The product of the REF2 gene is required for optimal levels of endonucleolytic cleavage at the 3' ends of yeast mRNA, prior to the addition of a poly(A) tail. To test the role of the previously demonstrated nonspecific affinity of REF2 for RNA in this process, we have identified RNA binding mutants in vitro and tested them for function within the cell. One REF2 variant, with an internal deletion of 82 amino acids (269-350), displays a 10-fold reduction in RNA binding, yet still retains full levels of processing activity in vivo. Conversely, a series of carboxyl-terminal deletions that maintain full RNA binding capability have progressively decreasing activity. These results rule out a major role for the central RNA binding domain of REF2 in mRNA 3' end processing and demonstrate the importance of the carboxyl-terminal region. To ask if the stimulatory role of REF2 depends on interactions with other proteins, we used a two-hybrid screen to identify a new protein termed FIR1 (Factor Interacting with REF) encoded on chromosome V. FIR1 interacts with two independent regions of REF2, one of which (amino acids 268-345) overlaps the RNA binding domain and is dispensible for REF2 function, whereas the other (amino acids 391-533) is located within the critical carboxyl-terminus. As with REF2, FIR1 has a small but detectable role in influencing the efficiency of poly(A) site use. Yeast strains containing a disrupted FIR1 gene are slightly less efficient in the use of cryptic poly(A) sites located within the lacZ portion of an ACT1-lacZ reporter construct. Likewise, a double delta ref2, delta fir1 mutant is more defective in processing of a reporter CYC1 poly(A) site than delta ref2 alone. This synergistic response provides additional support for the interaction of FIR1 with REF2 in vivo, and suggests that a number of gene products may be involved in regulating the cleavage reaction in yeast.  相似文献

The effects of the SAR (14 and 38) and of the methodology of introduction of nickel (wetness impregnation and ion exchange) on catalyst activation were investigated in Ni/USY model catalysts submitted to two activation treatments: reduction (A) and reduction/calcination/reduction, successively (B). the catalysts prepared by wetness impregnation, a marked increase in the catalytic activity after treatment B was observed. On the basis of the catalyst efficiency toward ethane hydrogenolysis, the lower SAR catalyst presents a threefold higher activity. On the contrary, the catalysts prepared by ion exchange, as well as a model catalyst prepared by wetness impregnation over a USY exhaustively exchanged with nickel ions, do not present measurable increasing of the catalytic activity under the same activation conditions. Monitoring the catalyst activation by EXAFS indicates the formation of nickel clusters in the impregnated catalysts after activation treatment B, which should be responsible for the high catalytic activity. However, TEM images reveal a bimodal particle size distribution, with large (ca. 20 nm) and small nickel particles. Temperature-programmed reduction performed under the same conditions of catalyst activation suggests that only the small nickel particles are activated under the experimental conditions adopted in this work.  相似文献

Several thermally coupled distillation sequences have been proposed to improve the thermal inefficiency of conventional distillation sequences. Particularly, for the separation of ternary mixtures, structures that perform a lateral extraction in one of the columns of the integrated arrangement have been shown to provide significant energy savings. The structure of existing sequences, based on conventional distillation columns, might provide the basis for alternate thermally coupled designs. In this paper, it is shown how a thermally coupled system derived from an indirect conventional sequence can provide energy savings through a proper optimization of the interconnecting streams.  相似文献

Abstract

The calcium (Ca), sodium (Na), and potassium (K) contents of blue gum, maritime pine, and cork increased after these lignocellulosics were stirred with the filtrate of a cement suspension or a solution of calcium hydroxide. The adsorbed cations could be released easily by treatment with an acidic solution. It is suggested that those raw materials, and lignocellulosics in general, act as cation exchange substrates when they are mixed with cement and water to make wood–cement composites. The implications that such phenomena may have on cement hardening reactions, and how they could hinder them are discussed. The possibility that the phenomenon is related to well‐known compatibility problems that many woods present when manufacturing wood–cement composites is discussed.  相似文献

