This paper reports the application of an amperometric biosensor for rapid and specific Salmonella Typhimurium detection in milk. This device was developed from self-assembled monolayer technique on a gold screen-printed electrode, using cysteamine thiol. Polyclonal antibodies were oriented by protein A immobilization. The biosensor structure was characterized by cyclic voltammetry, Fourier transform infrared spectroscopy, and scanning electron microscopy. The analytical response was obtained by a chronoamperometry technique, using a direct-sandwich peroxidase-labeled system. The biosensor device showed a qualitative behavior with a very low limit of detection of 10 CFU mL?1 and a detection time of 125 min. The biosensor specificity was demonstrated in pure and mixed samples with strains of Escherichia coli and Citrobacter freundii. The performance of the biosensor was found satisfactory, and the device was tested in skimmed and whole milk samples, being able to detect S. Typhimurium quickly, without an enrichment step. This structure of immunosensor assembly can be expended in future studies for other food matrices and bacterial species, making it a useful tool to ensure food safety.  相似文献

Automation of testing is an essential requirement to render it viable for software development. Although there are several testing techniques and criteria in many different domains, developing methods to test programs with complex outputs remains an unsolved challenge. This setting includes programs with graphical output, which produce images or interface windows. One possible approach towards automating the testing activity is the use of automatic oracles in which a reference image, taken as correct, can be used to establish a correctness measure in the tested program execution. A method that uses concepts of content‐based image retrieval to facilitate oracle automation in the domain of programs with graphics output is presented. Two case studies, one using a computer‐aided diagnostic system and one using a Web application, are presented, including some reflections and discussions that demonstrate the feasibility of the proposed approach. In this paper, we consider the single machine scheduling problem with quadratic earliness and tardiness costs, and no machine idle time. We propose a genetic approach based on a random key alphabet and present several algorithms based on this approach. These versions differ on the generation of both the initial population and the individuals added in the migration step, as well as on the use of local search. The proposed procedures are compared with the best existing heuristics, as well as with optimal solutions for the smaller instance sizes. The computational results show that the proposed algorithms clearly outperform the existing procedures and are quite close to the optimum. The improvement over the existing heuristics increases with both the difficulty and the size of the instances. The performance of the proposed genetic approach is improved by the initialization of the initial population, the generation of greedy randomized solutions, and the addition of the local search procedure. Indeed, the more sophisticated versions can obtain similar or better solutions and are much faster. The genetic version that incorporates all the considered features is the new heuristic of choice for small and medium size instances.  相似文献

The effect of chemical disorder over the martensitic phase transformation of the Ni50Mn36Sn14 Heusler-type alloy was systematically investigated by performing X-ray diffractometry (DRX), DC magnetization and 57Fe-doping and 119Sn-Mössbauer spectroscopy measurements. DRX patterns are characteristics of a L21-type chemically disordered structure, where the presence of this disorder was first evaluated by analyzing the relative intensity of the (1 1 1) DRX reflection, which varies in the case of Fe-doped and practically disappears for the milled samples. In consequence, the magnetic properties of Fe-doped well-milled samples related to the martensitic phase transformation change substantially. 300 K 57Fe-Mössbauer spectroscopy data suggest that the changes in the magnetic properties related to the martensitic transformation are intrinsically correlated to the ferromagnetic and paramagnetic fractions, which are respectively associated with Fe atoms replacing Mn- and Sn-sites. In the case of milled samples, the drastic reduction of alloy magnetization was explained by the increase of the number of Mn atoms in the shell regions, which have a reduced magnetic moment comparatively to those in the grain cores. The magnetization change and the temperature transition in the martensitic transformation are governed by the grain core. The initial magnetic properties and martensitic transformation can be recovered by a subsequent annealing on the milled sample.  相似文献

