The field of drug delivery focuses primarily on delivering small organic molecules or DNA/RNA as therapeutics and has largely ignored the potential for delivering catalytically active transition metal ions and complexes. The delivery of a variety of transition metals has potential for inducing apoptosis in targeted cells. The chief aims of this work were the development of a suitable delivery vector for a prototypical transition metal, Cu2+, and demonstration of the ability to impact cancer cell viability via exposure to such a Cu-loaded vector. Carboxylate-functionalized nanoparticles were synthesized by free radical polymerization and were subsequently loaded with Cu2+ via binding to particle-bound carboxylate functional groups. Cu loading and release were characterized via ICP MS, EDX, XPS, and elemental analysis. Results demonstrated that Cu could be loaded in high weight percent (up to 16 wt.%) and that Cu was released from the particles in a pH-dependent manner. Metal release was a function of both pH and the presence of competing ligands. The toxicity of the particles was measured in HeLa cells where reductions in cell viability greater than 95% were observed at high Cu loading. The combined pH sensitivity and significant toxicity make this copper delivery vector an excellent candidate for the targeted killing of disease cells when combined with an effective cellular targeting strategy.  相似文献

The mobile agent paradigm has been adopted by several systems in the area of wireless sensor networks as it enables a flexible distribution and placement of application components on nodes, at runtime. Most agent placement and migration algorithms proposed in the literature, assume that the communication rates between agents remain stable for a sufficiently long time to amortize the migration costs. Then, the problem is that frequent changes in the application-level communication may lead to several non-beneficial agent migrations, which may actually increase the total network cost, instead of decreasing it. To tackle this problem, we propose two distributed algorithms that take migration decisions in an online fashion, trying to deal with fluctuations in agent communication. The first algorithm is more of theoretical value, as it assumes infinite storage to keep information about the message exchange history of agents, while the second algorithm is a refined version that works with finite storage and limited information. We describe these algorithms in detail, and provide proofs for their competitive ratio vs. an optimal oracle. In addition, we evaluate the performance of the proposed algorithms for different parameter settings through a series of simulated experiments, also comparing their results with those achieved by an optimal static placement that is computed with full (a posteriori) knowledge of the execution scenarios. Our theoretical and experimental results are a strong indication for the robustness and effectiveness of the proposed algorithms.  相似文献

Plug-in hybrid electric vehicle (PHEV) technology is receiving attention as an approach to reducing US dependency on foreign oil and greenhouse gas (GHG) emissions from the transportation sector. PHEVs require large batteries for energy storage, which affect vehicle cost, weight, and performance. We construct PHEV simulation models to account for the effects of additional batteries on fuel consumption, cost, and GHG emissions over a range of charging frequencies (distance traveled between charges). We find that when charged frequently, every 20 miles or less, using average US electricity, small-capacity PHEVs are less expensive and release fewer GHGs than hybrid electric vehicles (HEVs) or conventional vehicles. For moderate charging intervals of 20–100 miles, PHEVs release fewer GHGs, but HEVs have lower lifetime costs. High fuel prices, low-cost batteries, or high carbon taxes combined with low-carbon electricity generation would make small-capacity PHEVs cost competitive for a wide range of drivers. In contrast, increased battery specific energy or carbon taxes without decarbonization of the electricity grid would have limited impact. Large-capacity PHEVs sized for 40 or more miles of electric-only travel do not offer the lowest lifetime cost in any scenario, although they could minimize GHG emissions for some drivers and provide potential to shift air pollutant emissions away from population centers. The tradeoffs identified in this analysis can provide a space for vehicle manufacturers, policymakers, and the public to identify optimal decisions for PHEV design, policy and use. Given the alignment of economic, environmental, and national security objectives, policies aimed at putting PHEVs on the road will likely be most effective if they focus on adoption of small-capacity PHEVs by urban drivers who can charge frequently.  相似文献

