Nuclear magnetic resonance therapy (NMRT) is discussed as a participant in repair processes regarding cartilage and as an influence in pain signaling. To substantiate the application of NMRT, the underlying mechanisms at the cellular level were studied. In this study microRNA (miR) was extracted from human primary healthy and osteoarthritis (OA) chondrocytes after NMR treatment and was sequenced by the Ion PI Hi-Q™ Sequencing 200 system. In addition, T/C-28a2 chondrocytes grown under hypoxic conditions were studied for IL-1β induced changes in expression on RNA and protein level. HDAC activity an NAD(+)/NADH was measured by luminescence detection. In OA chondrocytes miR-106a, miR-27a, miR-34b, miR-365a and miR-424 were downregulated. This downregulation was reversed by NMRT. miR-365a-5p is known to directly target HDAC and NF-ĸB, and a decrease in HDAC activity by NMRT was detected. NAD+/NADH was reduced by NMR treatment in OA chondrocytes. Under hypoxic conditions NMRT changed the expression profile of HIF1, HIF2, IGF2, MMP3, MMP13, and RUNX1. We conclude that NMRT changes the miR profile and modulates the HDAC and the NAD(+)/NADH signaling in human chondrocytes. These findings underline once more that NMRT counteracts IL-1β induced changes by reducing catabolic effects, thereby decreasing inflammatory mechanisms under OA by changing NF-ĸB signaling.  相似文献

Pompe disease is an autosomal recessive disorder caused by a deficiency in the enzyme acid alpha-glucosidase. The late-onset form of Pompe disease (LOPD) is characterized by a slowly progressing proximal muscle weakness, often involving respiratory muscles. In LOPD, the levels of GAA enzyme activity and the severity of the clinical pictures may be highly variable among individuals, even in those who harbour the same combination of GAA mutations. The result is an unpredictable genotype–phenotype correlation. The purpose of this study was to identify the genetic factors responsible for the progression, severity and drug response in LOPD. We report here on a detailed clinical, morphological and genetic study, including a whole exome sequencing (WES) analysis of 11 adult LOPD siblings belonging to two Italian families carrying compound heterozygous GAA mutations. We disclosed a heterogeneous pattern of myopathic impairment, associated, among others, with cardiac defects, intracranial vessels abnormality, osteoporosis, vitamin D deficiency, obesity and adverse response to enzyme replacement therapy (ERT). We identified deleterious variants in the genes involved in autophagy, immunity and bone metabolism, which contributed to the severity of the clinical symptoms observed in the LOPD patients. This study emphasizes the multisystem nature of LOPD and highlights the polygenic nature of the complex phenotype disclosed in these patients.  相似文献

Building resilience to flooding is a commitment of several universities; however, student interest in flood education programs is unclear. The goals of this research are three-fold: 1) to determine the origin of flood messaging on the Old Dominion University (ODU) campus, 2) to assess on-campus flood awareness, and 3) to evaluate the interest in additional flood education. This study evaluates student awareness of flooding via a survey of ODU students and contextual analysis of University warning messages. Many students experienced reduced access to campus as a result of flooding and expressed an interest in additional flood information. Some students reported receiving flood-related information through in-class instruction or orientation-based programming. However, the content varies in detail, and ODU could formally integrate additional resources into outreach and flood education programming. These findings could support the development of a campus wide flood awareness program at ODU and other universities.  相似文献

Potential applications for colorants derived from the spalting fungi Chlorociboria aeruginascens, Chlorociboria aeruginosa, Scytalidium cuboideum, and Scytalidium ganodermophthorum are of growing interest across a variety of fields, from wood and textile dyes, to solar cells. Previous research found that these dyes could be carried in natural oils, specifically raw linseed oil, instead of organic solvents, thereby eliminating the toxicity issues surrounding such carriers. In this work, dyes from the aforementioned fungi were extracted, carried in raw linseed oil, and mixed with a range of oil and acrylic paints to determine if the oil carrier was appropriate for delivering the dyes into a paint matrix. The oil-based dyes did not mix evenly with the acrylic paints; however, the acrylic paints maintained the color of the dyes. In contrast, the oil-based paints mixed well with the dyes, but the dyes decolorized (completely lost color) due to the presence of refined linseed oil in the paint base. Artists’ paints do not appear to be compatible with fungal dyes when carried in oil; however, there are still many potential applications for the oil-based dyes, such as wood and textile dyeing.  相似文献

Influence of Substituents on the T1 Energy and the S1-T1 Energy Gap in ω-Phosphorus Substituted trans-Styrenes The fluorescence and the phosphorescence of 4-substituted ω-diphenylphosphinyl trans-styrenes 1 and analogous diphenylthiophosphinyl trans-styrenes 2 at 77 K indicate together with some oxygen-induced singlet-triplet absorption spectra a bathochromic shift of the lowest singlet (S1) and the lowest triplet level (T1) relatively to the S1 and T1 level of the unsubstituted compounds 1a and 2a , respectively. Donor substituents X in 4-position give rise to a shift of S1, which is in the most cases much greater than that of T1, so that the S1–T1 energy gap decreases. P(Y)R1R2 groups (Y = O, S) in ω-position of 1 and 2 effect a small bathochromic shift of the T1 energy level only (130–900 cm−1 relatively to unsubstituted styrene). S1 is shifted 860–2080 cm−1 to lower energies, however.  相似文献

5-Hydroxymethylfurfural has become a substance of interest since recent results showed a possible carcinogenic potential in consequence of a metabolic activation by sulfotransferases. 5-Hydroxymethylfurfural is formed either by acid catalysed degradation of reducing sugars or via the Maillard reaction. This work provides an overview of foods potentially containing high amounts of 5-hydroxymethylfurfural. It comprises dried fruits with a high sugar content that were exposed to heat for a long time. The concentration ranges from very low in, e. g. figs (1 mg/kg) to plums that contained up to 2,200 mg/kg. Several types of roasted coffee were analysed that contained from 300 to 2,900 mg/kg of 5-hydroxymethylfurfural. In a small human study with seven healthy volunteers the urine excretion of unmetabolised 5-hydroxymethylfurfural was investigated. After uptake of 20 g of plum jam containing 24 mg of 5-hydroxymethylfurfural, 163 microg (mean) were excreted within 6 h, an equivalent of 0.75% of the ingested 5-hydroxymethylfurfural.  相似文献