In this work we investigate the laminar flow through square–square sudden contractions with various contraction ratios (CR=2.4, 4, 8 and 12), using a Newtonian fluid and a shear-thinning viscoelastic fluid. Visualizations of the flow patterns were carried out using streak line photography and detailed velocity field measurements were performed using particle image velocimetry. The experimental results are compared with numerical predictions obtained using a finite-volume method. For the Newtonian fluid, a corner vortex is found upstream of the contraction and increasing flow inertia leads to a reduction of the vortex size. Good agreement is observed between experiments and numerical simulations. For the shear-thinning fluid flow a corner vortex is also observed upstream of the contraction independently of the contraction ratio. Increasing the elasticity of the flow, while still maintaining low inertia flow conditions, leads to a strong increase of the vortex size, until an elastic instability sets in and the flow becomes time-dependent at De≈200, 300, 70 and 450 for CR=2.4, 4, 8 and 12, respectively. At low contraction ratios, viscoelasticity brings out an anomalous divergent flow upstream of the contraction. For both fluids studied the flow presents a complex three-dimensional helical vortex structure which is well predicted by numerical simulations. However, for the viscoelastic fluid flow the maximum Deborah number achieved in the numerical simulations is about one order of magnitude lower than the critical Deborah number for the onset of the elastic instability found in the experiments.  相似文献

The aim of this work was to develop an understanding of the major difficulties associated with the scale‐up of the technology for the synthesis of poly(vinyl chloride) (PVC) block copolymers that contain hydrophilic segments, thus providing important directions to be followed in order to produce such new materials on the industrial scale. The synthesis was carried out in a two‐step process. First, the macroinitiator α,ω‐di(iodo)poly(hydroxypropyl acrylate) was synthesized in an aqueous medium by (single electron transfer)/(degenerative chain transfer) living radical polymerization (SET‐DTRP) catalyzed by Na2S2O4. The block copolymer was then prepared by SET‐DTRP of vinyl chloride (VC) from the iodine‐terminal active chain ends of the macroinitiator, thereby leading to the formation of the block copolymer poly(vinyl chloride)‐b‐poly(hydroxypropyl acrylate)‐b‐poly(vinyl chloride). This report covers important aspects related to the characterization of the block copolymer produced and to the identification of the major limitations that must be overcome in order to produce this new material on the industrial scale. The results clearly show the differences between the theoretical predictions and the block copolymer compositions obtained by using a suspension polymerization method, which is the most‐used polymerization process in the PVC industry. We present a study of the structural and magnetic properties of Zn1?xFexO (0.00≤x≤0.09) nanoparticles prepared by a combustion reaction method. X-ray diffraction patterns of all samples showed sharp and intensive peaks of hexagonal wurtzite structure of ZnO without any evidence of spurious crystalline phases in all samples. The magnetic properties were measured by using a vibrating sample magnetometer. Hysteresis loop indicated that all samples are ferromagnetic at room temperature and the Curie temperature increased with increasing iron content. Room temperature ferromagnetism can be explained in terms of intrinsic properties and the presence of defects in Fe- doped ZnO nanoparticles.  相似文献

This study evaluates the effects of a westernized diet during the perinatal period on the maternal performance and growth and development of rat offspring. Female Wistar rats were fed with either a control (C) diet, with casein as the protein source or a westernized (W) diet, during pregnancy and lactation. The pups were divided, eight per group, into the same diet groups as their dams. During lactation, the body weight (day 1, W = 6.85 ± 0.62 g, C = 5.81 ± 0.49, p < 0.05; day 21, W = 55.42 ± 3.78, C = 47.75 ± 3.45, p < 0.001) and somatic growth (body length day 1, W = 53.24 ± 2.16, C = 50.641 ± 1.79, p < 0.05; day 21, W = 124.8, C = 119.903 ± 3.71, p < 0.001) in the male offspring showed significant differences among the groups. The physical appearance and reflex maturation showed differences between day 1 and day 3. With the westernized diet, during the perinatal period, no alterations in maternal weight gain, gestation or performance were observed; however, changes in the coefficients of feed efficiency and energy during lactation were noted. Besides, blood glucose was found to be elevated at the end of lactation (C = 3.67 ± 0.35 mmol/l, W = 5.2 0 ± 0.49 mmol/l). At 21 days, the male pups from the dams on the westernized diet were 15 % heavier, and the maturation of the neural reflexes and physical characteristics were found to occur earlier. Therefore, the consumption of a westernized diet during the perinatal period was independent of maternal energy intake, and influenced the growth and development of offspring.  相似文献
