Glycosyltransferases in the Golgi apparatus synthesize cell wall polysaccharides and elaborate the complex glycans of glycoproteins. To investigate the targeting of this type of enzyme to plant Golgi compartments, we generated transgenic Arabidopsis plants expressing alpha-2,6-sialyltransferase, a glycosyltransferase of the mammalian trans-Golgi cisternae and the trans-Golgi network. Biochemical analysis as well as immunolight and immunoelectron microscopy of these plants indicate that the protein is targeted specifically to the Golgi apparatus. Moreover, the protein is predominantly localized to the cisternae and membranes of the trans side of the organelle. When supplied with the appropriate substrates, the enzyme has significant alpha-2,6-sialyltransferase activity. These results indicate a conservation of glycosyltransferase targeting mechanisms between plant and mammalian cells and also demonstrate that glycosyltransferases can be subcompartmentalized to specific cisternae of the plant Golgi apparatus.  相似文献

The thermal stability of two-phase ( + ) lamellar microstructure in Ti-Al-Mo PST (polysynthetically twinned that has single colony) crystals, containing C or Si, was investigated. In addition, the variation of -phase volume fraction in Ti-Al-Mo-(C,Si) systems was investigated at several temperatures. Ti-46Al-1.5Mo-0.2C and Ti-46Al-1.5Mo-1.0Si alloys did not recrystallized (stable in this paper) during heat treatments at various heating rates and temperatures. Moreover, the -phase volume fractions of Ti-46Al-1.5Mo-0.2C and Ti-46Al-1.5Mo-1.0Si alloys which were stable compositions, changed less than those of Ti-47Al and Ti-46Al-1.5Mo alloys which were unstable compositions. The instability of the latter alloys was caused by their relatively higher variation of -phase volume fraction during heating. Therefore, it is suggested that the variation of -phase volume fraction is an important factor in controlling the thermal stability of lamellar microstructure.  相似文献

We consider the problem of determining which of a set of experts has tastes most similar to a given user by asking the user questions about his likes and dislikes. We describe a simple algorithm for generating queries for a theoretical model of this problem. We show that the algorithm requires at most opt(F)(ln(|F|/opt(F)) + 1) + 1 queries to find the correct expert, where opt(F) is the optimal worst-case bound on the number of queries for learning arbitrary elements of the set of experts F. The algorithm runs in time polynomial in |F| and |X| (where X is the domain) and we prove that no polynomial-time algorithm can have a significantly better bound on the number of queries unless all problems in NP have n O(log log n) time algorithms. We also study a more general case where the user ratings come from a finite set Y and there is an integer-valued loss function on Y that is used to measure the distance between the ratings. Assuming that the loss function is a metric and that there is an expert within a distance from the user, we give a polynomial-time algorithm that is guaranteed to find such an expert after at most 2opt(F, ) ln + 2( + 1)(1 + deg(F, )) queries, where deg(F, ) is the largest number of experts in F that are within a distance 2 of any f F.  相似文献

Many algorithmic efforts have been made to address technical issues in designing a streaming media caching proxy. Typical of those are segment-based caching approaches that efficiently cache large media objects in segments which reduces the startup latency while ensuring continuous streaming. However, few systems have been practically implemented and deployed. The implementation and deployment efforts are hindered by several factors: 1) streaming of media content in complicated data formats is difficult; 2) typical streaming protocols such as RTP often run on UDP; in practice, UDP traffic is likely to be blocked by firewalls at the client side due to security considerations; and 3) coordination between caching discrete object segments and streaming continuous media data is challenging. To address these problems, we have designed and implemented a segment-based streaming media proxy, called SProxy. This proxy system has the following merits. First, SProxy leverages existing Internet infrastructure to address the flash crowd. The content server is now free of the streaming duty while hosting streaming content through a regular Web server. Thus, UDP based streaming traffic from SProxy suffers less dropping and no blocking. Second, SProxy streams and caches media objects in small segments determined by the object popularity, causing very low startup latency, and significantly reducing network traffic. Finally, prefetching techniques are used to pro-actively preload uncached segments that are likely to be used soon, thus providing continuous streaming. SProxy has been extensively tested and we show that it provides high quality streaming delivery in both local area networks and wide area networks (e.g., between Japan and the U.S.).  相似文献

