Experimental studies of plasma membrane permeabilization, caused by single, intense, submicrosecond square wave pulses, indicate that the product of electric field amplitude and pulse duration (the electrical impulse) can be considered a similarity or scaling factor. A model based on the hypothesis that the intensity of membrane permeabilization effects is linearly dependent on the electric charge transferred through the permeabilized membrane, provides results, which are consistent with the empirical observations. For multiple pulses, bioelectric effects caused by ultrashort pulses were found to scale with the square root of the pulse number. This square root dependence on the pulse number points to a statistical motion of cells between pulses with respect to the applied electric field, and can be explained using an extension of the random walk statistical results to random rotations. Besides membrane permeabilization, the scaling law has also been shown to hold for secondary bioelectric effects, which are caused by permeability changes in the plasma membrane or subcellular membranes. 相似文献

We report investigations on polycrystalline LaCo1?x Al x O3 (x = 0–0.9) bulk samples. The solid state synthesized samples showed a coexistence of rhombohedral and monoclinic phases in the intermediate concentrations (0.2 ≤ x ≤ 0.5) and pure rhombohedral phase otherwise. The observed effect of Al doping on dc transport has been analysed on the basis of small polaron hopping mechanism. The magnetisation results presented give evidence of weak ferromagnetism and anomalous temperature dependence of coercivity which we associate to the canting of the localised high-spin Co(III) and anti-symmetric exchange interactions at low temperatures. 相似文献

Gravure is a high throughput printing process, normally associated with speed, quality, and long print runs. It is widely used for printing on shrink films and other substrates. The shrink films, in particular, polyvinyl chloride (PVC) and glycol-modified polyethylene terephthalate (PET-G), are two dominant substrates widely consumed and printed by gravure process. The PVC and PET-G offer different properties which greatly influence the printability. The surface energy of the substrate determines the adhesion and wettability of ink, while electrical properties such as surface and volume resistivity impact electrostatic assist (ESA) performance. The introduction of ESA in gravure further improved the print quality by eliminating dot skips with reduced impression pressure. However, print defects such as print mottle is inevitable. Print mottle occurs due to a discrepancy between substrate, ink, and process parameters which degrade the print quality. These complexities need to be addressed to deliver higher productivity with less print waste. Therefore, the study investigates the effect of process parameters, i.e., substrate type, line screen, air gap (distance between charge bar and impression roller), viscosity, voltage, and speed, and aims to quantify their effect numerically on defect minimization. The Design of Experiments (DOE) was generated for the above-mentioned parameters and analyzed to extract the best combination of process parameters. The optimized setting showed a reduction in solid mottle by 54% and 57% for PET-G and PVC, respectively. 相似文献

Multiple myeloma (MM) is a hematological disease marked by abnormal growth of B cells in bone marrow. Inherent chromosomal instability and DNA damage are major hallmarks of MM, which implicates an aberrant DNA repair mechanism. Studies have implicated a role for CDK12 in the control of expression of DNA damage response genes. In this study, we examined the effect of a small molecule inhibitor of CDK12–THZ531 on MM cells. Treatment of MM cells with THZ531 led to heightened cell death accompanied by an extensive effect on gene expression changes. In particular, we observed downregulation of genes involved in DNA repair pathways. With this insight, we extended our study to identify synthetic lethal mechanisms that could be exploited for the treatment of MM cells. Combination of THZ531 with either DNA-PK inhibitor (KU-0060648) or PARP inhibitor (Olaparib) led to synergistic cell death. In addition, combination treatment of THZ531 with Olaparib significantly reduced tumor burden in animal models. Our findings suggest that using a CDK12 inhibitor in combination with other DNA repair inhibitors may establish an effective therapeutic regimen to benefit myeloma patients. 相似文献

In vitro probiotic attributes of Pediococcus acidilactici, viz. tolerance to acidity and bile salt, phenol resistance, lactic acid production, antioxidant activity, cell surface hydrophobicity, lysozyme resistance and the presence of β‐galactosidase, proteolytic, lipase and peptidoglycan hydrolase activities, were studied. The strain was found to be rich in different proteolytic activities, β‐galactosidase and antioxidant activities, and produced lactic acid. The studies conducted support that P. acidilactici national collection of dairy cultures (NCDC) 252 is a potential probiotic for humans with all the essential basic probiotic properties. This testing is useful to gain insight into this strain and its mechanism of action. All these attributes add to its therapeutic importance and industrial significance. 相似文献

A rat epididymal protein of 27 kDa was identified using neonatal tolerization. This study reports the production and characterization of a polyclonal antiserum to this protein. ELISA was used to demonstrate that this antiserum reacts strongly with epididymal sperm proteins, but has little or no reactivity with testicular proteins. Western blot analysis revealed that this polyclonal antiserum recognized a 27 kDa protein extracted from the corpus epididymidis as well as from spermatozoa from the corpus and cauda epididymides, and immunostaining revealed the presence of the protein in the corpus to cauda epididymides. Stronger reactivity was observed in the supranuclear region and stereocilla of principal cells of the corpus epididymidis and in the luminal content of the corpus and cauda epididymides. The testicular section showed no reactivity. Treatment with the antiserum resulted in time- and dose-dependent agglutination of rat spermatozoa. By indirect immunofluorescence, the antiserum localized proteins in the mid-piece region of rat spermatozoa. Studies were carried out to determine the age at which the protein first became apparent during postnatal development. The protein was expressed from day 40 onwards, as demonstrated by western blot analysis. The androgen regulation of this protein was ascertained by castration and supplementation studies. Expression of this protein showed a decline starting at day 14 after castration and by day 21 the protein was absent; however, androgen replacement resulted in the reappearance of the protein. The results of these studies indicate that the protein identified is specific to the epididymis, and is regulated by development and androgens. The importance of epididymis-specific proteins that are regulated by androgens in sperm maturation is discussed, and the need to ascertain the sequence of the protein and clone the cognate gene is indicated. 相似文献

Pressure driven techniques (viz. reverse osmosis and nanofiltration) have the potentiality to remove the pesticides from water. The observations revealed that pesticides removal mostly depends upon the molecular weight (size exclusion) and hydrophobicity (log P) of the pesticides. Interfacial polymerization of m‐phenylene diamine (MPD) and trimesoyl chloride (TMC) on the polysulfone membranes impart the salt rejection property in it.
