factors in melanogenesis, i.e., tyrosinase (Tyr), Hps4, Rab27A, and Rab32·Rab38 (Rab32/38), and then compared their phenotypes with the phenotypes of corresponding mutant mouse melanocyte cell lines, i.e., melan-c, melan-le, melan-ash, and Rab32-deficient melan-cht cells, respectively. The results showed that Tyr and Rab27A are also indispensable for melanin synthesis and peripheral melanosome distribution, respectively, in B16-F1 cells, but that Hps4 or its downstream targets Rab32/38 are not essential for Tyr transport in B16-F1 cells, suggesting the existence of a Rab32/38-independent Tyr transport mechanism in B16-F1 cells. We then performed comprehensive knockdown screening of Rab small GTPases and identified Rab10 and Rab24, previously uncharacterized Rabs in melanocytes, as being involved in Tyr transport under Rab32/38-null conditions. Our findings indicate a difference between the Tyr transport mechanism in melanocytes and B16-F1 cells in terms of Rab32/38-dependency and a limitation in regard to using melanoma cells as a model for melanocytes, especially when investigating the mechanism of endosomal Tyr transport.  相似文献
