Children residing in a low-endemic region (LER), a high-endemic region (HER), and a leprosy colony contact population (CP) were evaluated for lepromin response as well as reactivity to the Mycobacterium leprae-specific synthetic antigen, ND-BSA. The mean reactivity to ND-BSA in the LER group (OD 0.03 +/- 0.03, N = 71) was significantly lower (p < 0.001) than that in the contact population (OD 0.14 +/- 0.09, N = 140) as well as the population residing in the HER (OD 0.09 +/- 0.08, N = 1340). ELISA-positive results were the highest (21.4%) with the CP group and lowest (0.0%) in the LER group, suggesting that it was a measure of the extent of exposure of M. leprae. In the contact population, females showed a preponderance for ELISA positivity over males (p < 0.005), a finding not observed with the HER population. The Mitsuda responses showed a Gaussian-type distribution in all of the three populations examined with the mean response being highest in the LER (6.0 mm +/- 2.9) and lowest in the HER (4.5 mm +/- 2.0) groups. The percent positivity for the Mitsuda reaction was found to be highest in the LER (93.0%) and lowest in the HER (88.3%) groups. The Mitsuda response thus appears to be independent of M. leprae exposure, and its interpretation in a given population needs consideration of several factors, such as nutritional, environmental, etc.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Gastrointestinal complications in human immunodeficiency virus (HIV) infection are indicative of impaired intestinal mucosal immune system. We used simian immunodeficiency virus (SIV)-infected rhesus macaques as an animal model for HIV to determine pathogenic effects of SIV on intestinal T lymphocytes. Intestinal CD4(+) T-cell depletion and the potential for cytokine responses were examined during SIV infection and compared with results for lymphocytes from lymph nodes and blood. Flow cytometric analysis demonstrated severe depletion of CD4(+)CD8(-) single-positive T cells and CD4(+)CD8(+) double-positive T cells in intestinal lamina propria lymphocytes (LPL) and intraepithelial lymphocytes (IEL) during primary SIV infection which persisted through the entire course of SIV infection. In contrast, CD4(+) T-cell depletion was gradual in peripheral lymph nodes and blood. Flow cytometric analysis of intracellular gamma interferon (IFN-gamma) and interleukin-4 (IL-4) production following short-term mitogenic activation revealed that LPL retained same or higher capacity for IFN-gamma production in all stages of SIV infection compared to uninfected controls, whereas peripheral blood mononuclear cells displayed a gradual decline. The CD8(+) T cells were the major producers of IFN-gamma. There was no detectable change in the frequency of IL-4-producing cells in both LPL and peripheral blood mononuclear cells. Thus, severe depletion of CD4(+) LPL and IEL in primary SIV infection accompanied by altered cytokine responses may reflect altered T-cell homeostasis in intestinal mucosa. This could be a mechanism of SIV-associated enteropathy and viral pathogenesis. Dynamic changes in intestinal T lymphocytes were not adequately represented in peripheral lymph nodes or blood.  相似文献

Highly integrated, flexible, and ultrathin wireless communication components are in significant demand due to the explosive growth of portable and wearable electronic devices in the fifth‐generation (5G) network era, but only conventional metals meet the requirements for emerging radio‐frequency (RF) devices so far. Here, it is reported on Ti3C2Tx MXene microstrip transmission lines with low‐energy attenuation and patch antennas with high‐power radiation at frequencies from 5.6 to 16.4 GHz. The radiation efficiency of a 5.5 µm thick MXene patch antenna manufactured by spray‐coating from aqueous solution reaches 99% at 16.4 GHz, which is about the same as that of a standard 35 µm thick copper patch antenna at about 15% of its thickness and 7% of the copper weight. MXene outperforms all other materials evaluated for patch antennas to date. Moreover, it is demonstrated that an MXene patch antenna array with integrated feeding circuits on a conformal surface has comparable performance with that of a copper antenna array at 28 GHz, which is a target frequency in practical 5G applications. The versatility of MXene antennas in wide frequency ranges coupled with the flexibility, scalability, and ease of solution processing makes MXene promising for integrated RF components in various flexible electronic devices.  相似文献

Walnut blight is a significant above-ground disease of walnuts caused by Xanthomonas arboricola pv. juglandis (Xaj). The secreted form of chorismate mutase (CM), a key enzyme of the shikimate pathway regulating plant immunity, is highly conserved between plant-associated beta and gamma proteobacteria including phytopathogens belonging to the Xanthomonadaceae family. To define its role in walnut blight disease, a dysfunctional mutant of chorismate mutase was created in a copper resistant strain Xaj417 (XajCM). Infections of immature walnut Juglans regia (Jr) fruit with XajCM were hypervirulent compared with infections with the wildtype Xaj417 strain. The in vitro growth rate, size and cellular morphology were similar between the wild-type and XajCM mutant strains, however the quantification of bacterial cells by dPCR within walnut hull tissues showed a 27% increase in XajCM seven days post-infection. To define the mechanism of hypervirulence, proteome analysis was conducted to compare walnut hull tissues inoculated with the wild type to those inoculated with the XajCM mutant strain. Proteome analysis revealed 3296 Jr proteins (five decreased and ten increased with FDR ≤ 0.05) and 676 Xaj417 proteins (235 increased in XajCM with FDR ≤ 0.05). Interestingly, the most abundant protein in Xaj was a polygalacturonase, while in Jr it was a polygalacturonase inhibitor. These results suggest that this secreted chorismate mutase may be an important virulence suppressor gene that regulates Xaj417 virulence response, allowing for improved bacterial survival in the plant tissues.  相似文献

