In this paper, we investigated the effects of neonatal exposure to methoxychlor (MXC), a synthetic organochlorine used as an insecticide with estrogenic, antiestrogenic, and antiandrogenic activities on ovarian follicles of adult pigs. Piglets were injected with MXC (20 μg/kg body weight) or corn oil (controls) from postnatal Day 1 to Day 10 (n = 5 per group). Then, mRNA expression, protein abundance and immunolocalization of growth and differentiation factor 9 (GDF9), bone morphogenetic protein 15 (BMP15), anti-Müllerian hormone (AMH) and cognate receptors (ACVR1, BMPR1A, BMPR1B, TGFBR1, BMPR2, and AMHR2), as well as FSH receptor (FSHR) were examined in preantral and small antral ovarian follicles of sexually mature gilts. The plasma AMH and FSH levels were also assessed. In preantral follicles, neonatal exposure to MXC increased GDF9, BMPR1B, TGFBR1, and BMPR2 mRNAs, while the levels of AMH and BMP15 mRNAs decreased. In addition, MXC also decreased BMP15 and BMPR1B protein abundance. Regarding small antral follicles, neonatal exposure to MXC upregulated mRNAs for BMPR1B, BMPR2, and AMHR2 and downregulated mRNAs for AMH, BMPR1A, and FSHR. MXC decreased the protein abundance of AMH, and all examined receptors in small antral follicles. GDF9 and BMP15 were immunolocalized in oocytes and granulosa cells of preantral follicles of control and treated ovaries. All analyzed receptors were detected in the oocytes and granulosa cells of preantral follicles, and in the granulosa and theca cells of small antral follicles. The exception, however, was FSHR, which was detected only in the granulosa cells of small antral follicles. In addition, MXC decreased the plasma AMH and FSH concentrations. In conclusion, the present study may indicate long-term effects of neonatal MXC exposure on GDF9, BMP15, AMH, and FSH signaling in ovaries of adult pigs. However, the MXC effects varied at different stages of follicular development. It seems that neonatal MXC exposure may result in accelerated initial recruitment of ovarian follicles and impaired cyclic recruitment of antral follicles.  相似文献

In this work we present studies on applicability of transition metal additives as sintering and electrical conductivity aids for cerium gadolinium oxide electrolyte. The nanosized Ce0.85Gd0.15O1.925 powder obtained by coprecipitation method was modified with Cr3+, Fe3+, Ni2+ or Cu2+ ions. Using high-intensity high-resolution X-ray powder diffraction data we have determined that Cr, Fe and Ni ions do not incorporate into the cerium gadolinium oxide surface or bulk when sintered at 1300 °C, but react with Gd ions to form Cr0.9Gd0.1O, GdFeO3 and GdNiO3 phases, while Cu incorporates in the material up to 0.7 mol% with a significant fraction of remaining material showing poorly crystalline CuO phase. The nanosized Ce0.85Gd0.15O1.925 material shows already improved sintering properties than previous reports but full sintering is not achieved below 1300 °C, however Cr, Fe and mainly Cu impregnation allows full sintering at 1300 °C. 0.5 mol% Ni impregnated material sintered at 1500 °C shows enhanced grain boundary conductivity that probably indicates that Ni incorporates into Ce0.84Gd0.15O1.925 above 1300 °C. The global results indicate, however, that optimization of ceria microstructure is at least of equal importance for sinterability and grain boundary conductivity than impregnation of the material with transition metal ions.  相似文献

Optical properties of the ultrananocrystalline diamond films were studied by multi-sample method based on the combination of variable angle spectroscopic ellipsometry and spectroscopic reflectometry applied in the range 0.6–6.5 eV. The films were deposited by PECVD in a conventional bell jar (ASTeX type) reactor using dual frequency discharge, microwave cavity plasma and radio frequency plasma inducing dc self-bias at a substrate holder. The optical model of the samples included a surface roughness described by the Rayleigh–Rice theory and a refractive index profile in which Drude approximation was used. The results conformed with the present understanding of the polycrystalline diamond growth on the silicon substrate because the existence of silicon carbide and amorphous hydrogenated carbon film between the silicon substrate and nucleation layer was proved.  相似文献

腐植酸对土壤环境的影响仍然是科学研究需要讨论的话题。本研究目的是调查11种泥炭淤泥质土壤的理化性质与从中分离的腐植酸属性的相关关系。本次测量的土壤参数有:总碳和有机碳、容重、灰分、腐殖化度和吸水指数。选择的腐植酸参数有:280 nm吸光度,465 nm和655 nm、260 nm和655 nm的吸光度比值,元素组成,H/C,O/H,O/C,C/N,氧化程度,羧基和酚基官能团含量。统计分析显示:容重、腐殖化度、吸水指数的升高导致腐植酸参数280 nm吸光度、含氧量、O/H、O/C、氧化度、官能团含量的显著升高。研究还发现土壤属性参数与腐植酸的碳氢含量及H/C比值呈明显负相关。因此,只要测量一些土壤参数,即使没有耗时分离腐植酸,弄清腐植酸的性质也成为可能。  相似文献