Using coal to produce transportation fuels could improve the energy security of the United States by replacing some of the demand for imported petroleum. Because of concerns regarding climate change and the high greenhouse gas (GHG) emissions associated with conventional coal use, policies to encourage pathways that utilize coal for transportation should seek to reduce GHGs compared to petroleum fuels. This paper compares the GHG emissions of coal-to-liquid (CTL) fuels to the emissions of plug-in hybrid electric vehicles (PHEV) powered with coal-based electricity, and to the emissions of a fuel cell vehicle (FCV) that uses coal-based hydrogen. A life cycle approach is used to account for fuel cycle and use-phase emissions, as well as vehicle cycle and battery manufacturing emissions. This analysis allows policymakers to better identify benefits or disadvantages of an energy future that includes coal as a transportation fuel. We find that PHEVs could reduce vehicle life cycle GHG emissions by up to about one-half when coal with carbon capture and sequestration is used to generate the electricity used by the vehicles. On the other hand, CTL fuels and coal-based hydrogen would likely lead to significantly increased emissions compared to PHEVs and conventional vehicles using petroleum-based fuels.  相似文献

The scope of this paper is to summarise a methodology developed for TRENDS (TRansport and ENvironment Database System-TRENDS). The main objective of TRENDS was the calculation of environmental pressure indicators caused by transport. The environmental pressures considered are associated with air emissions from the four main transport modes, i.e. road, rail, ships and air. In order to determine these indicators a system for calculating a range of environmental pressures due to transport was developed within a PC-based MS Access environment. Emphasis is given on the latest features incorporated in the model and their applications. One of the recently developed features of the software provides an option for simple scenario analysis including vehicle dynamics (such as turnover and evolution) for all EU15 member states. This feature is called the Transport Activity Balance module (TAB) and enables the production of collective results for all transport modes as well as a comparative assessment of air emissions produced by the various modes. Traffic activity and emission data obtained according to a basic (reference) scenario are displayed for the time period 1970-2020. In addition, a detailed assessment of the results produced by TRENDS was conducted by means of comparison with data found in the literature. Finally, vehicle emissions produced by the model for the EU15 member states were spatially disaggregated for the base year, 1995 and GIS maps were generated. Examples of these maps are displayed in this document, for the various modes of transport considered in the study.  相似文献

Quality of frozen cucumber can significantly be improved by means of osmotic dehydration as a pre-freezing treatment. The objective of this work was to study the effect of osmotic pre-treatment with alternative osmotic solutes – oligofructose and a high DE maltodextrin – on the quality and functional properties of frozen cucumber tissue. Colour, texture and sensory characteristics of pre-treated and conventionally frozen samples were comparatively measured at 3 months time, during storage at four different temperatures, − 5, − 8, − 12, − 15 °C. Colour change was kinetically studied, and its temperature dependence was modelled by the Arrhenius equation. Dehydrofrozen samples exhibited significantly improved stability, with the rates of colour change being reduced up to 36.7% for osmotically pre-treated cucumbers, compared to the untreated samples. The results indicate that osmodehydrofrozen compared to conventionally frozen sliced cucumbers show improved firmness for prolonged storage period. Sensory evaluation also showed good organoleptic quality in osmodehydrofrozen cucumber slices.

Industrial relevance

The poor quality of frozen cucumber can significantly be improved by means of the cryoprotection accomplished by a pre-freezing osmotic dehydration step. Osmotic dehydration at mild temperatures can preserve product texture, and enhance flavour and other sensory properties (colour). In this paper, the effect of osmotic pre-treatment on the quality and shelf life of frozen cucumber tissue was investigated. The aim was to develop a method of processing cucumber that maintains palatable product while optimizing quality at the beginning and during frozen storage.  相似文献

This paper develops a multiscale connectivity theory for shapes based on the axiomatic definition of new generalized connectivity measures, which are obtained using morphology-based nonlinear scale-space operators. The concept of connectivity-tree for hierarchical image representation is introduced and used to define generalized connected morphological operators. This theoretical framework is then applied to establish a class of generalized granulometries, implemented at a particular problem concerning soilsection image analysis and evaluation of morphological properties such as size distributions. Comparative results demonstrate the power and versatility of the proposed methodology with respect to the application of typical connected operators (such as reconstruction openings). This multiscale connectivity analysis framework aims at a more reliable evaluation of shape/size information within complex images, with particular applications to generalized granulometries, connected operators, and segmentation.  相似文献
