The transport of hydrogen during fusion welding of the titanium alloy Ti-6Al4V is analyzed. A coupled thermodynamic/kinetic treatment is proposed for the mass transport within and around the weld pool. The modeling indicates that hydrogen accumulates in the weld pool as a consequence of the thermodynamic driving forces that arise; a region of hydrogen depletion exists in cooler, surrounding regions in the heat-affected zone and beyond. Coupling with a hydrogen diffusion-controlled bubble growth model is used to simulate bubble growth in the melt and, thus, to make predictions of the hydrogen concentration barrier needed for pore formation. The effects of surface tension of liquid metal and the radius of preexisting microbubble size on the barrier are discussed. The work provides insights into the mechanism of porosity formation in titanium alloys.  相似文献

Three-layer alumina-based composites with Al2O3-containing mullite as the outer layer and ZrO2-containing alumina as the inner layer were fabricated by die-pressing and reaction bonding. The effects of the outer-layer compositions on the densification behavior of three-layer composites were investigated. The existence of residual stresses in the layers was verified using indentation methods. Compared with single-layer ceramics, three-layer composites exhibit an improved fracture toughness and an excellent damage resistance due to the presence of the compressive stresses in the outer layers.  相似文献

The relationship between 3 witness factors and identification accuracy, as well as calibration and diagnosticity of confidence, was investigated. A total of 384 participants in an eyewitness experiment rated their facial recognition skill, general memory skill, and self-reported encoding strategy on a questionnaire presented after the photo-confrontation. Participants who rated themselves to be good face recognizers showed a slightly higher overall accuracy with a more diagnostic confidence-accuracy relation. Participants who reported that they relied on a holistic encoding strategy were associated with more accurate identifications and a stronger confidence-accuracy relation than those who reported an analytic encoding strategy. The biological activities of omega-3 fatty acids (n-3 FAs) have been under extensive study for several decades. However, not much attention has been paid to differences of dietary forms, such as triglycerides (TGs) versus ethyl esters or phospholipids (PLs). New innovative marine raw materials, like krill and fish by-products, present n-3 FAs mainly in the PL form. With their increasing availability, new evidence has emerged on n-3 PL biological activities and differences to n-3 TGs. In this review, we describe the recently discovered nutritional properties of n-3 PLs on different parameters of metabolic syndrome and highlight their different metabolic bioavailability in comparison to other dietary forms of n-3 FAs.  相似文献

High-quality three-dimensional (3D) microscopy allows detailed, unrestricted and non-destructive imaging of entire volumetric tissue specimens and can therefore increase the diagnostic accuracy of histopathological tissue analysis. However, commonly used IgG antibodies are oftentimes not applicable to 3D imaging, due to their relatively large size and consequently inadequate tissue penetration and penetration speed. The lack of suitable reagents for 3D histopathology can be overcome by an emerging class of single-domain antibodies, referred to as nanobodies (Nbs), which can facilitate rapid and superior 2D and 3D histological stainings. Here, we report the generation and experimental validation of Nbs directed against the human endothelial cell-selective adhesion molecule (hESAM), which enables spatial visualization of blood vascular networks in whole-mount 3D imaging. After analysis of Nb binding properties and quality, selected Nb clones were validated in 2D and 3D imaging approaches, demonstrating comparable staining qualities to commercially available hESAM antibodies in 2D, as well as rapid and complete staining of entire specimens in 3D. We propose that the presented hESAM-Nbs can serve as novel blood vessel markers in academic research and can potentially improve 3D histopathological diagnostics of entire human tissue specimens, leading to improved treatment and superior patient outcomes.  相似文献

Pancreatic beta cell dysfunction caused by metabolic and inflammatory stress contributes to the development of type 2 diabetes (T2D). Butyrate, produced by the gut microbiota, has shown beneficial effects on glucose metabolism in animals and humans and may directly affect beta cell function, but the mechanisms are poorly described. The aim of this study was to investigate the effect of butyrate on cytokine-induced beta cell dysfunction in vitro. Mouse islets, rat INS-1E, and human EndoC-βH1 beta cells were exposed long-term to non-cytotoxic concentrations of cytokines and/or butyrate to resemble the slow onset of inflammation in T2D. Beta cell function was assessed by glucose-stimulated insulin secretion (GSIS), gene expression by qPCR and RNA-sequencing, and proliferation by incorporation of EdU into newly synthesized DNA. Butyrate protected beta cells from cytokine-induced impairment of GSIS and insulin content in the three beta cell models. Beta cell proliferation was reduced by both cytokines and butyrate. Expressions of the beta cell specific genes Ins, MafA, and Ucn3 reduced by the cytokine IL-1β were not affected by butyrate. In contrast, butyrate upregulated the expression of secretion/transport-related genes and downregulated inflammatory genes induced by IL-1β in mouse islets. In summary, butyrate prevents pro-inflammatory cytokine-induced beta cell dysfunction.  相似文献

The main aim of the investigation was to quantify the influence of production-related cross-lines on static mechanical properties (tensile, flexural and shear) of an oxide-oxide CMC as a comparison between specimens with cross-lines and specimens without cross-lines in tested regions. Investigated material was a weak-matrix oxide-oxide CMC (WHIPOX?) made of Nextel? 610 fibers (3000 denier) and alumina matrix with a special winding pattern. Mechanical tests at room temperature revealed that cross-lines were local weak regions in a wound component. Spatial separation of the cross-line within the composite (2?mm shift from layer to layer) did not improve the negative influence of the cross-lines on mechanical properties. Fractographic investigations revealed that cross-lines acted as a trigger of material failure.  相似文献

Oxygen uptake by soft bottom sediments was measured in situ with an oxygen electrode in a bell jar. Values in the range 0·3-3·0 g O2 m−2 d−1 were obtained at 19 localities in fresh and brackish water. Comparative measurements were made in the laboratory on sediment cores. These gave consistently lower values than the in situ measurements. Laboratory experiments showed that the oxygen uptake depended on the oxygen concentration and that the temperature coefficient decreased with increasing temperature. There was no simple correlation between oxygen uptake and content organic matter in sediments.  相似文献
