Arbequina table olives are processed as “naturally green olives”, they are directly placed in brine and fermentation starts spontaneously. Olives are harvested just before they change to ‘turning colour’. Different salt concentrations are used depending on the producer. The aim of the study was to evaluate how (i) the ripeness of the olive when it is harvested and (ii) the salt concentration of the brine influence the different microorganism populations in brine during the fermentation of Arbequina table olives.The results showed that the Enterobacteriaceae population lasted longer in black and turning colour olives than in green olives, whereas the growth of lactic acid bacteria was delayed in green olives. A higher salt concentration favoured the elimination of Enterobacteriaceae and hindered yeast growth. The main yeast species identified were Pichia anomala, Candida sorbosa and Candida boidinii, while Lactobacillus plantarum was the only lactic acid bacteria species involved in the process. In a sensory test, panellists preferred green olives and were not able to tell the laboratory-scale processed olives from a commercial sample, nor could they distinguish green olives from different brines.  相似文献

This study aimed at using essential oil (EO) alone or combined EO with nisin and γ‐irradiation to control Salmonella Typhimurium during the refrigerated storage of mini‐carrots. Peeled mini‐carrots were inoculated with S. Typhimurium at a final concentration of approximately 7 log CFU/g. Inoculated samples were coated by 5 different coating solutions: (i) nisin solution at final concentration of 103 IU/mL; (ii) mountain savory EO solution at 0.35%; (iii) carvacrol solution at 0.35%; (iv) mountain savory EO at 0.35% plus nisin solution of 103 IU/mL; or (v) carvacrol at 0.35% plus nisin solution of 103 IU/mL. Coated mini‐carrots were then irradiated at 0.5 or 1.0 kGy and compared to an unirradiated control sample. Samples were kept at 4 °C and microbial analyses were conducted at days 1, 3, 6, and 9. The results showed that mini‐carrots coated by carvacrol plus nisin solution or mountain savory EO plus nisin solution in combination with irradiation at 1.0 kGy completely eliminated S. Typhimurium to under the detection limit during the storage. Thus, the combined treatments using carvacrol plus nisin or mountain savory EO plus nisin coating solution and irradiation at 1.0 kGy could be used as an effective method for controlling S. Typhimurium in mini‐carrots.  相似文献

Most studies dealing with greenhouse gas (GHG) emissions from large boreal reservoirs were conducted during the ice‐free period. In this paper, the potential methane (CH4) and carbon dioxide emissions are estimated for two hydroelectric reservoirs, as well as for a small experimental reservoir from boreal latitudes (northern Quebec, Canada) at the ice break‐up event through diffusion (diffusive fluxes) and release of bubbles (bubbling fluxes). The results of this preliminary study suggest that the winter diffusive fluxes at the air–water interface of the sampled reservoirs represent < 7% of their cumulative carbon emissions during the ice‐free period. Furthermore, the release upon ice‐break of CH4 bubbles accumulated under the ice cover during the winter could represent  2% of the summer carbon emissions from hydroelectric reservoirs in northern Quebec. The results presented herein suggest that the GHG emissions upon ice break‐up from the boreal reservoirs investigated are a small, but non‐negligible, component of their annual GHG emissions.  相似文献

We have previously showed that plasma membrane cholesterol and GM1 ganglioside content are responsible for the opposite sensitivity of mouse leukemic T cells to ATP. We also reported that the sensitivity of CD4+ and CD8+ T cells to ATP depends on their stage of differentiation. Here, we show that CD4+ and CD8+ T cells from B6 mice express different levels of membrane GM1 and P2X7 but similar levels of cholesterol. Thus, in CD4+ T cells, membrane cholesterol content negatively correlated with ATP/P2X7-induced CD62L shedding but positively correlated with pore formation, phosphatidylserine externalization, and cell death. By contrast, in CD8+ T cells, cholesterol, GM1, and P2X7 levels negatively correlated with all these ATP/P2X7-induced cellular responses. The relationship between cholesterol and P2X7-induced cellular responses was confirmed by modulating cholesterol levels either ex vivo or through a high-fat diet. Membrane cholesterol enrichment ex vivo led to a significant reduction in all P2X7-induced cellular responses in T cells. Importantly, diet-induced hypercholesterolemia in B6 mice was also associated with decreased sensitivity to ATP in CD4+ and CD8+ T cells, highlighting the relationship between cholesterol intake and the amplitudes of P2X7-induced cellular responses in T cells.  相似文献

