Enterohemorrhagic E. coil (EHEC) serotype O157:H7 is one of the major pathogens, responsible for the severe disease outbreaks. EHEC causes diseases in humans through production of shiga-like toxin leading to bloody diarrhea. The toxin is encoded by stx2 gene in E. coli. The current methodology for detection of EHEC relies on fluorogenic-substrate based culture media or nucleic acid amplification based Real-Time Polymerase Chain Reaction assays that are either time consuming or need expensive instrumentation. In this study, the optical properties of gold nanoparticles (GNPs) have been exploited for detection of nucleic acid of Escherichia coli O157:H7. The stx2 gene representing EHEC signature has been targeted using the gold nanoparticle probes. Gold nanoparticles (GNPs) of 20 +/- 0.2 nm were synthesised by citrate reduction method and characterised by spectroscopy and transmission electron microscopy. The GNPs were functionalised with 19 and 22 bp of thiolated single stranded DNA complementary to target highly conserved 149 bp region of stx2 gene. Transmission Electron Microscopy revealed the hybridization, aggregation and reduction in the interparticle distances of the GNP probes in the presence of target DNA. The aggregation and the spectral shift in the plasmon band observed with 10(6) copies of target DNA indicates feasibility of a simple and quick colorimetric 'spot and read' test in contrast to amplification based detection methods. 相似文献

An optimized design for a 210 kg alloy, TiMn alloy based hydrogen storage system for stationary application is presented. A majority of the studies on metal hydride hydrogen systems reported in literature are based on system scale less than 10 kg, leaving questions on the design and performance of large-scale systems unanswered. On the basis of sensitivity to various design and operating parameters such as thermal conductivity, porosity, heat transfer coefficient etc., a comprehensive design methodology is suggested. Following a series of performance analyses, a multi-tubular shell and tube type storage system is selected for the present application which completes the absorption process in 900 s and the desorption process in 2000 s at a system gravimetric capacity of 0.7% which is a vast improvement over similar studies. The study also indicates that after fifty percent reaction completion, heat transfer ceases to be the major controlling factor in the reaction. This could help prevent over-designing systems on the basis of heat transfer, and ensure optimum system weight. 相似文献

Various mechanisms exist to prevent a potentially deleterious maternal immune response that results in compromising survival of semiallogeneic fetus. In pregnancy, there is a necessary early preimplantation inflammatory stage followed by a postimplantation anti-inflammatory stage. Thus, there is a biphasic 'immune response' observed during the course of pregnancy. We provide the evidence that capacitation of sperm induced the expression of a2 isoform of V-ATPase (ATP6V0A2 referred to as a2V), leukemia inhibitory factor (Lif), Il1b, and Tnf in the sperm. Capacitated sperm also released cleaved N-terminal domain of a2V-ATPase (a2NTD), which upregulates the gene expression of Lif, Il1b, Tnf, and monocyte chemotactic protein-1 (Ccl2 (Mcp1)) in the uterus. Unfertilized eggs had low a2V expression, but after fertilization, the expression of a2V increased in zygotes. This increased level of a2V expression was maintained in preimplantation embryos. Seminal plasma was necessary for upregulation of a2V expression in preimplantation embryos, as mating with seminal vesicle-deficient males failed to elicit an increase in a2V expression in preimplantation embryos. The infiltration of macrophages into the uterus was significantly increased after insemination of both sperm and seminal plasma during the preimplantation period of pregnancy. This dynamic infiltration into the uterus corresponded with the uterine a2V expression through the induction of Ccl2 expression. Furthermore, the polarization ratio of M1:M2 (pro-inflammatory/anti-inflammatory) macrophages in the uterus fluctuated from a ratio of 1.60 (day 1) to 1.45 (day 4) when female mice were inseminated with both sperm and seminal plasma. These data provide evidence that exposure to semen may initiate an inflammatory milieu by inducing a2V and cytokine/chemokine expression, which triggers the influx of macrophages into the preimplantation uterus during the onset of pregnancy and ultimately leads to successful pregnancy outcome. 相似文献