We consider the problem of decentralized binary detection in a sensor network where the sensors have access to side information that affects the statistics of their measurements, or reflects the quality of the available channel to a fusion center. Sensors can decide whether or not to make a measurement and transmit a message to the fusion center ("censoring"), and also have a choice of the mapping from measurements to messages. We consider the case of a large number of sensors, and an asymptotic criterion involving error exponents. We study both a Neyman-Pearson and a , Bayesian formulation, characterize the optimal error exponent, and derive asymptotically optimal strategies for the case where sensor decisions are only allowed to depend on locally available information. Furthermore, we show that for the Neyman-Pearson case, global sharing of side information ("sensor cooperation") does not improve asymptotic performance, when the Type I error is constrained to be small.  相似文献

NdBa2Cu3O7−δ (NdBCO) films were grown on rolling-assisted biaxially textured substrates (RABiTS) via pulsed laser deposition. c-Axis-oriented epitaxial NdBCO films with high performance were obtained under optimal deposition conditions. Transmission electron microscopy analysis shows that the NdBCO film grown on RABiTS has a clear interface with a CeO2 cap layer and a nearly perfect lattice structure. The NdBCO film exhibits higher T c of 93.7 K and better in-field J c in magnetic fields and at all field orientations, compared to pure YBCO films.  相似文献

In this work, an aqueous acidic thin‐layer‐based strategy for fabricating nanostructures on silicon by using atomic force microscopy (AFM) nanolithography is presented. The approach involves the formation of microscale droplets via dilute hydrofluoride (DHF) etching, the conversion of the droplets to acidic thin layers by AFM‐probe scanning, and subsequent lithographic operations using a biased probe in the aqueous layers. By varying the concentration of the acidic DHF layers, the thin layers can facilitate the creation of both positive and negative patterns, such as oxide dots and Si pores, through anodic oxidation and dissolution. In particular, the anodic oxidation in the acidic media is associated with the field‐enhanced nonequilibrium dissociation of the weak electrolyte. The Si pore structure formation is related to the field‐assisted dissolution of anodic oxides and the Si substrate. The acidic‐layer‐based technique allows switching between different lithographic modes by changing the acidity of the DHF layers, and is complementary to bulk solution‐based and local meniscus‐based approaches in AFM nanolithography. In principle, this method can also be extended to other materials that have similar reactions with DHF.  相似文献

There are three main approaches for reconstructing 3D models of buildings. Laser scanning is accurate but expensive and limited by the laser’s range. Structure-from-motion (SfM) and multi-view stereo (MVS) recover 3D point clouds from multiple views of a building. MVS methods, especially patch-based MVS, can achieve higher density than do SfM methods. Sophisticated algorithms need to be applied to the point clouds to construct mesh surfaces. The recovered point clouds can be sparse in areas that lack features for accurate reconstruction, making recovery of complete surfaces difficult. Moreover, segmentation of the building’s surfaces from surrounding surfaces almost always requires some form of manual inputs, diminishing the ease of practical application of automatic 3D reconstruction algorithms. This paper presents an alternative approach for reconstructing textured mesh surfaces from point cloud recovered by patch-based MVS method. To a good first approximation, a building’s surfaces can be modeled by planes or curve surfaces which are fitted to the point cloud. 3D points are resampled on the fitted surfaces in an orderly pattern, whose colors are obtained from the input images. This approach is simple, inexpensive, and effective for reconstructing textured mesh surfaces of large buildings. Test results show that the reconstructed 3D models are sufficiently accurate and realistic for 3D visualization in various applications.  相似文献

In this paper, the problem of global decentralised stabilisation for a class of uncertain large-scale feedforward nonlinear systems is investigated. The system under consideration is allowed to contain unknown non-Lipschitz continuous nonlinear terms. The design of the global decentralised controllers takes a two steps procedure. First of all, based on the adding a power integrator technique and the homogeneous domination approach a local homogeneous decentralised controller is proposed for each subsystem of the large-scale feedforward nonlinear system. Then, we integrate a series of nested saturation functions with the homogeneous decentralised controllers and adjust the saturation levels to ensure globally asymptotic stability of the closed-loop system. Simulation studies are conducted to illustrate the effectiveness of the proposed control method.  相似文献