For a successful implementation of newly proposed silicon-based latent heat thermal energy storage systems, proper ceramic materials that could withstand a contact heating with molten silicon at temperatures much higher than its melting point need to be developed. In this regard, a non-wetting behavior and low reactivity are the main criteria determining the applicability of ceramic as a potential crucible material for long-term ultrahigh temperature contact with molten silicon. In this work, the wetting of hexagonal boron nitride (h-BN) by molten silicon was examined for the first time at temperatures up to 1750 °C. For this purpose, the sessile drop technique combined with contact heating procedure under static argon was used. The reactivity in Si/h-BN system under proposed conditions was evaluated by SEM/EDS examinations of the solidified couple. It was demonstrated that increase in temperature improves wetting, and consequently, non-wetting-to-wetting transition takes place at around 1650 °C. The contact angle of 90° ± 5° is maintained at temperatures up to 1750 °C. The results of structural characterization supported by a thermodynamic modeling indicate that the wetting behavior of the Si/h-BN couple during heating to and cooling from ultrahigh temperature of 1750 °C is mainly controlled by the substrate dissolution/reprecipitation mechanism.  相似文献

The aim of this research was to observe the relationship between zeta potential, morphology, surface area, porosity, chemical composition, and ecotoxicity of nanocomposite powders such as Au/TiO2, Ag2O/TiO2, PdO/TiO2, Ag/TiO2/SiO2, Ag/N(C)TiO2, and SiO2/TiO2 from which Ag2O/TiO2, Ag/N(C)/TiO2, and Ag/TiO2/SiO2 were exhibiting good antimicrobial properties. It was observed, that nanomaterials characterized by similar morphology and zeta potential revealed the similar toxic behavior. The samples of higher agglomeration and higher zeta potential, especially Ag/TiO2/SiO2 xerogel and TiO2/SiO2 aerogel were generally less ecotoxic to water organisms and plants. They were also not genotoxic in concentrations up to 500 and 250 mg/L, respectively.  相似文献

Rhabdomyosarcoma (RMS) is a malignant soft tissue cancer that develops mostly in children and young adults. With regard to histopathology, four rhabdomyosarcoma types are distinguishable: embryonal, alveolar, pleomorphic and spindle/sclerosing. Currently, increased amounts of evidence indicate that not only gene mutations, but also epigenetic modifications may be involved in the development of RMS. Epigenomic changes regulate the chromatin architecture and affect the interaction between DNA strands, histones and chromatin binding proteins, thus, are able to control gene expression. The main aim of the study was to assess the role of protein arginine methyltransferases (PRMT) in the cellular biology of rhabdomyosarcoma. In the study we used two pan-inhibitors of PRMT, called AMI-1 and SAH, and evaluated their effects on proliferation and apoptosis of RMS cells. We observed that AMI-1 and SAH reduce the invasive phenotype of rhabdomyosarcoma cells by decreasing their proliferation rate, cell viability and ability to form cell colonies. In addition, microarray analysis revealed that these inhibitors attenuate the activity of the PI3K-Akt signaling pathway and affect expression of genes related to it.  相似文献

Aniline was oxidized with silver nitrate in aqueous solutions of sulfonic acids: camphorsulfonic, methanesulfonic, sulfamic, or toluenesulfonic acids. Polyaniline–silver composites were produced slowly in 4 weeks in good yield, except for the reaction, which took place in sulfamic acid solution, where the yield was low. Polyaniline in the emeraldine form was identified with UV–visible, FTIR, and Raman spectra. Thermogravimetric analysis was used to determine the silver content, which was close to the theoretical prediction of 68.9 wt.%. Transmission electron microscopy demonstrated the presence of silver nanoparticles of ca 50 nm average sizes as the dominating species, and hairy polyaniline nanorods having diameter 150–250 nm accompanied them. The highest conductivity of 880 S cm−1 was found with the composite prepared in methanesulfonic acid solution. Its conductivity decreased with temperature increasing in the 70–315 K range, which is typical of metals such as silver. The conductivity of composites prepared in solutions of other acids was lower and increased with increasing temperature. Such dependence is typical of semiconductors, reflecting the dominating role of polyaniline in the conductivity behaviour. It is proposed that interfaces between the polyaniline matrix and dispersed silver nanoparticles play a dominating role in macroscopic level of conductivity.  相似文献

The immunosuppressive effects of UV radiation have been well documented. This suppression has been attributed to the action of the cis form of urocanic acid (UCA), a photoproduct of trans-UCA, a natural constituent of the skin. Here, we show that mouse spleen cells preincubated with cis-UCA have a diminished proliferative response to allogeneic cells in MLC and to stimulation with anti-CD3 mAb. Cells preincubated with cis-UCA also had a decreased ability to serve as APC and to stimulate the proliferation of allogeneic lymphocytes in MLC. Simultaneously, the production of IL-2 and IFN-gamma by cells preincubated with cis-UCA was decreased. However, IL-10 gene expression and IL-10 protein secretion by spleen cells stimulated in the presence of cis-UCA were significantly enhanced. The principal cell population displaying the cis-UCA-induced elevated production of IL-10 was CD4+ T cells, which were shown to be a direct target of cis-UCA action. This was also supported by the observation that production of IL-10 by stimulated splenic non-T cells or by macrophages was not altered by cis-UCA. The enhanced production of IL-10 by activated CD4+ T cells may represent a novel pathway of UVB radiation-induced, cis-UCA-mediated immunosuppression. We suggest that the elevated production of IL-10 by activated CD4+ T cells may account for the suppressor T cell phenomena described in UV-irradiated recipients.  相似文献
