The effectiveness of antimicrobial mixtures against Listeria innocua 7, used as a L. monocytogenes surrogate, was investigated in broth and a food system. Synergistic effects were found for nisin (Nis), potassium sorbate (PS), calcium propionate (CP) and sodium lactate (SL), Nis + PS being the most effective binary mixture that exhibited listericidal activity in broth. To assess the effect of adding lactocin AL705 (AL705) to Nis + organic acid salt combinations, tridimensional isobolograms were generated. Sub-MIC combinations of the antimicrobials exerted bactericidal activity against L. innocua 7 after AL705 addition to the binary mixtures. However, when applied on Sardo cheese contaminated with L. innocua 7 (initial inoculum 4.45 ± 0.06 CFU g−1), only Nis + PS + AL705 produced count reductions respect to the control, reaching 3.04 ± 0.35 CFU g−1 counts after 15 days at 15 °C. Recommended drying treatments may not enhance destruction of pathogens that could be present on home-dried foods. In this study, the effects of traditional and modified treatments on Salmonella were evaluated during preparation, home-type dehydration (60 degrees C for 6 h), and storage of potato slices. Potato slices inoculated with five strains of Salmonella (8.4 log CFU/ g) were left untreated or were treated by steam blanching (88 degrees C for 10 min), water blanching (88 degrees C for 4 min), 0.105% citric acid blanching (88 degrees C for 4 min), or 0.210% citric acid blanching (88 degrees C for 4 min). Slices were then dried (6 h for 60 degrees C) and aerobically stored for up to 30 days at 25 +/- 3 degrees C. Cells were enumerated on tryptic soy agar with 0.1% pyruvate (TSAP) and on xylose lysine deoxycholate agar. Salmonella populations were reduced by 4.5 to 4.8 CFU/g and by >5.4 log CFU/g immediately following steam and water blanching, respectively. Populations were below the detection limit (0.80 log CFU/g) immediately following acid blanching, except for samples blanched in 0.105% citric acid and recovered on TSAP. After dehydration (6 h for 60 degrees C), Salmonella reductions on blanched potato slices (5.3 to 5.6 log CFU/g) were significantly greater (P < 0.05) than those on untreated samples (1.9 to 2.7 log CFU/g). Populations on all samples continued to decrease throughout 30 days of storage but still were 3.1 to 3.9 log CFU/g on untreated samples. In comparison, bacterial populations on blanched samples were undetectable by direct plating following 30 days of storage (regardless of blanching method). Blanching treatments used in this study improved the effectiveness of drying for inactivating Salmonella inoculated onto potato slices and, therefore, may enhance the safety of the product. 相似文献

Pyrrolizidine alkaloids (PAs) are secondary metabolites of plant families such as Asteraceae or Boraginaceae and are suspected to be genotoxic carcinogens. Recent investigations revealed their frequent occurrence in honey and particularly in tea. To obtain a comprehensive overview of the PA content in animal- and plant-derived food from the European market, and to provide a basis for future risk analysis, a total of 1105 samples were collected in 2014 and 2015. These comprised milk and milk products, eggs, meat and meat products, (herbal) teas, and (herbal) food supplements collected in supermarkets, retail shops, and via the internet. PAs were detected in a large proportion of plant-derived foods: 91% of the (herbal) teas and 60% of the food supplements contained at least one individual PA. All types of (herbal) teas investigated were found to contain PAs, with a mean concentration of 460 µg kg?1 dry tea (corresponding to 6.13 µg L?1 in [herbal] tea infusion). The highest mean concentrations were found in rooibos tea (599 µg kg?1 dry tea, 7.99 µg L?1 tea infusion) and the lowest in camomile tea (274 µg kg?1 dry tea, 3.65 µg L?1 tea infusion). Occurrence of PAs in food supplements was found to be highly variable, but in comparable ranges as for (herbal) tea. The highest concentrations were present in supplements containing plant material from known PA-producing plants. In contrast, only 2% of the animal-derived products, in particular 6% of milk samples and 1% of egg samples, contained PAs. Determined levels in milk were relatively low, ranged between 0.05 and 0.17 µg L?1 and only trace amounts of 0.10–0.12 µg kg?1 were found in eggs. The presence or absence of filbertone in 21 admixtures of olive oil with virgin and refined hazelnut oils obtained using various processing techniques from different varieties and geographical origins was evaluated by solid phase microextraction and multidimensional gas chromatography (SPME–MDGC). The obtained results showed that the sensitivity achievable with the proposed procedure was enough to detect filbertone and, hence, to establish the adulteration of olive oil of different varieties with virgin hazelnut oils in percentages of up to 7%. The very low concentrations in which filbertone occurs in some refined hazelnut oils made difficult its detection in specific admixtures. In any case, the minimum adulteration level to be detected depends on the oil varieties present in the adulterated samples. In the present study, the presence of R- and S-enantiomers of filbertone could be occasionally detected in olive oils adulterated with 10–20% of refined hazelnut oil. 相似文献