Abstract— In this paper we explain how macro‐cells (real PDP cells scaled‐up a hundred times) with external and removable electrodes have been validated by comparison with real panels and modeling and used to optimize the luminous efficacy of real PDPs. We illustrate the application of the macroscopic PDP tool to optimize the electrode configuration of short‐gap discharges towards higher luminous efficacy, as well as its use in conjunction with 2D and 3D modeling to lower the operating voltages of high‐efficacy long‐gap discharges triggered by auxiliary electrodes.  相似文献

Cutinase from the fungus Fusarium solani pisi is a lipolytic enzyme able to hydrolyze both aggregated and soluble substrates. It therefore provides a powerful tool for probing the mechanisms underlying lipid hydrolysis. Lipolytic enzymes have a catalytic machinery similar to those present in serine proteinases. It is characterized by the triad Ser, His, and Asp (Glu) residues, by an oxyanion binding site that stabilizes the transition state via hydrogen bonds with two main chain amide groups, and possibly by other determinants. It has been suggested on the basis of a covalently bond inhibitor that the cutinase oxyanion hole may consist not only of two main chain amide groups but also of the Ser42 O gamma side chain. Among the esterases and the serine and the cysteine proteases, only Streptomyces scabies esterase, subtilisin, and papain, respectively, have a side chain residue which is involved in the oxyanion hole formation. The position of the cutinase Ser42 side chain is structurally conserved in Rhizomucor miehei lipase with Ser82 O gamma, in Rhizopus delemar lipase with Thr83 O gamma 1, and in Candida antartica B lipase with Thr40 O gamma 1. To evaluate the increase in the tetrahedral intermediate stability provided by Ser42 O gamma, we mutated Ser42 into Ala. Furthermore, since the proper orientation of Ser42 O gamma is directed by Asn84, we mutated Asn84 into Ala, Leu, Asp, and Trp, respectively, to investigate the contribution of this indirect interaction to the stabilization of the oxyanion hole. The S42A mutation resulted in a drastic decrease in the activity (450-fold) without significantly perturbing the three-dimensional structure. The N84A and N84L mutations had milder kinetic effects and did not disrupt the structure of the active site, whereas the N84W and N84D mutations abolished the enzymatic activity due to drastic steric and electrostatic effects, respectively.  相似文献

Nickel and nickel-phosphorous matrix composite coatings reinforced by TiO2, SiC and WC particles were produced under direct and pulse current conditions from an additive-free Watts' type bath. The influence of the variable electrolysis parameters (type of current, frequency of current pulses and current density) and the reinforcing particles properties (type, size and concentration in the bath) on the surface morphology and the structure of the deposits was examined. It is demonstrated that the embedding of ceramic particles modifies in various ways the nickel electrocrystallisation process. On the other hand, Ni-P amorphous matrix is not affected by the occlusion of the particles. Overall, the imposition of pulse current conditions leads to composite coatings with increased embedded percentage and more homogenous distribution of particles in the matrix than coatings produced under direct current regime.  相似文献

Transient liquid phase bonding processes have been performed to join two carbon steel tubes using Fe96.2B3.8 wt% amorphous ribbons as interlayers. Welding experiments were performed at the temperature T ≈ 1,250 °C for different durations and under pressures of 2, 3 and 4 MPa. From metallographic inspection it is concluded that the bonding process ends in 7.0 min if a pressure of 4 MPa is applied whereas the process results incomplete if less pressure is applied. A 1D model using a finite element method has been developed for the simulation of a transient liquid phase bonding process. This model was applied to the bonding of steel/Fe–B glassy metals/steel. The computed results allow us to understand the role played by the variables involved in the bonding process.  相似文献

The synthesis, crystal structure, electrochemical and optical properties of a novel blue emitting ladder pentaphenylene are reported and compared with those of its 3π−2 spiro parent, in order to study the effect of the spiro-linked fluorene rings. Anodic oxidation of the novel compound results via electropolymerization, in the formation of an electroactive material that displays interesting electrochemical and optical properties.  相似文献