A viral infection involves entry and replication of viral nucleic acid in a host organism, subsequently leading to biochemical and structural alterations in the host cell. In the case of SARS-CoV-2 viral infection, over-activation of the host immune system may lead to lung damage. Albeit the regeneration and fibrotic repair processes being the two protective host responses, prolonged injury may lead to excessive fibrosis, a pathological state that can result in lung collapse. In this review, we discuss regeneration and fibrosis processes in response to SARS-CoV-2 and provide our viewpoint on the triggering of alveolar regeneration in coronavirus disease 2019 (COVID-19) patients.  相似文献

This paper is concerned with the experimental and numerical study of face milling of Ti-6Al-4 V titanium alloy. Machining is carried out by uncoated carbide cutters in the presence of an abundant supply of coolant. Experimental analysis is conducted by focusing on the measurement of specific cutting energy, surface integrity and tool performance. The experimental analysis is supplemented by simulations from a 3D finite element model (FEM) of face milling simulation where needed. A tool wear model parameterized from FEM predictions of the tool-chip interface temperature, contact stress and chip velocity is presented. Tool wear patterns are described in terms of various cutting conditions and the influence of tool wear on surface integrity is investigated. Tool wear predictions based on the 3D FEM simulation show good agreement with experimental tool wear measurements. The highest cutting speed realized for the cutting tool material is 182.9 m/min (600 sfpm). Good surface integrity in terms of favorable residual stress and surface finish is achieved under the machining conditions used with limited tool wear. Residual stresses imparted to the machined surface are shown to be compressive.  相似文献

ABSTRACT

The molar fluid composition of either the reservoir fluid or the well stream is determined by combining the true boiling point (TBP’) distillation data with gas chromatographic (GC) analysis of the light ends. For the purpose of thermodynamic simulation of phase behavior of petroleum reservoir fluids, in addition to the compositional data, physical properties of the pseudo fractions, i.e. density and molecular weight are required. A major drawback of the TBP distillation is the fact that the fractions contain typically 20 -30 % of the material outside the defined boiling range. Another significant issue is the use of generalized density and molecular weight data in the absence of experimentally determined values. This can introduce major inaccuracies in the phase behavior calculations because the generalized value of density and molecular weight significantly differ in each oil based on the paraffin-naphthene-aromatic distribution and its geographic origin.

In this work we have performed the true TBP distillation of 7 stabilized North Sea oil samples. All the oils were distilled from carbon number 6 to 19 and the distillation was terminated at C20+, which was termed as the residue. We have performed analysis of the Cm fraction of each oil by gas chromatography. Subsequently, the specific gravity and molecular weights of the TBP fractions were determined and compared with the generalized values, which indicated major differences. In addition, the superiority of the PVT calculations for a volatile oil and a gas condensate using the experimentally determined specific gravity and molecular weight of the pseudo fractions against the generalized properties is also demonstrated.  相似文献

Viscosity of heavy oils is a key parameter in designing potential enhanced oil recovery methods. The recently proposed semiempirical viscosity model by Lindeloff et al. [Web of Science ®] [Google Scholar] was studied in depth. This model is the simplest yet powerful tool for medium-heavy oil viscosities. Based on the analysis, appropriate adjustments have been suggested, with the major one being the division of the pressure-viscosity profile into three different regions. New modifications have improved the overall fit, including the saturated viscosities at low pressures. The %AAD for the data sets studied was found to be 1.37. However, with the limited geographically diverse data, it is not possible to make a comprehensive predictive model. A few limitations of the new model are also stated.  相似文献

Solvent entropy is a force to consider in protein folding andprotein design but is difficult to model. It is investigatedhere in the context of the hp model: Two types of residues,hydrophobic and hydrophilic, are modeled on a lattice. Ninechains and two- and three-dimensional simulations are compared.We show that considering solvent entropy alone, efficient foldingof lattice chains (identification of the native fold) can beachieved by an entropy-driven simulation on its own. Moreover,in a detailed comparison over a wide range of parameters, entropy-guidedsearching outperforms an energy-driven search in the model.The combination of energy- and entropy-driven search yieldsthe most efficient searching. It is compared in detail withthe above results, indicating also how this solvent shell modelmay advantageously be implemented in more complex protein modelingsimulations.  相似文献