The aim of this study was to apply the PIXE (Particle Induced X-ray Emission) and PDMS (Plasma Desorption Mass Spectrometry) techniques to characterize airborne dust particles containing metals. Aerosols generated at a mineral-sand processing plant were characterized in this study. The aerosol samples were collected at a plant that processes mineral sands to obtain rutile, ilmenite, zircon, and monazite concentrates. A cascade impactor with six stages was used to collect mineral dust particles with aerodynamic diameters in the range of 0.64 to 19.4 mum. The particles impacted on each stage of the cascade impactor were analyzed by PIXE, which permits the determination of the elemental mass air concentration and the MMAD (Mass Median Aerodynamic Diameter). The chemical compositions of the aerosol samples were identified by PDMS analysis. This study shows that, by using PIXE and PDMS techniques, it is possible to determine the chemical compounds in which the elements are associated in the aerosol particles. Based on the results of the PIXE analysis, the elemental mass concentrations and the MMADs were determined.  相似文献

The synthesis of the title complexes was achieved via the reaction of -p-dichlorobenzene- -cyclopentadienyliron cations with 4,4′-bis(4-hydroxyphenyl)valeric acid to produce the diiron complexes which were then reacted with a number of arylazo dyes to give cationic bis(cyclopentadienyliron)arene complexes containing the arylazo dyes. These iron-containing monomers were subsequently polymerized via nucleophilic aromatic substitution using 1,8-octanedithiol, 4,4′-thiobisbenzenethiol, or bisphenol A to produce the desired coloured cationic organoiron polymers. The weight – average molecular weights were estimated to range from 11,800 to 31,600. UV–vis studies conducted in dimethylformamide (DMF) showed that the metallated polymers exhibited of 412–491 nm. Addition of HCl to the polymer solution caused a bathochromic shift into the range of 515–530 nm. Thermogravimetric analysis (TGA) revealed that the iron moieties were cleaved between 205 and 248 °C while the polyether/thioether backbone degraded between 380 and 613 °C. Differential scanning calorimetry (DSC) showed that the polymers exhibited glass transition temperatures (Tg) ranging from 106 to 184°C.This paper is dedicated to Professor Richard J. Osmotic dehydration assisted by ultrasound (ODAU) at low temperatures reduces water activity (aw) and maintains nutrients. The influence of solution concentration (SC; 20 to 60° Brix, xylitol and sorbitol) and ultrasound application time (tus, 0 to 40 min) in ODAU of yacon was studied with the aid of a response surface method. The optimum condition with respect to mass transfer parameters, aw, and fructan retention was SC of 60° Brix for both solutions and tus of 2.67 min for xylitol samples and 0 min for sorbitol samples. The application of ultrasound improved dehydration but resulted in depolymerization of fructans.  相似文献

Mercury is a severe environmental pollutant with neurotoxic effects, especially when exposed for long periods. Although there are several evidences regarding mercury toxicity, little is known about inorganic mercury (IHg) species and cerebellum, one of the main targets of mercury associated with the neurological symptomatology of mercurial poisoning. Besides that, the global proteomic profile assessment is a valuable tool to screen possible biomarkers and elucidate molecular targets of mercury neurotoxicity; however, the literature is still scarce. Thus, this study aimed to investigate the effects of long-term exposure to IHg in adult rats’ cerebellum and explore the modulation of the cerebellar proteome associated with biochemical and functional outcomes, providing evidence, in a translational perspective, of new mercury toxicity targets and possible biomarkers. Fifty-four adult rats were exposed to 0.375 mg/kg of HgCl2 or distilled water for 45 days using intragastric gavage. Then, the motor functions were evaluated by rotarod and inclined plane. The cerebellum was collected to quantify mercury levels, to assess the antioxidant activity against peroxyl radicals (ACAPs), the lipid peroxidation (LPO), the proteomic profile, the cell death nature by cytotoxicity and apoptosis, and the Purkinje cells density. The IHg exposure increased mercury levels in the cerebellum, reducing ACAP and increasing LPO. The proteomic approach revealed a total 419 proteins with different statuses of regulation, associated with different biological processes, such as synaptic signaling, energy metabolism and nervous system development, e.g., all these molecular changes are associated with increased cytotoxicity and apoptosis, with a neurodegenerative pattern on Purkinje cells layer and poor motor coordination and balance. In conclusion, all these findings feature a neurodegenerative process triggered by IHg in the cerebellum that culminated into motor functions deficits, which are associated with several molecular features and may be related to the clinical outcomes of people exposed to the toxicant.  相似文献