The electronic properties of graphene are strongly influenced by electrostatic forces arising from long-range charge scatterers and by changes in the local dielectric environment. This makes graphene extremely sensitive to the surface charge density of cells interfacing with it. Here, we developed a graphene transistor array integrated with microfluidic flow cytometry for the "flow-catch-release" sensing of malaria-infected red blood cells at the single-cell level. Malaria-infected red blood cells induce highly sensitive capacitively coupled changes in the conductivity of graphene. We examine a number of distinct situations relating to heterogeneous catalysts where either a single atom (or ion), or a very small cluster of atoms functions as the locus of chemical turnover in various distinct kinds of conversion. There is little doubt that individual ions at certain crystallographic sites in nanoporous solids can indeed act as single-site catalysts. The situation concerning nanoclusters of pure metal (or bimetallic entities) is rather more ambiguous. What was hitherto thought to be an effective catalyst made up of a small cluster of Pt supported on ??-Al2O3 (for hydrogenation) now seems to be a single atom of Pt attached to a 5-coordinated AlIII ion. And in the case of Au or Pt on other supports, there is evidence that a single Pt atom, positively charged, but surrounded by alkali-metal ions, is a powerful catalyst for the water?Cgas shift (CO + H2O ?? by reactive magnetron sputtering. The comparison between SiO2 and SiN

host matrices in the optical properties of the multilayers is detailed. Structural analysis was made on the multilayer structures
using Fourier transform infrared spectroscopy. The effect of specific annealing treatments on the optical properties is studied
and we report a higher visible luminescence with a control over the thermal budget when SiO2 is replaced by the SiN
x

The development of a safe and efficient method for hydrogen storage is essential for the use of hydrogen with fuel cells for vehicular applications. Hollow glass microspheres (HGMs) have characteristics suitable for hydrogen storage and are expected to be a potential hydrogen carrier to be used for energy release applications. The HGMs with 10–100 μm diameters, 100–1000 Å pore width and 3–8 μm wall thicknesses are expected to be useful for hydrogen storage. In our research we have prepared HGMs from amber glass powder of particle size 63–75 μm using flame spheroidisation method. The HGMs samples with magnesium and iron loading were also prepared to improve the heat transfer property and thereby increase the hydrogen storage capacity of the product. The feed glass powder was impregnated with calculated amount of magnesium nitrate hexahydrate salt solution to get 0.2–3.0 wt% Mg loading on HGMs. Required amount of ferrous chloride tetrahydrate solution was mixed thoroughly with the glass feed powder to prepare 0.2–2 wt% Fe loaded HGMs. Characterizations of all the HGMs samples were done using FEG-SEM, ESEM and FTIR techniques. Adsorption of hydrogen on all the Fe and Mg loaded HGMs at 10 bar pressure was conducted at room temperature and at 200 °C, for 5 h. The hydrogen adsorption capacity of Fe loaded sample was about 0.56 and 0.21 weight percent for Fe loading 0.5 and 2.0 weight percentage respectively. The magnesium loaded samples showed an increase of hydrogen adsorption from 1.23 to 2.0 weight percentage when the magnesium loading percentage was increased from 0 to 2.0. When the magnesium loading on HGMs was increased beyond 2%, formation of nano-crystals of MgO and Mg was seen on the HGMs leading to pore closure and thereby reduction in hydrogen storage capacity. 相似文献

The seeds of Zanthoxylum armatum DC, on hydrodistillation, yielded 1.2% of the essential oil. The oleoresins were extracted by a Soxhlet extractor using ethanol, ethyl acetate, and isopropyl alcohol. Gas chromatography-mass spectrometry analysis of the essential oil resulted in the identification of 38 components, of which linalool (62%) and limonene (18.1%) were the major components. The major components of oleoresins were linoleic acid, palmitoleic acid, and linalool. The antioxidant potential of essential oil and oleoresins were evaluated by 2,2′-diphenyl picrylhydrazyl radical scavenging, Fe2+ chelating, ferric thiocyanate method, and various lipid peroxidation assays. The essential oil showed maximum antioxidant potential, whereas oleoresins showed moderate antioxidant activity. 相似文献