The environmental distribution and the biomagnification of a suite of perfluoroalkyl compounds (PFCs), including perfluorooctane sulfonate (PFOS) and C8 to C14 perfluorinated carboxylates (PFCAs), was investigated in the food web of the bottlenose dolphin (Tursiops truncatus). Surficial seawater and sediment samples, as well as zooplankton, fish, and bottlenose dolphin tissue samples, were collected at two U.S. locations: Sarasota Bay, FL and Charleston Harbor, SC. Wastewater treatment plant (WWTP) effluents were also collected from the Charleston area (n = 4). A solid-phase extraction was used for seawater and effluent samples and an ion-pairing method was used for sediment and biotic samples. PFCs were detected in seawater (range <1-12 ng/L), sediment (range <0.01-0.4 ng/g wet weight (ww)), and zooplankton (range 0.06-0.3 ng/g ww). The highest PFC concentrations were detected in WWTP effluents, whole fish, and dolphin plasma and tissue samples in which PFOS, C8 and C10-PFCAs predominated in most matrices. Contamination profiles varied with location suggesting different sources of PFC emissions. Biomagnification factors (BMFs) ranged from <1 to 156 at Sarasota Bay and <1 to 30 at Charleston. Trophic magnification factors (TMFs) for PFOS and C8-C11 PFCAs indicated biomagnification in this marine food web. The results indicate that using plasma and liver PFC concentrations as surrogate to whole body burden in a top marine predator overestimates the BMFs and TMFs. 相似文献

This study evaluated post-processing chemical solutions for their antilisterial effects on commercial smoked sausage formulated with or without 1.5% potassium lactate plus 0.05% sodium diacetate, and contaminated (approximately 3-4 log cfu/cm(2)) with 10-strain composite Listeria monocytogenes inocula prepared under various conditions. Inoculated samples were left untreated, or were immersed (2 min, 25 +/- 2 degrees C) in solutions of acetic acid (2.5%), lactic acid (2.5%), potassium benzoate (5%) or Nisaplin (0.5%, equivalent to 5000 IU/ml of nisin) alone, and in sequence (Nisaplin followed by acetic acid, lactic acid or potassium benzoate), before vacuum packaging and storage at 10 degrees C (48 days). Acetic acid, lactic acid or potassium benzoate applied alone reduced initial L. monocytogenes populations by 0.4-1.5 log cfu/cm(2), while treatments including Nisaplin caused reductions of 2.1-3.3 log cfu/cm(2). L. monocytogenes on untreated sausage formulated with antimicrobials had a lag phase duration of 10.2 days and maximum specific growth rate (mu(max)) of 0.089 per day, compared to no lag phase and mu(max) of 0.300 per day for L. monocytogenes on untreated product that did not contain antimicrobials in the formulation. The immersion treatments inhibited growth of the pathogen for 4.9-14.8 days on sausage formulated without potassium lactate-sodium diacetate; however, in all cases significant (P < 0.05) growth occurred by the end of storage. The antilisterial activity of chemical solutions was greatly enhanced when applied to product formulated with antimicrobials; growth was completely inhibited on sausage treated with acetic or lactic acid alone, and in sequence with Nisaplin. In general, habituation (15 degrees C, 7 days) of L. monocytogenes cells, planktonically or as attached cells to stainless-steel coupons in sausage homogenate prior to contamination of product, resulted in shorter lag phase durations compared with cells cultivated planktonically in a broth medium. Furthermore, when present, high levels of spoilage flora were found to suppress growth of the pathogen. Nimonic 80A is a nickel-chrome superalloy, commonly used due to its high resistance against creep, oxidation, and temperature corrosion. This paper presents the material constitutive models of Nimonic 80A superalloy. Johnson–Cook (JC) and modified JC model is preferred among the different material constitutive equations (Zerill Armstrong, Bodner Partom, Arrhenius type) due to its accuracy in the literature. Three different types of compression tests were applied to determine the equation parameters. Firstly, quasi-static tests were performed at room temperature. These tests were conducted at 10?3, 10?2, and 10?1 s?1 strain rates. Secondly, compression tests were performed at room temperature at high strain rates (370–954 s?1) using the Split-Hopkinson pressure bar. Finally, compression tests were performed at a temperature level from 24 to 200 °C at the reference strain rate (10?3 s?1). Johnson–Cook and modified JC model parameters of Nimonic 80A were determined with the data obtained from these tests, and they were finally verified statistically. 相似文献
